Adenosine has been reported to provide cytoprotection in the central nervous systems as well as myocardium by activating cell surface adenosine receptors. However, the exact target and mechanism of its action still remain controversial. The present study was performed to examine whether adenosine has a protective effect against Aβ-induced injury in neuroglial cells. The astrocyte-derived human neuroglioma cell line, A172 cells, and Aβ25∼35 were employed to produce an experimental Aβ-induced... -

ET-1 were increased, while that of angiotensin converting enzyme 2 was not changed. The expressions of CYP11B2 and angiotensin II receptors were not changed. C-type natriuretic peptide (CNP) expression was increased, while its receptors (natriuretic peptide receptor-B) were not changed. We suggest that the upregulation of local RAS and ET play a role in the progressive glomerular injury, and that the enhanced CNP activity also plays a compensatory role in obstructive uropathy in the glomerulus. -
Lysophosphatidylcholine Attenuates Endothelium-dependent Relaxation Responses through Inhibition of ACh-induced Endothelial [Ca2+]i Increase
Seong-ChunKwon, Yong-HoLee, TaicksangNam, Duck-SunAhn 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages

Lysophosphatidylcholine (LPC), which accumulates in atherosclerotic arteries, has been reported to inhibit endothelium-dependent relaxation (EDR) in many different species. However, the underlying mechanism of LPC-induced inhibition of EDR is still uncertain. In the present study, we measured simultaneously both isometric tension and cytosolic free Ca2 ([Ca2]i) in rabbit carotid strips, and examined the effect of LPC on tension and [Ca2]i. In carotid strips with... -
Calcium Ion Dynamics after Dexamethasone Treatment in Organotypic Cultured Hippocampal Slice
Hee-JungChae, Tong-HoKang, Ji-HoPark 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages

It is imperative to analyse brain injuries directly in real time, so as to find effective therapeutic compounds to protect brain injuries by stress. We established a system which could elucidate the real time Ca2 dynamics in an organotypic cultured hippocampal slice by the insults of artificial stress hormone, dexamethasone. The real time Ca2 dynamics could continuously be detected in cornus ammonis 3 (CA3) of the organotypic hippocampus for 8 hours under confocal microscopy. When... -

Laser Scanning Confocal Microscopy (LSCM). Furthermore, the role of sarcolemmal Na/Ca2 exchange (NCX) in cytosolic Ca2 efflux was studied in KB-R7943 and Na-free normal Tyrode's solution (143 mM LiCl ). When DNP was applied to cells loaded with Fluo-4 AM, Fluo-4 AM fluorescence intensity initially increased by 70⁑10% within 70⁑10 s, and later by 400⁑200% at 850⁑46 s. Fluorescence intensity of both Rhod-2 AM and TMRE were initially decreased by... -

a fluorogenic indicator Alamar blue. We found that TBH decreased cell viability in a time- and concentration-dependent manner. YS 49 concentration- and time-dependently increased HO-1 induction on SPAECs. As expected, YS 49 significantly decreased the TBH-induced cellular injury. In the presence of zinc protophorphyrin, HO-1 inhibitor, effect of YS 49 was significantly inhibited, indicating that HO-1 plays a protective role for YS 49. Furthermore, YS 49 showed free radical scavenging activity as... -
TASK-1 Channel Promotes Hydrogen Peroxide Induced Apoptosis

Hydrogen peroxide (H2O2) causes oxidative stress and is considered as an inducer of cell death in various tissues. Two-pore domain K (K2p) channels may mediate K efflux during apoptotic volume decreases (AVD) in zygotes and in mouse embryos. In the present study, we sought to elucidate linkage between K2p channels and cell death by H2O2. Thus K2p channels (TASK-1, TASK-3, TREK-1, TREK-2) were stably transfected in HEK-293 cells, and cytotoxicity assay was preformed using cell... -
Acetylcholine Induces Hyperpolarization Mediated by Activation of K(Ca) Channels in Cultured Chick Myoblasts
DoyunLee, JaeheeHan, Jae-YongPark 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2005, Vol.9 No.1 6 37-44 (8 pages)
Our previous report demonstrated that chick myoblasts are equipped with Ca2-permeable stretch- activated channels and Ca2-activated potassium channels (KCa), and that hyperpolarization-induced by KCa channels provides driving force for Ca2 influx through the stretch-activated channels into the cells. Here, we showed that acetylcholine (ACh) also hyperpolarized the membrane of cultured chick myoblasts, suggesting that nicotinic acetylcholine receptor (nAChR) may be another... -
Buffering Contribution of Mitochondria to the [Ca2+]i Increase by Ca2+ Influx through Background Nonselective Cation Channels in Rabbit Aortic Endothelial Cells

To prove the buffering contribution of mitochondria to the increase of intracellular Ca2+ level ([Ca2+]i) via background nonselective cation channel (background NSCC), we examined whether inhibition of mitochondria by protonophore carbonylcyanide m-chlorophenylhydrazone (CCCP) affects endothelial Ca2+ entry and Ca2+ buffering in freshly isolated rabbit aortic endothelial cells (RAECs). The ratio of fluorescence by fura-2 AM (R340/380) was measured in RAECs. Biological state was checked by... -
Cytotoxicological Effect of Tebufenozide, an Insect Growth Regulator (IGR): Stimulation of Filamentous Actin Reorganization and Enhancement of Hsp27 Expression in Drosophila Kc Cells

The cytotoxicological responses to insect growth regulator (IGR), using tebufenozide as ecdysteroid mimic, were investigated in Drosophila Kc cells. Treatment of Kc cells with tebufenozide showed significant growth inhibition and striking morphological changes including aggregation and elongation of the cells. In order to understand the cellular mechanism underlying the response of Drosophila cells to tebufenozide, immunofluorescence microscopy was performed. We found that treatment of Kc cells...
