Direct Block of Cloned K+ Channels, Kv1.5 and Kv1.3, by Cyclosporin A, Independent of Calcineurin Inhibition
BokHeeChoi, SangJuneHahn 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 10 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2005, Vol.9 No.6 7 353-362 (10 pages)
A (CsA), an immunosuppressant, with rat brain Kv1.5 (Kv1.5) channels, which were stably expressed in Chinese hamster ovary cells, was investigated using the whole-cell patch-clamp technique. CsA reversibly blocked Kv1.5 currents at 50 mV in a reversible concentration- dependent manner with an apparent IC50 of 1.0μM. Other calcineurin inhibitors (cypermethrin, autoinhibitory peptide) had no effect on Kv1.5 and did not prevent the inhibitory effect of CsA. Fast application of CsA led to a... -
Characterization of ETB Receptor-mediated Relaxation in Precontracted Mesenteric Artery from Streptozotocin-induced Diabetic Rats

Diabetes mellitus is associated with vascular complications, including an impairment of vascular function and alterations in the reactivity of blood vessels to vasoactive substances in various vasculature. In the present study, the authors have observed endothelin-B (ETB) receptor agonist-induced relaxation in precontracted mesenteric arterial segments from streptozotocin (STZ)-induced diabetic rats, which was not shown from control rats or in other arterial segments from diabetic rats.... -
The Role of Mitochondrial ATP-sensitive Potassium Channel on Intestinal Pacemaking Activity
ByungJooKim, KiWhanKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages

50μM glibenclamide, the resting membrane potential was depolarized. At 3mM 5-HDA, an inhibitor of the mitoKATP channels, inhibited the pacemaking activity of ICCs. Both the amplitude and the frequency were decreased. At 5 mM 5-HDA, both the amplitude and the frequency were completely abolished. Diazoxide, an opener of the mitoKATP channels, was applied to examine its effect on pacemaking activity of ICCs. At 50μM concentration, the pacemaking activity of ICCs was inhibited. Both the amplitude... -
The Effect of External Divalent Cations on Intestinal Pacemaking Activity

a mean amplitude of 500⁑50 pA or 30⁑1 mV and the frequency of 18⁑2 cycles/min. Treatments of the cells with external 0 mM Ca2 stopped pacemaking activity of ICC. In the presence of 2 mM Ca2, 0 mM external Mg2 depolarized the resting membrane potential, and there was no change in the frequency of pacemaking activity. However, 10 mM external Mg2 decreased the frequency of pacemaking activity (6.75⁑1 cycles/min, n=5). We replaced external 2... -
Expression of Nitric Oxide Synthase and Endothelin-1 in Human Uterine Artery from Full-Term Pregnancies

The aim of this study was to determine the roles of ET-1 and NO on uterine blood flow in pregnancy. Uterine arteries were isolated from 17 nonpregnant and 12 pregnant women. Nonpregnant group included patients with median age of 48.6⁑2.3 years who underwent hysterectomy, because of myoma. Pregnant group included patients with median age of 31.3⁑1.4 years undergoing cesarean delivery. ET-1 and ET-2 induced concentration-dependent contraction in isolated nonpregnant and pregnant... -

patch-clamp techniques. The spontaneous action potentials were increased by DADLE in 12 cells and decreased in 3 cells. The spike frequency and resting membrane potential of these cells were increased by DADLE. The depth of afterhyperpolarization was not affected by DADLE. The potassium currents were decreased in 20 cells and increased in 5 cells. These results suggest that DADLE increases the neuronal activity of the medial vestibular nuclear neurons by altering resting membrane potential. -
Modulation of Large Conductance Ca2+-activated K+ Channel of Skin Fibroblast (CRL-1474) by Cyclic Nucleotides

Potassium channels in human skin fibroblast have been studied as a possible site of Alzheimer disease pathogenesis. Fibroblasts in Alzheimer disease show alterations in signal transduction pathway such as changes in Ca2 homeostasis and/or Ca2-activated kinases, phosphatidylinositol cascade, protein kinase C activity, cAMP levels and absence of specific K channel. However, little is known so far about electrophysiological and pharmacological characteristics of... -

(SS) on calcium channel current (IBa) in guinea-pig gastric myocytes, IBa was recorded by using whole-cell patch clamp technique in single smooth muscle cells. Nicardipine (1μM), a L-type Ca2+ channel blocker, inhibited IBa by 98⁑1.9% (n=5), however IBa was decreased in a reversible manner by application of SS. The peak IBa at 0 mV were decreased to 95⁑1.1, 92⁑1.9, 82⁑4.0, 66⁑5.8, 10⁑2.9% at 1010, 109, 108, 107, 105 M of SS,... -

in HEK-293 cells, and cytotoxicity assay was preformed using cell counting kit-8 (CCK-8). Cell survival rates were calculated using the cytotoxicity assay data and dose-response curve was fitted to the H2O2 concentration. Ionic currents were recorded in cell-attached mode. The bath solution was the normal Ringer solution and the pipette solution was high K solution. In HEK-293 cells expressing TREK-1, TREK-2, TASK-3, H2O2 induced cell death did not change in comparison to non-transfected... -
Acetylcholine Induces Hyperpolarization Mediated by Activation of K(Ca) Channels in Cultured Chick Myoblasts

by KCa channels provides driving force for Ca2 influx through the stretch-activated channels into the cells. Here, we showed that acetylcholine (ACh) also hyperpolarized the membrane of cultured chick myoblasts, suggesting that nicotinic acetylcholine receptor (nAChR) may be another pathway for Ca2 influx. Under cell-attatched patch configuration, ACh increased the open probability of KCa channels from 0.007 to 0.055 only when extracellular Ca2 was present. Nicotine, a nAChR...
