in GenBank (National Center for Biotechnology Information, USA) was performed to compare the obtained nucleotide sequences with other sequences in the GenBank. To sequence the two APPV strains, overlap RT-PCR was conducted via one-step RT-PCR with the PrimeScript One-Step RT-PCR Kit and primers designed at the conserved regions according to the APPV sequences in GenBank. However, 5´ rapid... (GD-SD), were identified in two newborn piglet herds with congenital tremor from China. The open reading frame of the two strains shared an 83.5% nucleotide identity. Phylogenetically, the APPV strains were placed into two groups: GZ01 belonged to group I and GD-SD belonged to group II. A high viral load was detected in the cerebellum (quantification cycles < 26). Further studies should be carried out to thoroughly elucidate the development of congenital tremors caused by APPV. -

the cytoplasm. Overlap of the two different fluorescence signals appeared 12 h after transfection and continued to persist until the end of the experiment. These data indicate a possible interaction between DEV pUL49.5 and gM. Keywords: colocalization, duck enteritis virus, intracellular localization, molecular characterization, UL49.5 protein Introduction Duck enteritis virus (DEV) is an... domains. This protein was also predicted to localize in the cytoplasm and endoplasmic reticulum with a probability of 66.7%. Images taken by a fluorescence microscope at different time points revealed that the DEV pUL49.5 and gM proteins were both expressed in the cytoplasm. Overlap of the two different fluorescence signals appeared 12 h after transfection and continued to persist until the end of the experiment. These data indicate a possible interaction between DEV pUL49.5 and gM. -

of GFP and dsRed signal showed that these two signals overlap (Fig. 4A, B, and C). We also could not observe the ..PAGE:5 ER retention signal of GP3 in type II PRRSV 311 Fig. 4. Signal sequence of GP3 was fused with GFP to confirm its effect on localization. (A) GFP signal (green) was observed in the cytoplasm excluding the nucleus. No signal was observed in the plasma membrane and nuclear... The glycoprotein 3 (GP3) of type II porcine reproductive and respiratory syndrome virus has the characteristic domains of a membrane protein. However, this protein has been reported to be retained in the endoplasmic reticulum (ER) rather than transported to the plasma membrane of the cell. In this study, we performed confocal laser scanning microscopy analysis of variants of GP3 and foundthat the signal sequence of the GP3 led to confinement of GP3 in the ER, while the functional ortransmembrane... -

the primers NGF-1F and NGF-1R and 400 bp with the primers NGF-2F and primer NGF-2R. Overlapping PCR was performed to combine the PCR products of the partial dβ-NGF using NGF-1F and NGF-2R. This overlap PCR was performed for 35 cycles at 94 o C for 1 min, 58 o C for 1 min and 72 o C for 1 min. The size of this PCR product was confirmed to be approximately 660 bp (pcDNA1) using 1.5% agarose... Due to the therapeutic potential of gene therapy for neuronal injury, many studies of neurotrophic factors, vectors, and animal models have been performed. The presumed dog β-nerve growth factor (pdβ-NGF) was generated and cloned and its expression was confirmed in CHO cells. The recombinant pdβ-NGF protein reacted with a human β-NGF antibody and showed bioactivity in PC12 cells. The pdβb-NGF was shown to have similar bioactivity to the dog β-NGF. The recombinant pdβ-NGF plasmid was... -

TX, USA). In most cases, each electrode contained activities from only one RGC, although spike sorting was also performed. When it was difficult to sort spikes from a single-unit RGC due to spike overlap problem, we did not include the data from that RGC in data pool for further analysis. Continuously-modulated li ght was generated by a PC monitor, and then uniformly presented onto the MEA b... For successful restoration of visual function by a visual neural prosthesis such as retinal implant, electrical stimulation should evoke neural responses so that the information on visual input is properly represented. A stimulation strategy, which means a method for generating stimulation waveforms based on visual input, should be developed for this purpose. We proposed to use the decoding of visual input from retinal ganglion cell (RGC) responses for the evaluation of stimulus encoding... -

and size of expansions has been found (Day JW et al, 2003; Finsterer J, 2002; Ranum LP and Day JW, 2004). Similarities and features of DM1 and DM2 suggest overlap of mo lecular mechanisms in both diseases as well as some differences in the mechanisms of expansion formation and inheri tance and in the molecular expression of mutations. RNA models for molecular mechanisms of DM1 and DM2 A... Myotonic Dystrophies type 1 and 2 (DM1/2) are neuromuscular disorders which belong to a group of genetic diseases caused by unstable CTG triplet repeat (DM1) and CCTG tetranucleotide repeat (DM2) expansions. In DM1, CTG repeats are located within the 3' untranslated region of myotonin protein kinase (DMPK) gene on chromosome 19q. DM2 is caused by expansion of CCTG repeats located in the first intron of a gene coding for zinc finger factor 9 on chromosome 3q. The CTG and CCTG expansions are... -

25-base-pair overlap (Table 1). A 35 S promoter and a CaMV polyA terminator were inserted into the multiple cloning sites of the binary pGreenII 0000 expression vector to generate pGreen-35S (Hellens et al., 2000). The pGreen-35S was amplified by primers with 25-33 bases that were homologous to the viral genome fragments. The PCR amplification reactions were performed with PrimeSTAR GXL DNA... Soybean mosaic virus (SMV) is the predominant viral pathogen that affects the yield and quality of soy- bean. The natural host range for SMV is very narrow, and generally limited to Leguminosae. However, we found that SMV can naturally infect Pinellia ternata and Atractylodes macrocephala. In order to clarify the molecular mechanisms underlying the cross- family infection of SMV, we used double-stranded RNA extraction, rapid amplification of cDNA ends polymerase chain reaction and Gibson... -

of HLFPV genome, 11 primers pairs (available upon requested) for amplifying 11 sequence segments with overlap regions were designed and synthesized in SAN- GON BIOTECH Co., Ltd. basing on the HLFPV Japanese isolate (Yoshida et al., 2014). Then, 11 products of RT- PCR were cloned into pMD18-T vector. The obtained recombined vectors were sequenced and the obtained se- quences were combined into... Although limited progress have been made about pathogen system of Hibiscus rosa-sinensis and Hibiscus latent Fort Pierce virus (HLFPV), interaction between plant host and pathogen remain largely unknown, which led to deficiency of effective measures to control disease of hibiscus plants caused by HLFPV. In this study, infection of HLFPV in Hibiscus rosa-sinensis was firstly confirmed for the first time by traditional electron microscopy, modern reverse transcription polymerase chain reaction and...
