A one step reverse transcription PCR (RT-PCR) com- bined nested PCR was set up to increase efficiency in the diagnosis of canine distemper virus (CDV) infection after developement of nested PCR. Two PCR primer sets were designed based on the sequence of nucleocapsid gene of CDV Onderstepoort strain. One-step RT-PCR with the outer primer pair was revealed to detect 10 2 PFU/ml. The sensitivity was... dogs. Of the 51 blood samples from dogs clinically suspected of CD, 45 samples were revealed as positive by one-step RT-PCR combined with nested PCR. However, only 15 samples were identified as positive with a single one step RT-PCR. Therefore approximately 60% increase in the efficiency of the diagnosis was observed by the combined method. These results suggested that one step RT-PCR combined with nested PCR could be a sensitive, specific, and practical method for diagnosis of CDV infection. -

There are various perspectives on the roles and functions of museums. In modern times, socio-educational functions of museums are the most emphasized. Most museums offer programs for schools, but teachers still prefer not to visit them. The reason why teachers feel pressured to visit a museum is that they lack knowledge about museum exhibits. Also, it takes too much time and energy for individual teachers to reconstruct the curriculum in conjunction with museum exhibits. The link with the school... -

A One Step PrimeScript RT-PCR Kit (Perfect Real Time; TaKaRa Bio, Japan) was used to detect viral RNA, and Premix Ex Taq (Perfect Real Time) was used to detect the viral, protozoal, fungal, and bacterial DNAs. The real-time PCR assay was performed with the Applied Biosystems 7300 Real-Time PCR System (ABI 7300; Applied Biosystems, USA) for screening and with the LightCycler Nano (Roche... Bovine abortion, diarrhea, and respiratory disease complexes, caused by infectious agents, result in high and significant economic losses for the cattle industry. These pathogens are likely transmitted by various vectors and reservoirs including insects, birds, and rodents. However, experimental data supporting this possibility are scarce. We collected 117 samples and screened them for 44 bovine abortive, diarrheal, and respiratory disease complex pathogens by using Dembo polymerase chain... -

a first step, each cow underwent ultrasonographic examination of the ovaries to establish the presence of a corpus luteum (CL) with a mean diameter of at least 20 mm (Table 1). If a functional CL was present, the experiment started. To induce luteolysis, a PGF2 analogue (Cloprostenol 500 g/animal intramuscularly [i.m.], Estrumate; MSD Animal Health, Germany) was administered (day 0). A GnRH... This study was conducted to evaluate the effects of induced endometritis on uterine blood flow in cows. Transrectal Doppler sonography was performed on uterine arteries of six cyclic cows before and for 4 days after inducing acute endometritis by intrauterine infusion of 720 mg of policresulen, and for 4 days of the following estrous cycle. Time-averaged maximum velocity (TAMV) increased (p < 0.001) and pulsatility index (PI) decreased (p < 0.0001) within 1 h of policresulen... -
Quantitative real-time polymerase chain reaction for detecting Mycoplasma hyosynoviae and Mycoplasma hyorhinis in pen-based oral, tonsillar, and nasal fluids

As recently suggested [3], this step can affect detection of these mycoplasmas in pen-based oral fluids. As demonstrated here, this may be true for other specimens as well. Results of the present investigation suggest that the number of false negatives can be affected by the genomic DNA extraction protocol used depending on the study design and nature of sampling. Importantly, these findings... Mycoplasma (M.) hyorhinis and M. hyosynoviae are pathogens known to cause disease in pigs post-weaning. Due to their fastidious nature, there is increased need for culture-independent diagnostic platforms to detect these microorganisms. Therefore, this study was performed to develop and optimize quantitative real-time PCR (qPCR) assays to rapidly detect M. hyorhinis and M. hyosynoviae in pen-based oral fluids as well as nasal and tonsillar fluids as proxies for samples used in swine herd... -

one uterine horn were immersion fixed in neutral 4% formaldehyde for immunohistochemical analysis or snap frozen in liquid nitrogen and stored at 80 o C until real-time quantitative reverse transcription PCR (qRT-PCR) examination of gene expression and Western blot analysis were conducted. mRNA levels were measured with an RT-PCR assay. Venous blood samples (1.0 mL) were obtained from the... In this investigation, we studied the expression and localization of rat prostaglandin F (FP) receptor in uterine tissues of rats on gestational Days 10, 15, 18, 20, 21, 21.5 and postpartal Days 1 and 3 using Western blotting analysis, real-time PCR, and immunohistochemistry. A high level of immunoreactivity was observed on gestational Days 20, 21, and 21.5 with the most significant signals found on Day 20. FP receptor protein was expressed starting on gestational Day 15, and a fluctuating... -

Verocytotoxic Escherichia (E.) coli strains are responsible for swine oedema disease, which is an enterotoxaemia that causes economic losses in the pig industry. The production of a vaccine for oral administration in transgenic seeds could be an efficient system to stimulate local immunity. This study was conducted to transform tobacco plants for the seed-specific expression of antigenic proteins from a porcine verocytotoxic E. coli strain. Parameters related to an immunological response and... -

a denaturation step at 94 o C for 2 min, followed by 30 cycles of 94 o C for 30 sec, 58 o C (KA1) or 62 o C (GAPDH) for 30 sec, and 72 o C for 45 sec, with a final extension step at 72 o C for 10 min. Semi-quantitative RT- PCR analysis of the KA1 subunit expression was estimated after electrophoresis on 2% agarose gels (USB, USA) using ethidium bromide dye. To quantify the expression of the... The KA1 kainate receptor (KAR) subunit in the substantia gelatinosa (SG) of the trigeminal subnucleus caudalis (Vc) has been implicated in the processing of nociceptive information from the orofacial region. This study compared the expression of the KA1 KAR subunit in the SG of the Vc in juvenile, prepubescent and adult mice. RT-PCR, Western blot and immunohistochemistry analyses were used to examine the expression level in SG area. The expression levels of the KA1 KAR subunit mRNA and protein... -

of coat colour, horn types, limited polymorphic nature of the serum proteins or biochemical variants, etc. Molecular markers are more accurate and reliable than all other markers [9] because of their dense distribution in the genome, great variation, co- dominant inheritance and easy genotyping at DNA level. Among the various molecular genetic markers such as Restriction Fragment Length... Genetic variation in Barbari goats, a highly prolific breed distributed widely in the northern part of India, known for better milk and meat quality, was studied as a part of genetic characterization and conservation. The genomic DNA from 50 unrelated Barbari goats were amplified via PCR with a panel of 21 microsatellite markers, and resolved through 6 per cent denaturing polyacrylamide gel electrophoresis followed by silver staining. The number of alleles ranged from 4 to 11, with allele sizes...
