hours with 200 ng/ml LPS (in vitro). L-type Ca2 current (ICa,L) and transient outward K current (Ito) were measured using whole cell patch clamp techniques. Peak ICa,L was reduced in endotoxemic myocytes (ex vivo; 6.00.4 pA/pF, P<0.01) compared to normal myocytes (control; 10.90.6 pA/pF). Exposure to endotoxin in vitro also attenuated ICa,L (8.40.4 pA/pF, P<0.01). The amplitude of Ito on depolarization to 60 mV was reduced in endotoxin treated myocytes (16.51.5 pA/pF, P<0.01, ex... -
Hemorrhage- and Restraint-induced Analgesia in Male and Female Conscious Rats
SungYunOh, HeeSeokHan, DongKukAhn, JaeSikPark 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 9 Pages

It is well known that stress induces analgesia. This study was designed to demonstrate the stress-induced analgesia by employing hemorrhage and restraint and to investigate its mechanism and sex difference. The degree of pain was assessed by measuring the magnitude of jaw opening reflex produced by a noxious electrical stimulation in the dental pulp and by measuring the latency to withdraw the tail from a heat ray. Restraint showed an antinociceptive response. A significant increase in pain... -
Decreased Voltage Dependent K+ Currents in Cerebral Arterial Smooth Muscle Cells of One-Kidney, One-Clip Goldblatt Hypertensive Rat

whole cell mode of patch-clamp technique, the membrane potential and the 4-AP-sensitive Kv current in cerebral arterial smooth muscle cells were compared between normotensive rat and one-kidney, one-clip Goldblatt hypertensive rat (1K,1C-GBH rat). Cell capacitance of hypertensive rat was similar to that of normotensive rat. Cell capacitance of normotensive rat and 1K,1C-GBH rat were 20.8⁑2.3 and 19.5⁑1.4 pF, respectively. The resting membrane potentials measured in current clamp mode... -
Effects of Cl Channel Blockers on the Cardiac ATP-sensitive K+ Channel

manner with IC50 of 0.24 and 927μM, respectively. The inhibitory action of DIDS was irreversible whereas that of niflumic acid was reversible. Furthermore, DIDS-induced block was not recovered despite exposure to ATP (1 mM). In cell-attached and inside-out patches, DIDS blocked the pinacidil- or 2,4-dinitrophenol (DNP)-induced KATP channel openings. In contrast, niflumic acid did not block the pinacidil-induced KATP channel openings in inside-out patches, but inhibited it in cell-attached... -
Roles of Na+-Ca2+ Exchange in the Negative Force-Frequency Relationship

the basal tone during frequency reduciton. There were no differences in 45Ca uptakes between 0.3 Hz and 3 Hz stimulation except at 1 min when it was significantly low at 0.3 Hz than 3 Hz, illustrating net Ca2 losses. Monensin pretreatment enhanced the rate of this Ca2 loss. Taken together, it is concluded that Na-Ca2 exchange extrudes more SR released Ca2 out of the cell in proportion to the frequency, resulting in the negative rate staircase in the rat LA. -
Effects of Neonatal Footshock Stress on Glucocorticoid and 5- HT2A/2C Receptor Bindings and Exploratory Behavior
DongGooKim, SeoulLee, DongWonKang, JongSuLim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 9 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.6 4 677-685 (9 pages)
To investigate the effects of neonatal stress on behavior and neurochemistry, rats were exposed to the footshock stress on postnatal day (PND) 14 or PNDs 14 and 21. Rats were exposed to uncontrollable electric shocks delivered to the floor with a constant current (0.8 mA) for 5 sec period. Daily sessions consisted of 60 trials on a random time schedule with an average of 55 sec. The first exposure to footshocks on PND 14 decreased body weight gain for 1 day. However, the second exposure to... -
Effects of Dopamine and Haloperidol on Morphine-induced CREB and AP-1 DNA Binding Activities in Differentiated SH-SY5Y Human Neuroblastoma Cells
Soo-kyungKimGee-younKwon 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.6 3 671-676 (6 pages)
In the present study, we first examined whether the changes in the DNA binding activities of the transcription factors, cAMP response element binding protein (CREB) and activator protein-1 (AP-1) mediate the long-term effects of morphine in differentiated SH-SY5Y human neuroblastoma cells. The increases in CREB and AP-1 DNA binding activities were time-dependent up to 6 days of morphine treatment (1, 4, and 6 days). However, the significant reduction in the DNA binding activities of CREB and... -

distinctive biological activity of bombesin like peptide. On the contrary, the biological activity of BLI-K5 was similar to that of bombesin but its binding affinity to an anti-bombesin serum was low. The results indicate that three heterogeneous forms of BLI were coexpressed with bombesin and BLI-K2 in the skin of B. orientalis. All forms of the purified BLI in the present study were immunologically active but only BLI-K5 possessed the distinctive biological activity of bombesin like peptide. -
cAMP-Dependent Signalling is Involved in Adenosine-Stimulated Cl Secretion in Rabbit Colon Mucosa

(NPPB), glibenclamide. Amiloride, an epithelial Na channel blocker, and 4,4-diisothiocyanato-stilbene-2,2-disulphonate (DIDS), a Ca2-activated Cl channel blocker, had no effect. In the mucosa pre-stimulated with forskolin, adenosine did not show any additive effect, whereas carbachol resulted in a synergistic potentiation of the ISC response. The adenosine response was inhibited by 10 ㄍM H-89, an inhibitor of protein kinase A. These results suggest that the... -
Inhibitory Effect of Caffeine on Carbachol-Induced Nonselective Cationic Current in Guinea-Pig Gastric Myocytes
SungJoonKim, KyungWanMin, YoungChulKim, SangJinLee, InsukSo, KiWhanKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 7 Pages

In gastrointestinal smooth muscle, muscarinic stimulation by carbachol (CCh) activates nonselective cation channel current (ICCh) which is facilitated by intracellular [Ca2] increase. Caffeine is widely used in experiments to mobilize Ca2 from intracellular stores. This study shows a strong inhibitory effect of caffeine on ICCh in guinea-pig gastric myocyte. In this study, the underlying mechanism of the inhibitory effect of caffeine was investigated. ICCh was completely...
