연구의 목적은 갑상선세포(FRTL-5)에서 thyroglobulin (Tg)의 분비에 sericin이 영향을 주는지를 알려고 한다. Sericin에 의해서 Tg의 분비가 촉진되었지만 Tg-mRNA의 발현은 촉진되지 않았다. 이런 상태에서 소포체 샤페론(Bip & calreticulin)과 소포체 막 단백질(IRE1, PERK & ATF6)의 발현이 증가한 것이 확인되었다. 한편 IRE1의 하부 신호전달자인 XBP1의 mRNA splicing 이 약하게 확인되었지만 PERK의 하부 신호전달자인 $eIF2{alpha}$의 인산화는 일어나지 않았다. 그리고 sericin은 MTT assay 결과 cell viability을 촉진시키는... -

갑상선호르몬의 전구체인 thyroglobulin (Tg)이 어떻게 dimerization이 일어나는지를 조사하였다, 변성 혹은 비변성전기영동법으로 관찰한 결과 새롭게 만들어지기 시작한 Tg는 15분에 처음으로 구조적으로 불안정하지만 folded monomer가 관찰되었다. 그 이후에 monomer가 dimer로 바뀌어 결국 30분에 dimer Tg가 만들어지기 시작하여 40분에는 dimer Tg가 관찰되었다. 최종적으로 60분에 완전하게 dimerization된 Tg가 만들어졌다. -

Thyroglobulin과 반응하는 monoclonal antibody를 3클론(TN-1, TN-2 및 TN-3)을 확립하였다. 이들 중 1클론 (TN-2)은 thyroglobulin의 3차원적인 구조를 인식하면서 음전하를 띠는 자가항원의 일종인 phosphatidylserine과도 교차반응성을 나타내었다. 따라서 TN-2를 이용한 자가면역성 갑상선염의 진단 및 치료에의 적용효과가 기대된다. -

한다. GRP94 역시 ER내에 존재하는 molecular chaperone으로 알려지고 있지만 갑상선세포에서 단백질의 folding에 관여한다는 증거는 아직 불충분하다. 본 설험은 molecular chaperone을 세포내에서 overexpression시킬 수 있는 system을 확립하였다. 그 중에서 GRP94가 단백질의 folding에 직접적으로 관여한다는 증거를 얻기 위하여, endogenous GRP94를 code한 cDNA를 overexpression vector에 의해서 forced expression시킴으로 신생thyroglobulin의 folding에 직접적으로 관여하는 증거를 immun-oprecipitation으로 증명하였다. -
