vascular smooth muscle cell (VSMC) proliferation. VSMC was primarily cultured from rat aorta and confirmed by the immunocytochemistry with anti-smooth muscle myosin antibody. The number of viable VSMCs were counted, and lactate dehydrogenase (LDH) activity was measured to assess the degree of cell death. Concentrations of nitrite in the culture medium were measured as an indicator of NO production. LPS was introduced into the medium to induce the inducible nitric oxide synthase (iNOS) in VSMC,... -
Effects of Oxidative Stress on the Expression of Aldose Reductase in Vascular Smooth Muscle Cells

Aminoguanidine, a scanvenger of dicarbonyl, significantly down-regulated the MG-induced AR mRNA. In addition, the inhibition of AR activities with statil, an AR inhibitor, enhanced the cytotoxic effect of MG on SMC under normal glucose, suggesting a protective role of AR against MG-induced cell damages. These results imply that the induction of AR by MG may contribute to an important cellular detoxification of reactive aldehyde compounds generated under oxidative stress in extrahepatic tissues. -
Molecular Aspects of Organic Ion Transporters in the Kidney
SeokHoChaHitoshiEndou 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 16 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.2 1 107-122 (16 pages)
A function of the kidney is elimination of a variety of xenobiotics ingested and wasted endogenous compounds from the body. Organic anion and cation transport systems play important roles to protect the body from harmful substances. The renal proximal tubule is the primary site of carrier-mediated transport from blood into urine. During the last decade, molecular cloning has identified several families of multispecific organic anion and cation transporters, such as organic anion transporter... -
Role of Angiotensin II and Nitric Oxide in the Rat Paraventricular Nucleus

were markedly elevated 6 hrs after unilateral carotid artery ligation. Losartan injection (10μg/0.3μl) into the PVN augmented of the increment of AT1A and AT1B mRNAs It also increased ecNOS gene expression. In addition, AT1B mRNA levels increased after N-nitro-L-arginine methyl ester (L-NAME) injection (50μg/0.3μl) into the PVN. These results suggest that Ang II and NO in the rat PVN may interplay, at least in part, through regulation of gene expression of ecNOS and AT1B, respectively. -
Regulation of Immediate Early Gene Expression by Glutamate Receptor Activation in C6 Rat Glioma Cells
Jin-KooLee, Yung-HiKim, Seong-SooChoi, Hong-WonSuh 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 7 Pages

We have studied the effects of excitatory amino acids on the expression of the c-fos and c-jun mRNA in rat C6 glioma cells. The glutamate, N-methyl-D-aspartate (NMDA), and kainic acid (KA) increased c-fos mRNA level in a concentration-dependent manner. However, they did not affect c-jun mRNA level. In addition, forskolin and phorbol 12-myristate 13-acetate (PMA) increased c-fos mRNA level. Furthermore, PMA increased c-jun mRNA level whereas forskolin downregulated c-jun mRNA level. The... -
GLUT Phosphorylation May be Required to GLUT Translocation Mechanism
Jong-SikHah 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 10 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.6 8 487-496 (10 pages)
In this work, GLUTs phosphorylations by a downstream effector of PI3-kinase, PKC-ζ, were studied, and GLUT4 phosphorylation was compared with GLUT2 phosphorylation in relation to the translocation mechanism. Prior to phosphorylation experiment, PKC-ζ kinase activity was determined as 20.76⁑4.09 pmoles Pi/min/25 ng enzymes. GLUT4 was phosphorylated by PKC-ζ and the phosphorylation was increased on the vesicles immunoadsorpted from LDM and on GLUT4 immunoprecipitated from GLUT4-... -
Different Effects of Dopamine on Differential Rotational Mobility between Inner and Outer Monolayer of Synaptosomal Plasma Membrane Vesicles Isolated from Bovine Brain

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.5 10 409-415 (7 pages)
Fluorescence polarization of 1,6-diphenyl-1,3,5-hexatriene (DPH) was used to evaluate the effects of dopamine·HCl on the range of the rotatioanl mobility of bulk bilayer structure of the synaptosomal plasma membrane vesicles (SPMV) isolated from whole bovine brain. In a dose-dependent manner, dopamine decreased the anisotropy (γ), limiting anisotropy (γ∞) and order parameter (S) of DPH in the membranes. These indicate that dopamine increased the rotational mobility of the probe in the... -
Stationary Outward and Transient Ca2+-Dependent Currents in Hamster Oocytes
Yang-MiKim, Jae-HeeHan, Jong-SuKim, Seong-GeunHong 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.5 9 403-408 (6 pages)
The outward currents elicited in hamster eggs by depolarizing pulses were studied. The currents appeared to comprise at least two components, a transient outward component (Ito) and a steady-state outward component (I∞). Ito was transiently followed by the cessation of inward Ca2 current (ICa), and its current-voltage (I-V) relation was a mirror image of that of ICa. Either blockade of ICa by Co2 or replacement of Ca2 with Sr2 abolished Ito without change in I∞.... -
Changes of the Level of G Protein α-subunit mRNA by Withdrawal from Morphine and Butorphanol
SeikwanOh 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 9 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.4 4 291-299 (9 pages)
Morphine or butorphanol was continuously infused into cerebroventricle (i.c.v.) with the rate of 26 nmol/ μl/h for 3 days, and the withdrawal from opioid was rendered 7 hrs after the stopping of infusion. The expression of physical dependence produced by these opioids was evaluated by measuring the naloxone- precipitated withdrawal signs. The withdrawal signs produced in animals dependent on butorphanol (kappa opioid receptor agonist) were similar to those of morphine (mu opioid receptor... -

central activities related to gastrointestinal, cardiovascular and respiratory functions. In order to study ionic actions of CCK on each type of receptor, we investigated the effects of CCK-8S on neurons located in the NTS of the rat using whole-cell patch-clamp recordings in brainstem slices. Application of CCK-8S, under current clamp, produced a membrane depolarization accompanied by action potential firing. This CCK-evoked excitation was dose-dependent (10 nM∼10μM) and observed in more th...
