Calcium (Ca2+) is an intracellular second messenger associated with neuronal plasticity of the central nervous system. The calcium-binding proteins regulate the Ca2+-mediated signals in the cytoplasm and buffer the calcium concentration. This study examined temporal changes of three calcium-binding proteins (calretinin, calbindin and parvalbumin) in the medial vestibular nucleus (MVN) during vestibular compensation after unilateral labyrinthectomy (UL) in rats. Rats underwent UL, and the... -
Influence of SKF81297 on Catecholamine Release from the Perfused Rat Adrenal Medulla

a selective agonist of dopaminergic D1 receptor, on the secretion of catecholamines (CA) evoked by cholinergic stimulation and membrane-depolarization in the isolated perfused rat adrenal gland, and also to elucidate the mechanism involved. SKF81297 (10∼100μM) perfused into an adrenal vein for 60 min produced dose- and time-dependent inhibition of CA secretory responses evoked by ACh (5.32 mM), high K+ (56 mM), DMPP (100μM) and McN-A-343 (100μM). Also, in adrenal glands loaded with SKF81297... -
Developmental Disability Animal Model Based on Neonatal Lipopolysaccharide with Altered 5-HT Function

Developmental disability shows life-long behavioral abnormality with no significant physical malformation. This study was undertaken to develop an animal model for developmental disability by using two-factor approach. Lipopolysaccharide (LPS), a bacterial toxin, and NAN-190, a 5-HT1A receptor antagonist, were administered to Sprague-Dawley rats on postnatal day (PND) 5 to induce inflammation and an altered 5-HT system, respectively. Long-term alteration of behavior occurred in the drug-treated... -
Tetanus-induced LTD of Developing MNTB-LSO Synapses in Rat is Dependent on Postsynaptic Ca2+

Because synaptic refinement of medial nucleus of trapezoid body (MNTB) - lateral superior olive (LSO) synapses is most active during the first postnatal week and the long term depression (LTD) has been suggested as one of its mechanisms, LTD of MNTB-LSO synapses was investigated in neonatal rat brain stem slices with the whole cell voltage clamp technique. In Mg2 free condition, tetanus (10 stimuli at 10 Hz for 2 min) in the current clamp mode induced a robust LTD of isolated D,... -
A1 Receptor-mediated Protection against Amyloid Beta-induced Injury in Human Neuroglioma Cells

Adenosine has been reported to provide cytoprotection in the central nervous systems as well as myocardium by activating cell surface adenosine receptors. However, the exact target and mechanism of its action still remain controversial. The present study was performed to examine whether adenosine has a protective effect against Aβ-induced injury in neuroglial cells. The astrocyte-derived human neuroglioma cell line, A172 cells, and Aβ25∼35 were employed to produce an experimental Aβ-induced... -
Influence of ω-Conotoxin GVIA, Nifedipine and Cilnidipine on Catecholamine Release in the Rat Adrenal Medulla

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2007, Vol.11 No.1 4 21-30 (10 pages)
The present study was designed to establish comparatively the inhibitory effects of cilnidipine (CNP), nifedipine (NIF), and ω-conotoxin GVIA (CTX) on the release of CA evoked by cholinergic stimulation and membrane depolarization from the isolated perfused model of the rat adrenal medulla. CNP (3 μM), NIF (3μM), and CTX (3μM) perfused into an adrenal vein for 60 min produced greatly inhibition in CA secretory responses evoked by ACh (5.32⁓103 M), DMPP (104 M for 2 min),... -
Neurovascular Mechanisms in Stroke, Neurodegeneration and Recovery

The emerging concept of the“neurovascular unit”may enable a powerful paradigm shift for neuroscience. Instead of a pure focus on the“neurobiology”of disease, an opportunity now exists to return to a more integrative approach. The neurovascular unit emphasizes that signaling between vascular and neuronal compartments comprise the basis for both function and dysfunction in brain. Hence, brain disorders are not just due to death of neurons, but instead manifested as cell signaling perturbations... -
Effects of Zinc on Spontaneous Miniature GABA Release in Rat Hippocampal CA3 Pyramidal Neurons
Byung-JuChoi, Il-SungJang 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2006, Vol.10 No.2 1 59-64 (6 pages)
The effects of Zn2 on spontaneous glutamate and GABA release were tested in mechanically dissociated rat CA3 pyramidal neurons which retained functional presynaptic nerve terminals. The spontaneous miniature excitatory and inhibitory postsynaptic currents (mEPSCs and mIPSCs, respectively) were pharmacologically isolated and recorded using whole-cell patch clamp technique under voltage-clamp conditions. Zn2 at a lower concentration (30μM) increased GABAergic mIPSC frequency without... -
Direct Block of Cloned K+ Channels, Kv1.5 and Kv1.3, by Cyclosporin A, Independent of Calcineurin Inhibition

in a tail crossover phenomenon. Block of Kv1.5 by CsA was use-dependent. CsA also blocked Kv1.3 currents at 50 mV in a reversible concentration-dependent manner with an apparent IC50 of 1.1μM. The same effects of CsA on Kv1.3 were also observed in excised inside-out patches when applied to the internal surface of the membrane. The present results suggest that CsA acts directly on Kv1.5 currents as an open-channel blocker, independently of the effects of CsA on calcineurin activity. -
Effects of Nitric Oxide on Inhibitory Receptors of Rod Bipolar Cells of Rat Retina

The effects of nitric oxide (NO) on inhibitory neurotransmitter receptors and some types of inhibitory receptors in dissociated rod bipolar cell (RBC) were investigated. In the whole cell voltage-clamping mode, the gamma-aminobutyric acid (GABA) activated current showed both sustained and transient components. GABA activated transient current was fully blocked by bicuculine, a GABAA receptor antagonist. The cis-4-aminocrotonic acid (CACA), a GABAC receptor agonist, evoked the sustained current...
