Mechanism of Acetylcholine-induced Endothelium-dependent Relaxation in the Rabbit Carotid Artery by M3-receptor Activation
Yong-JinSong, Seong-ChunKwon 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages

[Ca2]i and vasorelaxation by ACh was siginificantly reduced by either 3μM gadolinium, 10μM lanthanum, or by 10μM SKF 96365. These results suggest that in rabbit carotid artery, ACh-evoked relaxation of 70 mM K-induced contractions appears to be mediated by the release of NO. ACh-evoked vasorelaxation is mediated via the M3 subtype, and activation of the M3 subtype is suggested to stimulate nonselective cation channels, leading to increase of [Ca2]i in endothelial cells. -
Activation of Vestibular Neurons Projecting to Autonomic Brain Stem Nuclei Following Acute Hypotension in Rats

Extracellular regulated protein kinase1/2 (pERK1/2) is one of the major regulatory factors for transcription of the c-fos oncogene in neurons. The purpose of this study was to evaluate the expression of phosphorylated ERK1/2 within the vestibular nuclei (VN) of rats following acute arterial hypotension. Following the acute arterial hypotension induced by rapid hemorrhage, a significant number of pERK1/2-immunoreactive neurons appeared bilaterally in the caudal aspect of the medial and inferior... -
Activation of Vestibular Neurons Projecting to Autonomic Brain Stem Nuclei Following Acute Hypotension in Rats

The purpose of the present study was to elucidate the possible involvement of the medial vestibular nucleus (MVN) and inferior vestibular nucleus (IVN) following acute hypotension in the vestibulo- autonomic reflex through vestibulosolitary or vestibuloventrolateral projections. Acute hypotension- induced cFos expression was assessed in combination with retrograde cholera toxin B subunit (CTb) tract tracing. After injection of CTb into the solitary region, CTb-labeled neurons were located... -

potential (AP) was significantly changed by stretch in the model simulation. The duration was decreased at initial fast phase of repolarization (AP duration at 20% repolarization level from 3.7 to 2.5 ms) and increased at late slow phase of repolarization (AP duration at 90% repolarization level from 62 to 178 ms). The resting potential was depolarized from 75 to 61 mV. This mathematical model of SACs may quantitatively predict changes in cardiomyocytes by mechanical stretch. -

characteristics of the rostral gustatory nucleus of the tractus solitarius and distribution of NO synthases (NOS). Neuronal NOS (nNOS) immunoreactivity was strongly detected along the solitary tract extending from rostral to caudal medulla. Resting membrane potentials of NTS neurons were 49.2⁑2 mV and action potential amplitudes were 68.5⁑2 mV with a mean duration measured at half amplitude of 1.7⁑0.3 ms. Input resistance, determined from the response to a 150 ms,... -
Stretch-activated K+ Channels in Rat Atrial Myocytes

Mechanical stimuli to the cardiac myocytes initiate many biochemical and physiological events. Stretch-activated cation channels have been suggested to mediate these events. In this study, cell-attached and inside-out excised-patch clamp methods were used to identify stretch-activated cation channels in adult rat atrial myocytes. Channel openings were increased in cell-attached configuration when negative pressure was applied to the pipette, and also in inside-out excised patches by negative... -
Reduction of Muscarinic K+ Channel Activity by Transferrin in Ischemic Rat Atrial Myocytes

It has been demonstrated that an unidentified cytosolic factor(s) reduces KACh channel function. Therefore, this study attempted to elucidate the cytosolic factor. Fresh cytosol isolated from normal heart (FC) depressed the KACh channel activity, but cytosol isolated from the ischemic hearts (IC) did not modulate the channel function. Electrophorectic analysis revealed that a protein of ∼80 kDa was markedly reduced or even lost in IC. By using peptide sequencing analysis and Western blot, this... -

to identify inhibitory interneurons were resting membrane potential (>70 mV) and action potential (AP) width (<0.9 msec at half amplitude). FS could be differentiated from LS, based on smaller amplitude of the AP (<∼50 mV) and shorter peak-to-trough time (P-T time) of the afterhyperpolarization (<4 msec). In addition to the shorter AP width, RSNP had the higher input resistance (>200 MΩ) and the shorter P-T time (<20 msec) than those of regular spiking pyramidal neurons. Confocal... -

Endothelins secreted from keratinocytes are intrinsic mitogens and melanogens of human melanocytes in UVB-induced hyperpigmentation. To elucidate the cellular mechanism of antimelanogenic activity of bamboo extract, the effects of three ingredients of bamboo extract on endothelin 1 (ET-1)-induced Ca2 mobilization were investigated in cultured human melanocytes. ET-1 receptors in human melanocytes were characterized by using specific antagonist, and ET-1 was found to increase intracellular... -
Curcumin Attenuates Glial Cell Activation But Cannot Suppress Hippocampal CA3 Neuronal Cell Death in i.c.v. Kanic Acid Injection Model

Kainic acid (KA) is a structural analogue of glutamate that interacts with specific presynaptic and postsynaptic receptors to potentiate the release and excitatory actions of glutamate. Systemic or intracerebroventricular (i.c.v.) administration of KA to experimental animals elicits multifocal seizures with a predominantly limbic localization, and results in neuronal death of cornu ammonia 1 (CA1), reactive gliosis and biochemical changes in the hippocampus and other limbic structures. Several...
