The aim of the present study was to investigate the effects of R-()-2,10,11-trihydroxy-N-propylnoraporphine [R-()-TNPA], a selective agonist of dopaminergic D2 receptor and S()-raclopride, a selective antagonist of dopaminergic D2 receptor, on the secretion of catecholamines (CA) evoked by cholinergic stimulation and membrane-depolarization in the isolated perfused model of the rat adrenal gland, and also to establish its mechanism of action. R-()-TNPA (10∼100μM)... -
Calcium Ions are Involved in Modulation of Melittin-induced Nociception in Rat: I. Effect of Voltage-gated Calcium Channel Antagonist

Melittin-induced nociceptive responses are mediated by selective activation of capsaicin-sensitive primary afferent fibers and are modulated by excitatory amino acid receptor, cyclooxygenase, protein kinase C and serotonin receptor. The present study was undertaken to investigate the peripheral and spinal actions of voltage-gated calcium channel antagonists on melittin-induced nociceptive responses. Changes in mechanical threshold and number of flinchings were measured after intraplantar (i.pl.)... -

to a more integrative approach. The neurovascular unit emphasizes that signaling between vascular and neuronal compartments comprise the basis for both function and dysfunction in brain. Hence, brain disorders are not just due to death of neurons, but instead manifested as cell signaling perturbations at the neurovascular interface. In this mini-review, we will examine 3 examples of this hypothesis: neurovascular mechanisms involved in the thrombolytic therapy of stroke, the crosstalk between... -

The functional expression of potassium (K) channels has electrophysiologically been studied in bone cells from several species, however, their identity and regulation of gene expressions in bone cells are not well known. In the present study, to investigate how K channel expressions are regulated by estrogen, we measured changes of transcript levels of various Ca2-activated (KCa) and ATP-sensitive K channels in rat osteoblastic ROS 17/2.8 cells after treatment with... -
Effects of Phenylephrine on the Excitability of Medial Vestibular Nuclear Neurons in Rats

potential of medial vestibular nuclear neurons was decreased by phenylephrine (n=15). Phenylephrine increased the amplitude of afterhyperpolarization without changes in the resting membrane potential and spike width. In voltage-clamp mode, the whole potassium currents of the medial vestibular nuclear neurons were increased by phenylephrine (n=12). These experimental results suggest that α-receptor mediates the inhibitory effects on the neuronal activity of the medial vestibular nuclear neuron. -
Effects of Zinc on Spontaneous Miniature GABA Release in Rat Hippocampal CA3 Pyramidal Neurons

mechanically dissociated rat CA3 pyramidal neurons which retained functional presynaptic nerve terminals. The spontaneous miniature excitatory and inhibitory postsynaptic currents (mEPSCs and mIPSCs, respectively) were pharmacologically isolated and recorded using whole-cell patch clamp technique under voltage-clamp conditions. Zn2 at a lower concentration (30μM) increased GABAergic mIPSC frequency without affecting mIPSC amplitude, but it decreased both mIPSC frequency and amplitude at... -

has direct effects on several receptors. Employing whole-cell patch clamp techniques in rat brain slice, we studied the effects of fluoxetine on corticostriatal synaptic transmission by measuring the change in spontaneous excitatory postsynaptic currents (sEPSC). Acute treatment of rat brain slice with fluoxetine (10μM) significantly decreased the amplitude of sEPSC (84.1⁑3.3%, n=7), but did not alter its frequency (99.1⁑4.7%, n=7). Serotonin (10μM) also significantly decreased the... -
pH-mediated Regulation of Pacemaker Activity in Cultured Interstitial Cells of Cajal

7 has been suggested to be required for intestinal pacemaking activity. TRPM7 produced large outward currents and small inward currents by voltage ramps, ranging from 100 to 100 mV from a holding potential of 60 mV. The inward current of TRPM7 was dramatically increased by a decrease in the extracellular pH. At pH 4.0, the average inward current amplitude measured at 100 mV was increased by about 7 fold, compared with the current amplitude at pH 7.4. Changes in the... -

for association and dissociation of CsA were 7.0μM1s1 and 8.1 s1, respectively. CsA slowed the deactivation time course, resulting in a tail crossover phenomenon. Block of Kv1.5 by CsA was use-dependent. CsA also blocked Kv1.3 currents at 50 mV in a reversible concentration-dependent manner with an apparent IC50 of 1.1μM. The same effects of CsA on Kv1.3 were also observed in excised inside-out patches when applied to the internal surface of the membrane. The presen... -

Striatum has important roles in motor control, habitual learning and memory. It receives glutamatergic inputs from neocortex and thalamus, and dopaminergic inputs from substantia nigra. We examined effects of dopamine (DA) on the corticostriatal synaptic transmission using in vitro extracellular recording technique in rat brain corticostriatal slices. Synaptic responses were elicited by stimulation of cortical glutamatergic inputs on the corpus callosum and recorded in the dorsal striatum....
