a widely used tyrosine kinase inhibitor, on cloned Shaw-type K currents, Kv3.1 which were stably expressed in Chinese hamster ovary (CHO) cells, using the whole-cell configuration of patch-clamp techniques. In whole-cell recordings, genistein at external concentrations from 10 to 100μM accelerated the rate of inactivation of Kv3.1 currents, thereby concentration-dependently reducing the current at the end of depolarizing pulse with an IC50 value of 15.71⁑0.67μM and a Hill... -

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2005, Vol.9 No.6 7 353-362 (10 pages)
The interaction of cyclosporine A (CsA), an immunosuppressant, with rat brain Kv1.5 (Kv1.5) channels, which were stably expressed in Chinese hamster ovary cells, was investigated using the whole-cell patch-clamp technique. CsA reversibly blocked Kv1.5 currents at 50 mV in a reversible concentration- dependent manner with an apparent IC50 of 1.0μM. Other calcineurin inhibitors (cypermethrin, autoinhibitory peptide) had no effect on Kv1.5 and did not prevent the inhibitory effect of CsA.... -

in the heart. This possibility was examined in this study by comparing NCX-induced extrusion of Ca2 after its release from SR in the presence and absence of global Ca2i transient in the isolated single rat ventricular myocytes by using patch-clamp technique in a whole-cell configuration. Global Ca2i transient was controlled by an internal dialysis with different concentrations of BAPTA added in the pipette. During stimulation with a ramp pulse from 100 mV to 10... -

To prove the buffering contribution of mitochondria to the increase of intracellular Ca2+ level ([Ca2+]i) via background nonselective cation channel (background NSCC), we examined whether inhibition of mitochondria by protonophore carbonylcyanide m-chlorophenylhydrazone (CCCP) affects endothelial Ca2+ entry and Ca2+ buffering in freshly isolated rabbit aortic endothelial cells (RAECs). The ratio of fluorescence by fura-2 AM (R340/380) was measured in RAECs. Biological state was checked by... -

A variety of G protein coupled receptors (GPCRs) are expressed in the presynaptic terminals of central and peripheral synapses and play regulatory roles in transmitter release. The patch-clamp whole-cell recording technique, applied to the calyx of Held presynaptic terminal in brainstem slices of rodents, has made it possible to directly examine intracellular mechanisms underlying the GPCR-mediated presynaptic inhibition. At the calyx of Held, bath-application of agonists for GPCRs such as GABAB... -

the rat primary visual cortex, using patch-clamp recording and confocal reconstruction, and classified inhibitory interneurons into fast spiking (FS), late spiking (LS), burst spiking (BS), and regular spiking non-pyramidal (RSNP) neurons according to their electrophysiological characteristics. Global parameters to identify inhibitory interneurons were resting membrane potential (>70 mV) and action potential (AP) width (<0.9 msec at half amplitude). FS could be differentiated from LS,... -
Effects of Noradrenaline on the Membrane Potential of Prostatic Neuroendocrine Cells of Rat

The prostate gland contains numerous neuroendocrine cells that are believed to influence the function of the prostate gland. Our recent study demonstrated the expression of both α1- and α2-ARs, signaling the release of stored Ca2 and the inhibition of N-type Ca2 channels, respectively, in rat prostate neuroendocrine cells (RPNECs). In this study, the effects of NA on the resting membrane potential (RMP) of RPNECs were investigated using a whole-cell patch clamp method. Fresh... -

Effects of oxidized low-density lipoprotein (ox-LDL), l-α-stearoyl-lysophosphatidylcholine (LPC), on intracellular Ca2 concentration were examined in mouse endothelial cells by measuring intracellular Ca2 concentration ([Ca2]i) with fura 2-AM and reverse transcription-polymerase chain reaction (RT-PCR). LPC increased [Ca2]i under the condition of 1.5 mM [Ca2]o but did not show any effect under the nominally Ca2-free condition. Even after the store... -

F2α (PGF2α) on membrane potentials, Ca2-activated K (KCa) channels, and delayed rectifier K (KV) channels using the patch-clamp technique in single rabbit middle cerebral arterial smooth muscle cells. PGF2α significantly hyperpolarized membrane potentials and increased outward whole-cell K currents. PGF2α increased open-state probability of KCa channels without the change of the open and closed kinetics. PGF2α increased the amplitudes of KV currents with a... -

Suppressive role of Na-Ca2 exchange in myocardial tension generation was examined in the negative frequency-force relationship (FFR) of electric field stimulated left atria (LA) from postnatal developing rat heart and in the whole-cell clamped adult rat ventricular myocytes with high concentration of intracellular Ca2 buffer (14 mM EGTA). LA twitch amplitudes, which were suppressed by cyclopiazonic acid in a postnatal age-dependent manner, elicited frequency-dependent and...
