Some species of the Trichoderma genus are reported as the major problem in oak wood mushroom production in Korea. In spite of economic loss by the fungi, scientific information on airborne Trichoderma species is not much available. To generate information for disease management development we analyzed airborne Trichoderma. A total of 1,063 fungal isolates were purely obtained from indoor air sampling of cultivation houses used for oak wood mushroom using sawdust media. Among the obtained... -
DevelDevelopment of a Quantitative Real-time Nucleic Acid Sequence based Amplification (NASBA) Assay for Early Detection of Apple scar skin viroid-time Nucleic Acid Sequence based Amplification (NASBA) Assay for Early Detection of Apple scar skin viroid

of the starting ASSVd RNA copy number of 10-fold dilutions each. Compared to the established RT-PCR methods, our method was more sensitive for detecting ASSVd. The real-time quantitative NASBA method will be fast, sensitive, and reliable for routine diagnosis and selection of viroid-free stock materials. Furthermore, real-time quantitative NASBA may be especially useful for detecting low levels in apple trees with early viroid-infection stage and for monitoring the influence on tree growth. -
Selection of Small Synthetic Antimicrobial Peptides Inhibiting Xanthomonas citri subsp. citri Causing Citrus Canker
한국식물병리학회 The Plant Pathology Journal 8 Pages
한국식물병리학회 The Plant Pathology Journal 2017, 33권 1호 10 87-94 (8 pages)
Citrus canker disease decreases the fruit quality and yield significantly, furthermore, emerging of streptomycin-resistant pathogens threatens the citrus industry seriously because of a lack of proper control agents. Small synthetic antimicrobial peptides (AMPs) could be a promising alternative. Fourteen hexapeptides were selected by using positional scanning of synthetic peptide combinatorial libraries. Each hexapeptide showed different antimicrobial spectrum against Bacillus, Pseudomonas,... -
Localization of Barley yellow dwarf virus Movement Protein Modulating Programmed Cell Death in Nicotiana benthamiana
한국식물병리학회 The Plant Pathology Journal 13 Pages
한국식물병리학회 The Plant Pathology Journal 2017, 33권 1호 6 53-65 (13 pages)
Barley yellow dwarf virus (BYDV) belongs to Luteovirus and is limited only at phloem related tissues. An open reading frame (ORF) 4 of BYDV codes for the movement protein (MP) of BYDV gating plasmodesmata (PD) to facilitate virus movement. Like other Luteoviruses, ORF 4 of BYDV is embedded in the ORF3 but expressed from the different reading frame in leaky scanning manner. Although MP is a very important protein for systemic infection of BYDV, there was a little information. In this study, MP... -

images detected changes before symptom appearance at low-density inoculation. The results of this study indicate that the P. cichorii infection severity can be detected by chlorophyll fluorescence assay and hyperspectral images prior to the onset of visible symptoms, indicating the feasibility of early detection of diseases. However, to detect disease development by hyperspectral imaging, more detailed protocols and analyses are necessary. Taken together, change in chlorophyll fluorescence is a... -
Development of a Rapid Detection Method for Potato virus X by Reverse Transcription Loop-Mediated Isothermal Amplification

a set of loop primers since one out of six primers showed PVX specificity. Based on real-time monitoring, RT-LAMP detected PVX around 30 min, compared to 120 min for RT-PCR. By adding a fluorescent reagent during the reaction, the extra step of visualization by gel electrophoresis was not necessary. RT-LAMP was conducted using simple inexpensive instruments and a regular incubator to evaluate whether RNA could be amplified at a constant temperature instead of using an expensive thermal cycler.... -
Development of a Model to Predict the Primary Infection Date of Bacterial Spot (Xanthomonas campestris pv. vesicatoria) on Hot Pepper
한국식물병리학회 The Plant Pathology Journal 11 Pages
한국식물병리학회 The Plant Pathology Journal 2014, 30권 2호 3 125-135 (11 pages)
A population model of bacterial spot caused by Xanthomonas campestris pv. vesicatoria on hot pepper was developed to predict the primary disease infection date. The model estimated the pathogen population on the surface and within the leaf of the host based on the wetness period and temperature. For successful infection, at least 5,000 cells/ml of the bacterial population were required. Also, wind and rain were necessary according to regression analyses of the monitored data. Bacterial spot on... -

to the rpf system. Through experiments using the rpfE-deficient mutant strain, we found that mutation in rpfE gene in Xoo reduced virulence, swarm motility, and production of virulence factors such as cellulase and extracellular polysaccharide. Disease progress by the rpfE-deficient mutant strain was significantly slowed compared to disease progress by the wild type and the number of the rpfE-deficient mutant strain was lower than that of the wild type in the early phase of infection in the... -
Research Articles : A Three-Year Field Validation Study to Improve the Integrated Pest Management of Hot Pepper
한국식물병리학회 The Plant Pathology Journal 11 Pages
한국식물병리학회 The Plant Pathology Journal 2013, 29권 3호 8 294-304 (11 pages)
To improve the integrated pest management (IPM) of hot pepper, field study was conducted in Hwasung from 2010 to 2012 and an IPM system was developed to help growers decide when to apply pesticides to control anthracnose, tobacco budworm, Phytophthora blight, bacterial wilt, and bacterial leaf spot. The three field treatments consisted of IPM sprays following the forecast model advisory, a periodic spray at 7-to-10-day intervals, and no spray (control). The number of annual pesticide... -
A Forecast Model for the First Occurrence of Phytophthora Blight on Chili Pepper after Overwintering
한국식물병리학회 The Plant Pathology Journal 13 Pages
한국식물병리학회 The Plant Pathology Journal 2012, 28권 2호 6 172-184 (13 pages)
An infection risk model for Phytophthora blight on chili pepper was developed to estimate the first date of disease occurrence in the field. The model consisted of three parts including estimation of zoosporangium formation, soil water content, and amount of active inoculum in soil. Daily weather data on air temperature, relative humidity and rainfall, and the soil texture data of local areas were used to estimate infection risk level that was quantified as the accumulated amount of active...
