Antidepressant drug paroxetine blocks the open pore of Kv3.1 potassium channel

a calculated KD value of 7.9 µM. The analyses of Kv3.1 tail current indicated that paroxetine did not affect ion selectivity and slowed its deactivation time course, resulting in a tail crossover phenomenon. Paroxetine inhibited Kv3.1 channels in a use-dependent manner. Taken together, these results suggest that paroxetine blocks the open state of Kv3.1 channels. Given the role of Kv3.1 in fast spiking of interneurons, our data imply that the blockade of Kv3.1 by paroxetine might elevate... -
Biphasic augmentation of alpha-adrenergic contraction by plumbagin in rat systemic arteries

Plumbagin, a hydroxy 1,4-naphthoquinone compound from plant metabolites, exhibits anticancer, antibacterial, and antifungal activities via modulating various signaling molecules. However, its effects on vascular functions are rarely studied except in pulmonary and coronary arteries where NADPH oxidase (NOX) inhibition was suggested as a mechanism. Here we investigate the effects of plumbagin on the contractility of skeletal artery (deep femoral artery, DFA), mesenteric artery (MA) and renal... -
Electrophysiological characteristics of R47W and A298T mutations in CLC-1 of myotonia congenita patients and evaluation of clinical features

The Korean Journal of Physiology & Pharmacology 2017, Vol.21 No.4 10 439-447 (9 pages)
Myotonia congenita (MC) is a genetic disease that displays impaired relaxation of skeletal muscle and muscle hypertrophy. This disease is mainly caused by mutations of CLCN1 that encodes human skeletal muscle chloride channel (CLC-1). CLC-1 is a voltage gated chloride channel that activates upon depolarizing potentials and play a major role in stabilization of resting membrane potentials in skeletal muscle. In this study, we report 4 unrelated Korean patients diagnosed with myotonia congenita... -
MDL-12330A potentiates TRAIL-induced apoptosis in gastric cancer cells through CHOP-mediated DR5 upregulation
The Korean Journal of Physiology & Pharmacology 9 Pages
The Korean Journal of Physiology & Pharmacology 2017, Vol.21 No.4 5 397-405 (9 pages)
MDL-12330A is a widely used adenylyl cyclase (AC) inhibitor that blocks AC/cAMP signaling. In this study, we demonstrated a novel antitumor activity of this drug in gastric carcinoma (GC) cell lines. In these GC cells, MDL-12330A reduced cell viability and induced cell death in a concentration-dependent manner. At a moderate concentration (~20 μM), MDL-12330A mainly induced apoptotic death whereas at concentrations greater than 20 μM, it increased non-apoptotic cell death. The induction of... -
Reactive oxygen species increase neuronal excitability via activation of nonspecific cation channel in rat medullary dorsal horn neurons
The Korean Journal of Physiology & Pharmacology 6 Pages
The Korean Journal of Physiology & Pharmacology 2017, Vol.21 No.4 2 371-376 (6 pages)
The caudal subnucleus of the spinal trigeminal nucleus (medullary dorsal horn; MDH) receives direct inputs from small diameter primary afferent fibers that predominantly transmit nociceptive information in the orofacial region. Recent studies indicate that reactive oxygen species (ROS) is involved in persistent pain, primarily through spinal mechanisms. In this study, we aimed to investigate the role of xanthine/xanthine oxidase (X/XO) system, a known generator of superoxide anion (O2·-), on... -
JS-III-49, a hydroquinone derivative, exerts anti-inflammatory activity by targeting Akt and p38
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2017, Vol.21 No.3 9 345-352 (8 pages)
Since previous studies have reported that hydroquinone (HQ) exerted immunosuppressive and anti-inflammatory activity, various HQ derivatives have been synthesized and their biological activities investigated. In this study, we explored the anti-inflammatory activity of JS-III-49, a novel HQ derivative, in macrophagemediated inflammatory responses. JS-III-49 suppressed the production of the inflammatory mediators nitric oxide (NO) and prostaglandin E2 (PGE2) and downregulated the mRNA expression... -
Activation of transient receptor potential vanilloid 3 by the methanolic extract of Schisandra chinensis fruit and its chemical constituent γ-schisandrin
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2017, Vol.21 No.3 5 309-316 (8 pages)
Transient receptor potential vanilloid 3 (TRPV3) is a non-selective cation channel with modest permeability to calcium ions. It is involved in intracellular calcium signaling and is therefore important in processes such as thermal sensation, skin barrier formation, and wound healing. TRPV3 was initially proposed as a warm temperature sensor. It is activated by synthetic small-molecule chemicals and plantderived natural compounds such as camphor and eugenol. Schisandra chinensis (Turcz.) Baill... -
Nortriptyline, a tricyclic antidepressant, inhibits voltage-dependent K+ channels in coronary arterial smooth muscle cells
SungEunShin, HongliangLi, HanSolKim, HyeWonKim, MiSeonSeo, Kwon-SooHa, Eun-TaekHan, Seok-HoHong, AmyL.Firth, Il-WhanChoi6, YoungMinBae, Won 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2017, Vol.21 No.2 10 225-232 (8 pages)
We demonstrated the effect of nortriptyline, a tricyclic antidepressant drug and serotonin reuptake inhibitor, on voltage-dependent K+ (Kv) channels in freshly isolated rabbit coronary arterial smooth muscle cells using a whole-cell patch clamp technique. Nortriptyline inhibited Kv currents in a concentration-dependent manner, with an apparent IC50 value of 2.86±0.52 µM and a Hill coefficient of 0.77±0.1. Although application of nortriptyline did not change the activation curve,...
