IC50 of 0.24 and 927μM, respectively. The inhibitory action of DIDS was irreversible whereas that of niflumic acid was reversible. Furthermore, DIDS-induced block was not recovered despite exposure to ATP (1 mM). In cell-attached and inside-out patches, DIDS blocked the pinacidil- or 2,4-dinitrophenol (DNP)-induced KATP channel openings. In contrast, niflumic acid did not block the pinacidil-induced KATP channel openings in inside-out patches, but inhibited it in cell-attached patches. DIDS and... -
Opening of ATP-sensitive K+ Channel by Pinacidil Requires Serine/ Threonine Phosphorylation in Rat Ventricular Myocytes
The Korean Journal of Physiology & Pharmacology 11 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.3 7 293-303 (11 pages)
The influences of specific protein phosphatase and protein kinase inhibitors on the ATP-sensitive K (KATP) channel-opening effect of pinacidil were investigated in single rat ventricular myocytes using patch clamp technique. In cell-attached patches, pinacidil (100 μM) induced the opening of the KATP channel, which was blocked by the pretreatment with H-7 (100 μM) whereas enhanced by the pretreatment with genistein (30 μM) or tyrphostin A23 (10 μM). In inside-out patches, pinacidil... -
The Effects of Sera from Amyotrophic Lateral Sclerosis Patients on Neuromuscular Transmission and Calcium Channels in Mice
The Korean Journal of Physiology & Pharmacology 17 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.1 13 101-117 (17 pages)
Amyotrophic lateral sclerosis (ALS) is a degenerative neuromuscular disease of unknown etiology in which the upper and lower motor neurons are progressively destroyed. Recent evidences support the role of autoimmune mechanisms in the pathogenesis of ALS. This study investigated the effects of sera from ALS patients on neuromuscular transmission in phrenic nerve-hemidiaphragm preparations and on calcium currents of single isolated dorsal root ganglion (DRG) cells in mice. Mice were injected with... -
The Mechanism of t-Butylhydroperoxide-Induced Apoptosis in IMR- 32 Human Neuroblastoma Cells
The Korean Journal of Physiology & Pharmacology 10 Pages
The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.1 3 19-28 (10 pages)
Apoptosis has been implicated in the pathophysiological mechanisms of various neurodegenerative diseases. In a variety of cell types, oxidative stress has been demonstrated to play an important role in the apoptotic cell death. However, the exact mechanism of oxidative stress-induced apoptosis in neuronal cells is not known. In this study, we induced oxidative stress in IMR-32 human neuroblastoma cells with tert- butylhydroperoxide (TBHP), which was confirmed by significantly reduced... -

The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.1 1 1-10 (10 pages)
We sought to find out the mechanism of vascular relaxation by extracellular K concentration ([K]o) in the cerebral resistant arteriole from rabbit. Single cells were isolated from the cerebral resistant arteriole, and using voltage-clamp technique barium-sensitive K currents were recorded, and their characteristics were observed. Afterwards, the changes in membrane potential and currents through the membrane caused by the change in [K]o was observed. In the smooth... -
The Role of Intracellular Mg2+ in Regulation of Ca2+-activated K+ Channel in Pulmonary Arterial Smooth Muscle Cells of the Rabbit
The Korean Journal of Physiology & Pharmacology 6 Pages
The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.5 9 611-616 (6 pages)
Although the Ca2+-activated K+ (IK,Ca) channel is known to play an important role in the maintenance of resting membrane potential, the regulation of the channel in physiological condition is not completely understood in vascular myocytes. In this study, we investigated the role of cytoplasmic Mg2+ on the regulation of IK,Ca channel in pulmonary arterial myocytes of the rabbit using the inside-out patch clamp technique. Mg2+ increased open probability (Po), but decreased the magnitude of single... -
The Effects of Intracellular Monocarboxylates on the ATP-sensitive Potassium Channels in Rabbit Ventricular Myocytes
The Korean Journal of Physiology & Pharmacology 9 Pages
The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.5 6 581-589 (9 pages)
A regulating mechanism of the ATP-sensitive potassium channels (KATP channels) is yet to fully explained. This study was carried out to investigate the effects of intracellular application of monocarboxylates (acetate, formate, lactate, and pyruvate) on KATP channels in isolated rabbit ventricular myocytes. Single channel currents of KATP channels were recorded using the excised inside-out or permeabilized attached (open-cell) patch-clamp technique at room temperature. Intracellular application... -
Tetrachloroauric Acid Depresses the Activation Processes of Phagocytic Cells
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.3 13 377-384 (8 pages)
thapsigargin-pretreated neutrophils was decreased by the addition of tetrachloroauric acid and genistein. The effect of staurosporine on intracellular Ca2+ mobilization was not observed. In conclusion, tetrachloroauric acid may suppress neutrophil responses through its inhibitory action on elevation of intracellular Ca2+ level and protein kinase C. It might exhibit an inhibitory effect on the action of protein tyrosine kinase. Tetrachloroauric acid depresses cytokine production by macrophages. -

ATP-sensitive K+ channels (KATP) were not identified in gastric smooth muscle cells. However, in tension recording of intact gastric circular muscle, lemakalim of KATP channels opener in other tissues suppressed mechanical contractions and this effect was blocked by glibenclamide, a specific inhibitor of KATP channels. The aims of this study were to investigate whether KATP channels exist in gastric smooth muscle of guinea-pig and to know its physiological role. Whole cell K'+ curr‘ents...
