value (4.4⁑0.8 Hz, p<0.001). This effect was blocked by yohimbine (2∼20μM), an α2-adrenoceptor antagonist and mimicked by clonidine (50μM), an α2-adrenoceptor agonist. In contrast, NE increased sIPSC frequency to 248⁑32% of the control (3.06⁑0.37 Hz, p<0.001) in 31 of 54 type II neurons, but decreased the frequency to 41⁑7% of the control (5.5⁑1.3 Hz) in the rest of type II neurons (p<0.001). In both types of PVN neurons, NE did not affect the mean amplitude... -
Nimodipine as a Potential Pharmacological Tool for Characterizing R-Type Calcium Currents
SeogBaeOh 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 9 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.6 8 511-519 (9 pages)
Nimopidine, one of dihydropyridine derivatives, has been widely used to pharmacologically identify L-type Ca currents. In this study, it was tested if nimodipine is a selective blocker for L-type Ca currents in sensory neurons and heterologous system. In mouse dorsal root ganglion neurons (DRG), low concentrations of nimodipine (<10μM), mainly targeting L-type Ca currents, blocked high-voltage-activated calcium channel currents by ∼38%. Interestingly, high concentrations of nimodipine... -
Forward-Mode Na+-Ca2+ Exchange during Depolarization in the Rat Ventricular Myocytes with High EGTA
Eun-GiKim, ChangMannKo 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.6 5 487-494 (8 pages)
During depolarization, extrusion of Ca2 from sarcoplasmic reticulum through forward-mode Na-Ca2 exchange was studied in the rat ventricular myocytes patch-clamped in whole-cell configuration. In order to confine the Ca2 responses in a micro-domain by limiting the Ca2 diffusion time, rat ventricular myocytes were dialyzed with high (14 mM) EGTA. K current was suppressed by substituting KCl with 105 mM CsCl and 20 mM TEA in the pipette filling solution and... -
The Inhibitory Effect of Opioid on the Hyperpolarization-Activated Cation Currents in Rat Substantia Gelatinosa Neurons
GeunHeeSeol, JunKim, SunHeeCho, WonKiKim, JongWhanKim, SangJeongKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.5 2 373-380 (8 pages)
The action of opioid on the hyperpolarization-activated cation current (Ih) in substantia gelatinosa neurons were investigated by using whole-cell voltage-clamp recording in rat spinal brain slices. Hyperpolarizing voltage steps revealed slowly activating currents in a subgroup of neurons. The half-maximal activation and the reversal potential of the current were compatible to neuronal Ih. DAMGO (1μM), a selective- opioid agonist, reduced the amplitude of Ih reversibly. This reduction was... -
Activation of ATP-sensitive Potassium Channels by the Predominant Metabolite of Isoflurane in Rabbit Ventricular Myocytes
JinHan, NariKim, EuiyongKim, SungjuKim, KangheeCho 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 11 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.2 7 165-175 (11 pages)
Background: Recent in vivo experimental evidence suggests that isoflurane-induced cardioprotection may involve KATP channel activation. However, it was demonstrated that isoflurane inhibited KATP channel activities in the inside-out patch mode. To explain this discrepancy, the present investigation tested the hypothesis that a metabolite of isoflurane, trifluoroacetic acid (TFA), contributes to isoflurnae-induced cardioprotection via KATP channel activation during myocardial ischemia and... -
The Alteration of Ca2+-activated K+ Channels in Coronary Arterial Smooth Muscle Cells Isolated from Isoproterenol-induced Cardiac Hypertrophy in Rabbit
NariKim, JinHan, EuiyongKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 10 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.2 5 147-156 (10 pages)
It has been proposed that Ca2-activated K (KCa) channels play an essential role in vascular tone. The alterations of the properties of coronary KCa channels have not been studied as a possible mechanism for impaired coronary reserve in cardiac hypertrophy. The present studies were carried out to determine the properties of coronary KCa channels in normal and hypertrophied hearts. These channels were measured from rabbit coronary smooth muscle cells using a patch clamp technique.... -

3-morpholinosydnonimine-N-ethylcarbamide (SIN-1, 50μM) and 8-(4-Chlorophenylthio)-guanosine-3',5'-cyclic monophosphate (8-pCPT-cGMP, 100μM), a membrane-permeable analogue of cGMP, increased the KCa channel activity in the cell-attached patch configuration, and the effect was removed upon washout of the drugs. In inside-out patches, single-channel current amplitude was not changed by SIN-1 and 8-pCPT-cGMP. Application of ATP (100μM), cGMP (100μM), ATPcGMP (100μM each), PKG (5 U/μl),... -

from pancreatic beta cells. We have investigated the effect of propofol on KATP channels in cultured single pancreatic beta cells of rats. Channel activity was recorded from membrane patches using the patch-clamp technique. In the inside-out configuration bath-applied propofol inhibited the KATP channel activities in a dose-dependent manner. The half-maximal inhibition dose (ED50) was 48.6⁑8.4μM and the Hill coefficient was 0.73⁑0.11. Single channel conductance calculated from the... -
Effect of Capsaicin on Delayed Rectifier K+ Current in Adult Rat Dorsal Root Ganglion Neurons

currents play multiple roles in the excitability of dorsal root ganglion (DRG) neurons. Influences on these currents change the shape of the action potential, its firing threshold and the resting membrane potential. In this study, whole cell configuration of patch clamp technique had been applied to record the blocking effect of capsaicin, a lipophilic alkaloid, on the delayed rectifier K current in cultured small diameter DRG neurons of adult rat. Capsaicin reduced the amplitude of... -
Roles of Ca2+-Activated K+ Conductances on Spontaneous Firing Patterns of Isolated Rat Medial Vestibular Nucleus Neurons

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.1 1 1-8 (8 pages)
To investigate the contributions of intrinsic membrane properties to the spontaneous activity of medial vestibular nucleus (MVN) neurons, we assessed the effects of blocking large and small calcium-activated potassium channels by means of patch clamp recordings. Almost all the MVN neurons recorded in neonatal (P13∼P17) rat were shown to have either a single deep after-hyperpolarization (AHP; type A cells), or an early fast and a delayed slow AHP (type B cells). Among the recorded MVN cells,...
