Testosterone Relaxes Rabbit Seminal Vesicle by Calcium Channel Inhibition
The Korean Journal of Physiology & Pharmacology 6 Pages
The Korean Journal of Physiology & Pharmacology 2008, Vol.12 No.2 6 73-78 (6 pages)
Recent studies have documented that testosterone relaxes several smooth muscles by modulating K+ channel activities. Smooth muscles of seminal vesicles play a fundamental role in ejaculation, which might involve testosterone. This study was aimed to assess the role of testosterone in seminal vesicular motility by studying its effects on contractile agents and on the ion channels of single vesicular myocytes in a rabbit model. The contractile responses of circular smooth muscle strips of rabbit... -
Relaxant Effect of Spermidine on Acethylcholine and High K+- induced Gastric Contractions of Guinea-Pig
The Korean Journal of Physiology & Pharmacology 6 Pages
The Korean Journal of Physiology & Pharmacology 2008, Vol.12 No.2 4 59-64 (6 pages)
In our previous study, we found that spermine and putrescine inhibited spontaneous and acetylcholine (ACh)-induced contractions of guinea-pig stomach via inhibition of L-type voltage- dependent calcium current (VDCCL). In this study, we also studied the effect of spermidine on mechanical contractions and calcium channel current (IBa), and then compared its effects to those by spermine and putrescine. Spermidine inhibited spontaneous contraction of the gastric smooth muscle in a... -

membrane depolarization in current-clamp mode and increased tonic inward pacemaker currents in voltage-clamp mode. The tonic inward currents by TXA2 were inhibited by intracellular application of GDP-Ղ-S. The pretreatment of ICC with Ca2+ free solution and thapsigargin, a Ca2+-ATPase inhibitor in endoplasmic reticulum, abolished the generation of pacemaker currents and suppressed the TXA2-induced tonic inward currents. However, chelerythrine or calphostin C, protein kinase C inhibitors,... -
PGE2 Regulates Pacemaker Currents through EP2-Receptor in Cultured Interstitial Cells of Cajal from Murine Small Intestine

The interstitial cells of Cajal (ICCs) are the pacemaker cells in gastrointestinal tract and generate electrical rhythmicity in gastrointestinal muscles. Therefore, ICC may be modulated by endogenous agents such as neurotransmitter, hormones, and prostaglandins (PGs). In the present study, we investigated the effects of prostaglandins, especially PGE2, on pacemaker currents in cultured ICCs from murine small intestine by using whole-cell patch clamp techniques. ICCs generated spontaneous slow... -
Cooperation of Gβ and Gαq Protein in Contractile Response of Cat Lower Esophageal Sphincter (LES)
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 2003, Vol.7 No.6 9 349-355 (7 pages)
We previously shown that LES contraction depends on M3 receptors linked to PTX insensitive Gq protein and activation of PLC. This results in production of IP3, which mediates calcium release, and contraction through a CaM dependent pathway. In the esophagus ACh activates M2 receptors linked to PTX sensitive Gi3 protein, resulting in activation of PLD, presumably, production of DAG. We investigated the role of PLC isozymes which can be activated by Gq or Gβ protein on ACh-induced contraction in... -
Dietary Ascorbate Supplementation Reduces Oxidative Tissue Damage and Expression of iNOS in the Kidney of Streptozotocin Induced Diabetic Rats

The Korean Journal of Physiology & Pharmacology 2003, Vol.7 No.1 8 39-45 (7 pages)
Reactive oxygen species (ROS) have been suggested to be contributory factors in complications of diabetes mellitus. In the present study, we investigated the generation of superoxide, the lipid peroxide level measured as thiobarbituric acid reactive substances, the vasorelaxation of isolated thoracic aorta and the iNOS expression in kidney of streptozotocin induced diabetic rats. Sprague Dawley rats were divided into four groups: control, ascorbate (400 mg/kg rat weight daily in drinking water),... -
Neurotensin Enhances Gastric Motility in Antral Circular Muscle Strip of Guinea-pig
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.3 7 227-234 (8 pages)
NT-induced contractile responses were abolished in Ca2-free solution and reduced greatly to near abolition by 10μM of verapamil or 0.2 mM of CdCl2. Verapamil attenuated the effects of NT on frequency and amplitude of the slow waves. Taken together, these results indicate that NT enhances contractility in guinea-pig gastric antral circular muscle and Ca2 influx through the voltage-operated Ca2 channel appears to play an important role in the NT-induced contractile mechanism. -
Decreased Voltage Dependent K+ Currents in Cerebral Arterial Smooth Muscle Cells of One-Kidney, One-Clip Goldblatt Hypertensive Rat
The Korean Journal of Physiology & Pharmacology 9 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.5 3 471-479 (9 pages)
The Kv channel activity in vascular smooth muscle cell plays an important role in the regulation of membrane potential and blood vessel tone. It was postulated that increased blood vessel tone in hypertension was associated with alteration of Kv channel and membrane potential. Therefore, using whole cell mode of patch-clamp technique, the membrane potential and the 4-AP-sensitive Kv current in cerebral arterial smooth muscle cells were compared between normotensive rat and one-kidney, one-clip... -
Effect of Imipramine on Calcium Utilization of Single Cells Isolated from Canine Detrusor
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.4 9 439-445 (7 pages)
ACh-induced contraction was significantly inhibited by imipramine. In Ca2-free PSS, ACh-induced contraction was less than those in normal PSS and it was not affected by the pretreatment with imipramine. Ca2-induced contraction in Ca2-free PSS was supressed by imipramine, but addition of A 23187, a calcium ionophore, overcomed the inhibitory effect of imipramine. High potassium-depolarization (40 mM KCl) evoked cell contraction, which was inhibited by imipramine. Caffeine, a... -

We sought to find out the mechanism of vascular relaxation by extracellular K concentration ([K]o) in the cerebral resistant arteriole from rabbit. Single cells were isolated from the cerebral resistant arteriole, and using voltage-clamp technique barium-sensitive K currents were recorded, and their characteristics were observed. Afterwards, the changes in membrane potential and currents through the membrane caused by the change in [K]o was observed. In the smooth...
