Changes of the Level of G Protein α-subunit mRNA by Withdrawal from Morphine and Butorphanol
SeikwanOh 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 9 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.4 4 291-299 (9 pages)
Morphine or butorphanol was continuously infused into cerebroventricle (i.c.v.) with the rate of 26 nmol/ μl/h for 3 days, and the withdrawal from opioid was rendered 7 hrs after the stopping of infusion. The expression of physical dependence produced by these opioids was evaluated by measuring the naloxone- precipitated withdrawal signs. The withdrawal signs produced in animals dependent on butorphanol (kappa opioid receptor agonist) were similar to those of morphine (mu opioid receptor... -

In order to provide a basis for studying the molecular mechanism of pharmacological action of local anesthetics and to develop a fluorescence spectroscopic method which can detect the microviscosity of native and model membranes using intramolecular excimerization of 1,3-di(l-pyrenyl)propane (Py-3-Py), we examined the effect of lidocaine·HCl on the microviscosity of model membranes of phosphatidylcholine fraction extracted from synaptosomal plasma membrane vesicles (SPMVPC). The excimer to... -
Relatoinship between Sarcoplasmic Reticular Calcium Release and Na+-Ca2+ Exchange in the Rat Myocardial Contraction

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.3 4 197-210 (14 pages)
Suppressive role of Na-Ca2 exchange in myocardial tension generation was examined in the negative frequency-force relationship (FFR) of electric field stimulated left atria (LA) from postnatal developing rat heart and in the whole-cell clamped adult rat ventricular myocytes with high concentration of intracellular Ca2 buffer (14 mM EGTA). LA twitch amplitudes, which were suppressed by cyclopiazonic acid in a postnatal age-dependent manner, elicited frequency-dependent and... -
Effect of Propofol, an Intravenous Anesthetic Agent, on KATP Channels of Pancreatic β-cells in Rats

ATP-sensitive potassium channels (KATP channels) play an important role in insulin secretion from pancreatic beta cells. We have investigated the effect of propofol on KATP channels in cultured single pancreatic beta cells of rats. Channel activity was recorded from membrane patches using the patch-clamp technique. In the inside-out configuration bath-applied propofol inhibited the KATP channel activities in a dose-dependent manner. The half-maximal inhibition dose (ED50) was 48.6⁑8.4μM... -
Effect of Capsaicin on Delayed Rectifier K+ Current in Adult Rat Dorsal Root Ganglion Neurons

dose dependent manner, and the concentration-dependence curve was well described by the Hill equation with KD value of 19.1μM. The blocking effect of capsaicin was reversible. Capsaicin (10 μM) shifted the steady- state inactivation curve in the hyperpolarizing direction by about 15 mV and increased the rate of inactivation. The voltage dependence of activation was not affected by capsaicin. These multiple effects of capsaicin may suggest that capsaicin bind to the region of K channel,... -
Role of K+ Channels to Resting Membrane Potential of Rabbit Middle Cerebral Arterial Smooth Muscle Cells

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.6 2 547-554 (8 pages)
The aim of the present study is to investigate the contribution of Ca2-activated K (KCa) channels and delayed rectifier K (KV) channels to the resting membrane potential (RMP) in rabbit middle cerebral arterial smooth muscle cells. The RMP and membrane currents were recorded using the whole-cell patch configuration and single KCa channel was recorded using the outside-out patch configuration. Using the pipette solution containing 0.05 mM EGTA, the RMP was... -
Changes in Phosphate Transporter Activity Evaluated by Phosphonoformic Acid Binding in Cadmium-Exposed Renal Brush-Border Membranes
JinMoChung, DoWhanAhn, KyoungRyongKim, YangSaengPark 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 7 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.5 8 513-519 (7 pages)
Direct exposure of renal tubular brush-border membranes (BBM) to free cadmium (Cd) causes a reduction in phosphate (Pi) transport capacity. Biochemical mechanism of this reduction was investigated in the present study. Renal proximal tubular brush-border membrane vesicles (BBMV) were isolated from rabbit kidney outer cortex by Mg precipitation method. Vesicles were exposed to 50∼200 μM CdCl2 for 30 min, then the phosphate transporter activity was determined. The range of Cd concentration... -
Decreased Voltage Dependent K+ Currents in Cerebral Arterial Smooth Muscle Cells of One-Kidney, One-Clip Goldblatt Hypertensive Rat

Kv current was smaller in 1K,1C-GBH rat than in normotensive rat. The voltage dependence of steady-state activation and inactivation of Kv channel determined by using double-pulse protocol showed no significant difference. These results suggest that 4-AP-sensitive Kv channels play a major role in the regulation of membrane potential in cerebral arterial smooth muscle cells and alterations of 4-AP- sensitive KV channels would contribute to hypopolarization of membrane potential in 1K,1C-GBH rat. -
Thyroid Hormone-Induced Alterations of Ca2+-ATPase and Phospholamban Protein Expression in Cardiac Sarcoplasmic Reticulum
HaeWonKim, KyungMinNoh, MiyoungPark, HeeRanLee, EunHeeLee 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.2 13 223-230 (8 pages)
Alterations of cardiovascular function associated with various thyroid states have been studied. In hyperthyroidism left ventricular contractility and relaxation velocity were increased, whereas these parameters were decreased in hypothyroidism. The mechanisms for these changes have been suggested to include alterations in the expression and/or activity levels of various proteins; α-myosin heavy chain, β- myosin heavy chain, β-receptors, the guanine nucleotide-binding regulatory protein, and... -
The Effects of Sera from Amyotrophic Lateral Sclerosis Patients on Neuromuscular Transmission and Calcium Channels in Mice
HaiDunYan, JimokKim, SungJunJung, JunKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 17 Pages

Amyotrophic lateral sclerosis (ALS) is a degenerative neuromuscular disease of unknown etiology in which the upper and lower motor neurons are progressively destroyed. Recent evidences support the role of autoimmune mechanisms in the pathogenesis of ALS. This study investigated the effects of sera from ALS patients on neuromuscular transmission in phrenic nerve-hemidiaphragm preparations and on calcium currents of single isolated dorsal root ganglion (DRG) cells in mice. Mice were injected with...
