Involvement of ERK1/2 and JNK Pathways in 17β-estradiol Induced Kir6.2 and SK2 Upregulation in Rat Osteoblast-like Cells
Jung-WookKim, Eun-KyoungYang 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages

and regulation of gene expressions in bone cells are not well known. In the present study, to investigate how K channel expressions are regulated by estrogen, we measured changes of transcript levels of various Ca2-activated (KCa) and ATP-sensitive K channels in rat osteoblastic ROS 17/2.8 cells after treatment with estrogen. Application of 17β- estradiol (E2) for 24 h and 48 h increased mRNA and protein expressions of inwardly rectifying K channel (Kir) 6.2 and type... -
Protective Effect of Defibrotide on Splanchnic Injury following Ischemia and Reperfusion in Rats

in intestine and thrombin induced adherence of neutrophils to superior mesentric artery segments. Superoxide anion and hydrogen peroxide production in 1μM formylmethionylleucylphenylalanine (fMLP)-activated PMNs was inhibited by defibrotide in a dose-dependent fashion. Defibrotide effectively scavenged hydrogen peroxide, but not hydroxyl radical. Treatment of SAO rats with defibrotide inhibited tumor necrosis factor-α, and interleukin-1β productions in blood in comparison with untreated rats.... -
Effects of Ambroxol, S-carboxymethylcysteine, Dextromethorphan and Noscapine on Mucin Release from Airway Goblet Cells

were metabolically radiolabeled and chased for 30 min in the presence of varying concentrations of the above agents to assess the effects on 3H-mucin release. Noscapine stimulated mucin release during 30 min of treatment period in a dose-dependent manner. However, ambroxol, S-carboxymethyl-L-cysteine and dextromethorphan showed no significant effect on mucin release during 30 min of treatment period. We conclude that noscapine can affect mucin release by acting on airway mucin-secreting cells. -
Regulation of Vacuolar H+-ATPase c Gene Expression by Oxidative Stress

was lost in mutants of the GC box. We confirmed by electrophoretic mobility shift and supershift assays that Sp1 protein specifically binds to the GC box. The promoter activity was not changed by the H2O2 treatment and incubation in hypoxic chamber, however, H2O2 increased the stability of ATP6L mRNA. These data suggest that the expression of the ATP6L gene by oxidative stresses is regulated at posttranscriptional level, whereas the GC box is important in basal activities of the promoter. -
Two Types of Voltage-activated Calcium Currents in Goldfish Horizontal Cells

40 mV, and the prepulse step of 90 mV was applied before command pulse between 65 and 55 mV. The transient Ca2 current was activated by depolarization to around 50 mV from a prepulse voltage of 90 mV lasting at least 400 ms and reached a maximal value near 25 mV. On the other hand, the sustained Ca2 current was induced by pre-inactivation for less than 10 ms duration. Its activation started near 10 mV and peaked at 20... -
Morphine-induced Modulation of Nociceptive Spinal Dorsal Horn Neuronal Activities after Formalin-induced Inflammatory Pain

of formalin, iontophoretically applied morphine attenuated the naturally and electrically evoked neuronal responses of dorsal horn neurons. However, neuronal responses after the formalin-induced inflammation were significantly increased by morphine. Bicuculline, GABAA antagonist, increased the naturally and electrically evoked neuronal responses of dorsal horn neurons. This increase in neuronal responses due to bicuculline after the formalin-induced inflammation was larger than that in the... -
Higher Expression of TRPM7 Channels in Murine Mature B Lymphocytes than Immature Cells
JinKyoungKim, JaeHongKo, JooHyunNam, Ji-EunWoo, KyeongMinMin, YungEEarm, SungJoonKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2005, Vol.9 No.2 1 69-76 (8 pages)
The stimulation of B cell receptors (BCR) by ligation with antibodies did not change the amplitude of MIC current. Also, increase of extracellular [Mg2]c to enhance the Mg2 influx did not affect the BCR ligation-induced death of WEHI-231 cells. Although the level of TRPM7 was not directly related with the cell death of immature B cells, the remarkable difference of TRPM7 might indicate a fundamental change in the permeability to divalent cations during the development of B cells. -
Effects of Proinflammatory Cytokines and Natural Products on Mucin Release from Cultured Hamster Tracheal Surface Epithelial Cells

for 30 min in the presence of varying concentrations of each agent to assess the effects on 3H-mucin release. The results were as follows: TNF-alpha, CTMA and LPD increased mucin release at the highest concentration, but IL-1beta did not. We conclude that CTMA and LPD can stimulate mucin release by directly acting on airway mucin-secreting cells, and suggest that these agents should be further investigated for the possible use as mild expectorants during the treatment of chronic airway diseases. -
Effects of Fetal Mesencephalic Cell Grafts on the Intrastriatal 6-hydroxydoapmine Lesioned Rats

SNc was close to that of contralateral SNc (91.9⁑9.8%) in the transplanted animals, while the ratio was approximately 32% in sham-grafted animals. These results indicate that grafted cells restored the activity for the dopaminergic neurons located in SNc, although they were transplanted into striatum. In addition, we showed that the implanted fetal cells expressed high level of glial cell line-derived neurotrophic factor (GDNF), suggesting that the transplanted fetal cells might serve as a... -
Characterization of Acetylcholine-induced Currents in Male Rat Pelvic Ganglion Neurons
Joong-HyunPark, Kyu-SangPark, Seung-KyuCha, Keon-IlLee, Min-JungKim, Jong-YeonPark, InDeokKong, Joong-WooLee 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages

by acetylcholine (ACh). Until now, however, the properties of ACh-induced currents and its receptors in pelvic ganglia have not clearly been elucidated. In the present study, biophysical characteristics and molecular nature of nicotinic acetylcholine receptors (nAChRs) were studied in sympathetic and parasympathetic major pelvic ganglion (MPG) neurons. MPG neurons isolated from male rat were enzymatically dissociated, and ionic currents were recorded by using the whole cell variant patch clamp...
