Reactive oxygen species (ROS) have been suggested to be contributory factors in complications of diabetes mellitus. In the present study, we investigated the generation of superoxide, the lipid peroxide level measured as thiobarbituric acid reactive substances, the vasorelaxation of isolated thoracic aorta and the iNOS expression in kidney of streptozotocin induced diabetic rats. Sprague Dawley rats were divided into four groups: control, ascorbate (400 mg/kg rat weight daily in drinking water),... -

high K (90 mM) external solution, which was also blocked by glibenclamide. The metabolic inhibition by the treatment with mitochondrial respiratory chain inhibitors (rotenone and antimycin) hyperpolarized the resting membrane potential of parasympathetic neurons, which was not observed in sympathetic neurons. The hyperpolarizing response to metabolic inhibition was partially blocked by glibenclamide. RT-PCR analysis revealed that MPG neurons mainly expressed the KATP channel subunits of... -
Effect of Doxorubicin on Catecholamine Release in the Isolated Perfused Rat Adrenal Gland

The present study was undertaken to investigate the effect of doxorubicin (DX) on secretion of catecholamines (CA) evoked by ACh, high K, DMPP and McN-A-343 from the isolated perfused rat adrenal gland and to establish the mechanism of its action. DX (107∼106 M) perfused into an adrenal vein for 60 min produced relatively dose- and time-dependent inhibition of CA secretory responses evoked by ACh (5.32⁓103 M), DMPP (104 M) and McN-A-343 (104 M).... -
Influence of Tacrine on Catecholamine Secretion in the Perfused Rat Adrenal Gland

selective neuronal nicotinic agonist, 104 M for 2 min) and McN-A-343 (a selective muscarinic M1-agonist, 104 M for 2 min) in relatively dose- and time- dependent manners. However, tacrine failed to affect CA secretion by high K (5.6⁓102 M). Tacrine itself at concentrations used in the present experiments did not also affect spontaneous CA output. Furthermore, in the presence of tacrine (104 M), CA secretory responses evoked by Bay-K-8644 (an activator of... -
Mediation of N-methyl-D-aspartate on Neuropeptide Y Expression Induced by Morphine in Mouse Cerebellum

The existence of opioid receptors in mammalian cerebellum except human, has not been clearly understood. In the present study, we found that NPY was inducible by morphine in the mouse cerebellar granular and Purkinje cell layers. We performed in situ RT-PCR and immunohistochemistry to characterize the NPY expression. The increase of NPY gene expression by morphine (30 mg/kg, i.p.) was inhibited by pretreatment with not only naloxone (100 mg/kg, i.p.) but also a noncompetitive NMDA antagonist,... -
Modulation of L-type Ca2+ Channel Currents by Various Protein Kinase Activators and Inhibitors in Rat Clonal Pituitary GH3 Cell Line

L-type Ca2 channels play an important role in regulating cytosolic Ca2 and thereby regulating hormone secretions in neuroendocrine cells. Since hormone secretions are also regulated by various kinds of protein kinases, we investigated the role of some kinase activators and inhibitors in the regulation of the L-type Ca2 channel currents in rat pituitary GH3 cells using the patch-clamp technique. Phorbol 12,13-dibutyrate (PDBu), a protein kinase C (PKC) activator, and... -
Role of Gap Junctions in the Endothelium-Dependent Hyperpolarization of Vascular Smooth Muscle Cells

in both endothelial and smooth muscle cells using the double patch clamp method demonstrates that these two cell types are indeed electrically coupled, indicating that they behave as a functional syncytium. The EDHF-induced hyperpolarization is produced by an activation of Ca2-sensitive K-channels that are inhibited by charybdotoxin and apamin. Agonists that release EDHF increase [Ca2]i in endothelial cells but not in smooth muscle cells. Inhibition of gap junctions with... -
The Effect of Carbon Monoxide on Contraction, Cytosolic Ca2+ Level and Ionic Currents in Guinea Pig Ileal Smooth Muscle

The aim of this study was to clarify the mechanism of the inhibitory action of carbon monoxide (CO) on contraction, by measuring cytosolic Ca2 level ([Ca2]i) and ionic currents in guinea-pig ileum. CO (10%) inhibited 40 mM KCl-induced contraction and this effect was blocked by ODQ (1μM), a soluble guanylyl cyclase (sGC) inhibitor. CO inhibited the 40 mM KCl-induced contraction without changing [Ca2]i. Cumulative addition of KCl induced a graded increase in [Ca2]i... -

(TMA-DPH) and anionic 3-[p-(6- phenyl)-1,3,5-hexatrienyl]-phenylpropionic acid (PRO-DPH) were utilized to examine the range of transbilayer asymmetric rotational mobility of the neuronal membranes. Dopamine had a greater increasing effect on the mobility of the inner monolayer as compared to the outer monolayer of the neuronal membranes. It has been proven that dopamine exhibits a selective rather than nonselective fluidizing effect within the transbilayer domains of the SPMV. -
Asymmetrical Distribution of P2Y Nucleotide Receptors in Rabbit Inner Medullary Collecting Duct Cells

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.4 6 311-318 (8 pages)
We cultured the rabbit inner medullary collecting duct (IMCD) cells as monolayers on collagen-coated membrane filters, and investigated distribution of the P2Y receptors by analyzing nucleotide-induced short circuit current (Isc) responses. Exposure to different nucleotides of either the apical or basolateral surface of cell monolayers stimulated Isc. Dose-response relationship and cross-desensitization studies suggested that at least 3 distinct P2Y receptors are expressed asymmetrically on the...
