The prostate gland contains numerous neuroendocrine cells that are believed to influence the function of the prostate gland. Our recent study demonstrated the expression of both α1- and α2-ARs, signaling the release of stored Ca2 and the inhibition of N-type Ca2 channels, respectively, in rat prostate neuroendocrine cells (RPNECs). In this study, the effects of NA on the resting membrane potential (RMP) of RPNECs were investigated using a whole-cell patch clamp method. Fresh... -
Thiol-dependent Redox Mechanisms in the Modification of ATP-Sensitive Potassium Channels in Rabbit Ventricular Myocytes

Cellular redox state is known to be perturbed during ischemia and that Ca2 and K channels have been shown to have functional thiol groups. In this study, the properties of thiol redox modulation of the ATP-sensitive K (KATP) channel were examined in rabbit ventricular myocytes. Rabbit ventricular myocytes were isolated using a Langendorff column for coronary perfusion and collagenase. Single-channel currents were measured in excised membrane patch configuration of... -

or chemotherapy drug. We observed that the synthesized peptide (HNI-38) prevented DNA-PKcs from binding to Ku70/Ku80, thus resulting in inactivation of DNA-PK activity. Consequently, the peptide treated cells exhibited poor to no DNA repair, and became highly sensitive to IR or chemotherapy drugs, and the growth of breast cancer cells was inhibited. Additionally, the results obtained in the present study also support the physiological role of resistance of cancer cells to IR or chemotherapy. -
Altered Delayed Rectifier K+ Current of Rabbit Coronary Arterial Myocytes in Isoproterenol-Induced Hypertrophy

and hypertrophied hearts. These channels were measured from rabbit coronary smooth muscle cells using a patch clamp technique. The main findings of the study are as follows: (1) the KDR current density was decreased without changes of the channel kinetics in isoproterenol-induced hypertrophy; (2) the sensitivity of coronary KDR channels to 4-AP was increased in isoproterenol-induced hypertrophy. From the above results, we suggest for the first time that the alteration of KDR channels may limit... -
Effects of Prostaglandin F2α on Membrane Potentials and K+ Currents in Rabbit Middle Cerebral Arterial Cells

potentials and increased outward whole-cell K currents. PGF2α increased open-state probability of KCa channels without the change of the open and closed kinetics. PGF2α increased the amplitudes of KV currents with a leftward shift of activation and inactivation curves and a decrease of activation time constant. Our results suggest that the activation of KCa and KV channels, at least in part, may lead to attenuate or counteract vasoconstriction by PGF2α in middle cerebral artery. -
Opening of ATP-sensitive K+ Channel by Pinacidil Requires Serine/ Threonine Phosphorylation in Rat Ventricular Myocytes

The influences of specific protein phosphatase and protein kinase inhibitors on the ATP-sensitive K (KATP) channel-opening effect of pinacidil were investigated in single rat ventricular myocytes using patch clamp technique. In cell-attached patches, pinacidil (100 μM) induced the opening of the KATP channel, which was blocked by the pretreatment with H-7 (100 μM) whereas enhanced by the pretreatment with genistein (30 μM) or tyrphostin A23 (10 μM). In inside-out patches, pinacidil... -
Effects of Dexamethasone and DHEA on the Changes of Glutamate and Polyamine Uptake in Rat Astrocytes by Lipopolysaccharide and Antimycin A

antimycin A decreased Vmax, but both drugs had little effect on Km. Dexamethasone also decreased basal Vmax without any significant effect on Km. And dexamethasone further decreased the antimycin A-induced decrease of Vmax. DHEA did not affect the kinetics of basal glutamate uptake and the change by LPS or antimycin A. 2. [14C]Putrescine uptake: LPS increased Vmax, and antimycin A decreased Vmax. They showed little effect on Km. Dexamethasone decreased Vmax of basal uptake and further decreased... -
External pH Effects on Delayed Rectifier K+ Currents of Small Dorsal Root Ganglion Neuron of Rat

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.2 4 165-172 (8 pages)
Under certain pathophysiological conditions, such as inflammation and ischemia, the concentration of H+ ion in the tissue surrounding neurons is changed. Variations in H+ concentration are known to alter the conduction and/of the gating properties of several types of ion channels. Several types of K+ channels are modulated by pH. In this study, the whole cell configuration of the patch clamp technique has been applied to the recording of the responses of change of external pH on the delayed... -
Distributional Patterns of Phospholipase C Isozymes in Heart and Brain of Spontaneously Hypertensive and Normotensive Rats

The phospholipase C (PLC)-mediated intracellular signal transduction pathway is considered to be involved in the regulation of blood pressure. However, little information is available concerning the distributional and functional significance of PLC in the genetic hypertensive rats. As the first step of knowing the role of PLC on hypertension, we investigated the distribution of 6 PLC isozymes (PLC-β1, -β3, -β4, -γ1, -γ2 and -δ1) in the heart and brain, which are concerned with...
