Effects of Arachidonic Acid on the Calcium Channel Current (IBa) and on the Osmotic Stretch-induced Increase of IBa in Guinea-Pig Gastric Myocytes
The Korean Journal of Physiology & Pharmacology 9 Pages
The Korean Journal of Physiology & Pharmacology 1997, Vol.1 No.4 10 435-443 (9 pages)
We employed the whole-cell patch clamp technique to investigate the effects of arachidonic acid (AA) on barium inward current through the L-type calcium channels (IBa) and on osmotic stretch-induced increase of IBa in guinea-pig antral gastric myocytes. Under isosmotic condition, AA inhibited IBa in a dose-dependent manner to 91.1⁑1.4, 72.0⁑3.2, 46.0⁑1.8, and 20.3⁑2.3% at 1, 5, 10, 30 mM, respectively. The inhibitory effect of AA was not affected by 10 ㄍM... -
Two Types of Voltage-dependent Outward Potassium Currents in Smooth Muscle Cells of Rabbit Basilar Atery
The Korean Journal of Physiology & Pharmacology 15 Pages
The Korean Journal of Physiology & Pharmacology 1997, Vol.1 No.2 7 169-183 (15 pages)
We have investigated the two types of voltage-dependent outward potassium (K) currents, i.e. delayed rectifier K current (IK(V)) and 'A-like' transient outward K current (Ito) with patch-clamp technique in single smooth muscle cells (SMCs) isolated from rabbit basilar artery, and investigated the characteristics of them. The time-courses of activation were well fitted by exponential function raised to second power (n2) in IK(v) and fourth power (n4) in Ito. The activation, inactivation and... -

In the present study, we observed change in intracellular Ca2+([Ca2+]i) as measured with the fluorescent Ca2+-indicator fura-2 in association with force development of the rat basilar arteries during activation byK+ depolarizing solution and U46619, a thromboxane analogue, in the absence and the presence of calcitonin-gent related peptide (CGRP). CGRP (30 and 100 nM) caused a concentration-dependent inhibition of U46619-induced contraction with decrease in [Ca2+]i, whereas it did not exert any... -
Modulatory Effect of the Tyrosine Kinase and Tyrosine Phosphatase on the ACh-activated K+ Channel in Adult Rat Atrial Cells
Chang, Kyeong-Jae, Rhie, Sang-Ho, Heo, Ilo, Kim, Yang-Mi, Haan, Jae-Hee, Hong, Seong-Geun 대한생리학회 대한생리학회지 10 Pages
3 209-218 (10 pages)
Acetylcholine (ACh) activates the inwardly rectifying muscarinic K+ channel in rat atrial cells via pertussis toxin (PTX)-sensitive G-protein (Gk) coupled with the muscarinic receptor (mAChR). Although this K+ (KACh) channel function has reported to be modulated by the phosphorylation process, a kinase and phosphatase involved in these processes are still unclear. Since either PKA or PKC was not effective on this ATP-modulation, the present study examined the possible involvement of the protein... -
Contractile and Electrical Responses of Guinea-pig Gastric Smooth Muscle to Bradykinin
Kim, Chul-Soo, Jun, Jae-Yeoul, Kim, Sung-Joon, So, In-Suk, Kim, Ki-Whan 10 Pages
5 233-242 (10 pages)
The nonapeptide bradykinin has been shown to exhibit an array of biological activities including relaxation/contraction of various smooth muscles. In order to investigate the effects of bradykinin on the contractility and the electrical activity of antral circular muscle of guinea-pig stomach, the isometric contraction and membrane potential were recorded. Also, using standard patch clamp technique, the Ca2+-activated K currents were recorded to observe the change in cytosolic Ca2+... -
Phorbol Ester-Induced Periodic Contraction in Isolated Rabbit Jugular Vein
8 Pages
4 225-232 (8 pages)
The present study was conducted to evaluate the effect of phorbol 12,13-dibutyrate (PDBu) on the contraction of rabbit jugular vein in vitro. PDBu concentrations of greater than 10 nM induced a periodic contraction which was composed of rapid contraction, plateau and slow relaxation. The frequency of periodic contraction increased as PDBu concentration increased. The PDBu-induced contraction was inhibited by staurosporine (100 nM), it was not changed by tetrodotoxin (1 μM). In Ca2+-free... -
Effects of High Glucose on Na,K-ATPase and Na/glucose Cotransporter Activity in Primary Rabbit Kidney Proximal Tubule Cells
12 Pages
7 69-80 (12 pages)
Renal proximal tubular hypertrophy and hyperfunction are known to be early manifestations of experimental and human diabetes. As the hypertrophy and hyperfunction have been suggested to be central components in the progression to renal failure, an understanding of their underlying causes is potentially important for the development of therapy. A primary rabbit kidney proximal tubule cell culture system was utilized to evaluate the possibility that the renal proximal tubular hypertrophy and... -
Effects of cGMP on the Contractility and Ca Movement in the Aorta of Normotensive Wistar-Kyoto Rats and Spontaneously Hypertensive Rats
Park, Hae-Kun, Jeon, Byeong-Hwa, Kim, Se-Hoon, Kim, Hoe-Suk, Chang, Seok-Jong 대한생리학회 대한생리학회지 10 Pages
8 181-190 (10 pages)
Endothelium-derived relaxing factor (EDRF) activates guanylate cyclase which mediates the formation of cGMP from GTP in vascular smooth muscle. It is well known that endothelium-dependent relaxation is impaired in spontaneously hypertensive rats (SHR). However, it is still unknown whether the impaired endothelium-dependent relaxation in SHR results from the reduced release of EDRF or from the decrease of vascular response to EDRF. We investigated the effects of cGMP on the contractility and Ca... -
Effects of Lemakalim, a Potassium Channel Opener, on the Contractility and Electrical Activity of the Antral Circular Muscle in Guinea-Pig Stomach

Synthetic potassium channel openers (KCOs) are agents capable of opening K-channels in excitable cells. These agents are known to have their maximal potency in the smooth muscle tissue, especially in the vascular smooth muscle. Much attention has been focused on the type of K-channel that is responsible for mediating the effects of KCOs. As the KCO-induced changes are antagonized by glibenclamide, an KATP (ATP-sensitive K-channel) blocker in the pancreatic β-cell, KATP was suggested to be the...
