Involvement of Spontaneously Formed Cyclic Nucleotides in Cat Gastric Muscle Relaxation

Muscle strips and muscle cells from cat stomach were used to investigate whether spontaneously formed cyclic nucleotides were involved in the inhibition of gastric smooth muscle contraction. A phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine (IBMX), increased the levels of both cyclic GMP (cGMP) and cyclic AMP (cAMP) in resting state cells, while decreasing acetylcholine-induced muscle contraction. Under the influence of IBMX, SQ22536, an adenylyl cyclase inhibitor and methylene blue, a... -
Moderate Elevation of Extracellular K+ Concentration Induces Vasorelaxation in Isolated Rat, Rabbit and Human Cerebral Arteries Role of Na Pump and Ba-Sensitive Process

species, while histamine (10∼50 ㄍM) evoked contraction only in the rabbits and induced relaxation in the rats and humans. Addition of K (2∼10 mM) to the control solution induced vasorelaxations. These effects were inhibited by the pretreatment with both ouabain (10 ㄍM) and Ba2 (0.1∼0.3 mM) in the rat, but only with ouabain (10 ㄍM) in the rabbit and human. These results suggest that K-induced vasorelaxation occurs via the stimulation of electrogenic Na... -
Sensitivity of Rabbit Cerebral Artery to Serotonin is Increased with the Moderate Increase of Extracellular K+

with the moderate increase of [K]O, using Mulvany-type myograph and [Ca2]c measurement. The contractions of basilar artery and branch of middle cerebral artery induced by histamine were not increased with the elevation of [K]O from 6 mM to 9 mM or 12 mM. On the contrary, the contractions induced by serotonin were significantly increased with the elevation of [K]O. The contractions were also significantly increased by the treatment with nitro-L-arginine (104 M... -
Characteristics of Ca2+ Stores in Rabbit Cerebral Artery Myocytes

of ryanodine, histamine could induce repetitive rise of [Ca2]c while the amplitude of peak rise became stepwisely decreased and eventually disappeared. These results suggest that two different Ca2-release mechanisms (caffeine-sensitive and histamine-sensitive) are present in rabbit cerebral artery myocyte and the corresponding pools overlap each other functionally. Increase of [Ca2]c by histamine seems to partially activate ryanodine receptors present in caffeine-sensitive... -
Cromakalim Blocks Membrane Phosphoinositide Activated Signals in the Guinea Pig Lung Mast Cells Stimulated with Antigen-Antibody Reactions

sensitized mast cells was decreased by calphostin C, staurosporine and cromakalim, respectively. The PLA2 activity was increased in the activated mast cells. The pretreatment of cromakalim did not significantly decrease PLA2 activity. These data show that cromakalim inhibits histamine release by continuously inhibiting signal transduction processes which is mediated via PLC pathway during mast cell activation, but that cromakalim does not affect PLA2 activity related to leukotriene release. -
Influence of Glucocorticoids on Cholinergic Stimulation-Induced Catecholamine Secretion from the Rat Adrenal Medulla

as membrane-depolarization, Ca2+ channel activation and the release of intracellular Ca2+. Furthermore, even in the presence of betamethasone (30ՌM), CA secretion evoked by ACh, excess K+, DMPP and McN-A-343 was also markedly inhibited. Taken together, the present results suggest that glucocorticoids cause the marked inhibition of CA secretion evoked by both cholinergic nicotinic and muscarinic receptor stimulation from the isolated perfused rat adrenal gland, indicating strongly that this... -

kinase C inhibitor, genistein, a protein tyrosine kinase inhibitor and chlorpromazine, a calmodulin inhibitor, inhibited superoxide production by fMLP, but they did not further affect the action of DBcAMP on the stimulatory action of fMLP. DBcAMP, histamine, adenosinetheophylline and genistein inhibited myeloperoxidease release evoked by fMLP, whereas BrcGMP, sodium nitroprusside and staurosporine did not affect it. The elevation of [Ca2]i evoked by fMLP was inhibited by genistein...
