Reoxygenation Stimulates EDRF(s) Release from Endothelial Cells of Rabbit Aorta

We have reported that hypoxia stimulates EDRF(s) release from endothelial cells and the release may be augmented by previous hypoxia. As a mechanism, it was hypothesized that reoxygenation can stimulate EDRF(s) release from endothelial cells and we tested the hypothesis via bioassay experiment. In the bioassay experiment, rabbit aorta with endothelium was used as EDRF donor vessel and rabbit carotid artery without endothelium as a bioassay test ring. The test ring was contracted by... -
The Role of Somatostatin in Nociceptive Processing of the Spinal Cord in Anesthetized Cats

Somatostatin (SOM) is one of the major neuropeptides in dorsal root ganglion cells, but its role in spinal nociceptive process has not been well known. In present study we aimed to investigate the effect of SOM on the response of dorsal horn neurons to the various types of peripheral nociceptive stimuli in anesthetized cats. Using carbon-filament microelectrode, the single cell activities of wide dynamic range neurons were recorded from the lumbosacral enlargement after noxious mechanical... -
Mechanism of Pituitary Adenylate Cyclase-Activating Polypeptide- Induced Inhibition on Catecholamine Secretion Evoked by Cholinergic Stimulation and Membrane Depolarization in the Rat Adrenal Gland

Bay-K-8644 and cyclopiazonic acid were not altered greatly in comparison to the case of PACAP-treatment only. Taken together, these results suggest that PACAP causes the marked inhibition of CA secretion evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors as well as by membrane depolarization, indicating that this effect may be mediated by inhibiting influx of extracellular calcium and release in intracellular calcium in the rat adrenomedullary chromaffin cells. -

by Mg2, the NMDA-induced responses being inhibited more strongly. Ca2 also inhibited the NMDA-induced responses current-dependently. Both inhibitory actions of Mg2 and Ca2 were additive, while Mg2 suppressed the EGTA-induced augmentation of WDR cell responses to NMDA and C-fiber stimulation. Magnesium had dual effects on the spontaneous activities of WDR cell. These experimental findings suggest that Mg2 is implicated in the modulation of pain in the rat... -
Evidence for Adenosine Triphosphate (ATP) as an Excitatory Neurotransmitter in Guinea-Pig Gastric Antrum

We explore the question of whether adenosine 5'-triphosphate (ATP) acts as an excitatory neurotransmitter in guinea-pig gastric smooth muscle. In an organ bath system, isometric force of the circular smooth muscle of guinea-pig gastric antrum was measured in the presence of atropine and guanethidine. Under electrical field stimulation (EFS) at high frequencies (>20 Hz), NO-mediated relaxation during EFS was followed by a strong contraction after the cessation of EFS (a... -
Properties of Spontaneous Activity in Gastric Smooth Muscle

Mammalian gastric smooth muscles generate spontaneous rhythmic contractions which are associated with slow oscillatory potentials (slow waves) and spike potentials. Spike potentials are blocked by organic Ca2-antagonists, indicating that these result from the activation of L-type Ca2-channel. However, the cellular mechanisms underlying the generation of slow wave remain unclear. Slow waves are insensitive to Ca2-antagonists but are blocked by metabolic inhibitors or low... -
The Mechanism of t-Butylhydroperoxide-Induced Apoptosis in IMR- 32 Human Neuroblastoma Cells

Apoptosis has been implicated in the pathophysiological mechanisms of various neurodegenerative diseases. In a variety of cell types, oxidative stress has been demonstrated to play an important role in the apoptotic cell death. However, the exact mechanism of oxidative stress-induced apoptosis in neuronal cells is not known. In this study, we induced oxidative stress in IMR-32 human neuroblastoma cells with tert- butylhydroperoxide (TBHP), which was confirmed by significantly reduced... -
Mechanism of Membrane Hyperpolarization by Extracellular K+ in Resistance-sized Cerebral Arterial Muscle Cell of Rabbit

currents at [K]o of 6 and 15 mM showed that the barium- sensitive outward current increased at 15 mM K. From the above results the following were concluded. Barium-sensitive K channel activity increased when [K]o is elevated and this leads to an increase in K-outward current. Consequently, the membrane potential hyperpolarizes, leading to the relaxation of resistant arteries, and this is thought to contribute to an increase in the local blood flow of brain. -

Frequency-force relationships (FFR) were studied in electrically field stimulated rat left atria (LA) by reducing the stimulation frequency from resting 3 Hz to test frequencies (0.1-1 Hz) for 5 minutes. The twitch amplitudes of LA elicited the typical negative staircases with 3-phased changes: the initial rapid increase, the second decrease and the following plateau at test frequencies. Verapamil (3⁓105 M) pretreatment elicited frequency-dependent suppression of the twitch... -

K current) in the mechanism. Mulvany myograph was used to study 24 rats, 18 rabbits, and 10 humans MCAs (216⁑3 ㄍm, 347⁑7 ㄍm, and 597⁑39 ㄍm in diameter when stretched to a tension equivalent to 55 mmHg). High K (125 mM) and PGF2α (1∼10 ㄍM) induced concentration-dependent contractions in all 3 species, while histamine (10∼50 ㄍM) evoked contraction only in the rabbits and induced relaxation in the rats and humans. Addition of K...
