Differential Functional Expression of Clotrimazole-sensitive Ca2+-activated K+ Current in Bal-17 and WEHI-231 Murine B Lymphocytes
HaifengZheng, JaeHongKo, JooHyunNam, YungEEarm, SungJoonKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2006, Vol.10 No.1 4 19-24 (6 pages)
The intermediate conductance Ca2-activated K channels (SK4, IKCa1) are present in lymphocytes, and their membrane expression is upregulated by various immunological stimuli. In this study, the activity of SK4 was compared between Bal-17 and WEHI-231 cell lines which represent mature and immature stages of murine B lymphocytes, respectively. The whole-cell patch clamp with high-Ca2 (0.8μM) KCl pipette solution revealed a voltage-independent K current that was blocked... -
Gene Transfer of Cu/ZnSOD to Cerebral Vessels Prevents Subarachnoid Hemorrhage-induced Cerebral Vasospasm

The preventive effects of gene transfer of human copper/zinc superoxide dismutase (Cu/ZnSOD) on the development of cerebral vasospasm after subarachnoid hemorrhage (SAH) were examined using a rat model of SAH. An experimental SAH was produced by injecting autologous arterial blood twice into the cisterna magna, and the changes in the diameter of the middle cerebral artery (MCA) were measured. Rats subjected to SAH exhibited a decreased diameter with an increased wall thickness of MCA that were... -
Inhibition of Nitric Oxide-induced Neuronal Apoptosis in PC12 Cells by Epigallocatechin Gallate

In the central nervous system, nitric oxide (NO) is associated with many pathological diseases such as brain ischemia, neurodegeneration and inflammation. The epigallocatechin gallate (EGCG), a major compound of green tea, is recognized as protective substance against neuronal diseases. This study is aimed to investigate the effect of EGCG on NO-induced cell death in PC12 cells. Administration of sodium nitroprusside (SNP), a NO donor, decreased cell viability in a dose- and time-dependent... -
Alteration of Immunoreactivity for SNARE Proteins in the Rat Hippocampus after Middle Cerebral Artery Occlusion

Soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNARE) proteins, composed of two presynaptic membrane proteins [synaptosomal-associated protein of 25 kDa (SNAP-25) and syntaxin] and a presynaptic vesicular protein [vesicle-associated membrane protein (VAMP)], serve as a core of exocytotic fusion machinery, which can be affected by ischemia. Synaptic protein in core region, striatum and cortex has been shown to alter after focal ischemia, however, little is known in... -
Changes in Phosphate Transporter Activity Evaluated by Phosphonoformic Acid Binding in Cadmium-Exposed Renal Brush-Border Membranes

in renal cortical tissues of Cd-exposed animals at the time of onset of renal dysfunction. The rate of sodium-dependent phosphate transport (Na-Pi cotransport) by BBMV was determined by 32P-labeled inorganic phosphate uptake, and the number of Na- Pi cotransporters in the BBM was assessed by Pi-protectable 14C-labeled phosphonoformic acid ([14C]PFA) binding. The exposure of BBMV to Cd decreased the Na-Pi cotransport activity in proportion to the Cd concentration in the... -
The Effects of Intracellular Monocarboxylates on the ATP-sensitive Potassium Channels in Rabbit Ventricular Myocytes

A regulating mechanism of the ATP-sensitive potassium channels (KATP channels) is yet to fully explained. This study was carried out to investigate the effects of intracellular application of monocarboxylates (acetate, formate, lactate, and pyruvate) on KATP channels in isolated rabbit ventricular myocytes. Single channel currents of KATP channels were recorded using the excised inside-out or permeabilized attached (open-cell) patch-clamp technique at room temperature. Intracellular application... -
Effect of Bromocriptine on 6-Hydroxydopamine-induced Lipid Peroxidation and Cytotoxicity in vitro and in vivo

The present study was to evaluate the protective effects of bromocriptine, which is known as D2 dopamine receptor agonist and used for the treatment of patients with Parkinson's disease (PD), on 6-hydroxydopamine (6-OHDA)-induced neurotoxicity in vitro and in vivo. Lipid peroxidation product (malondialdehyde; MDA) produced by the administration of 6-OHDA was profoundly reduced following the treatment of bromocriptine in a dose-dependent manner in rabbit brain homogenate. Quinone formation by... -
Inhibitory Effect of Caffeine on Carbachol-Induced Nonselective Cationic Current in Guinea-Pig Gastric Myocytes

In gastrointestinal smooth muscle, muscarinic stimulation by carbachol (CCh) activates nonselective cation channel current (ICCh) which is facilitated by intracellular [Ca2] increase. Caffeine is widely used in experiments to mobilize Ca2 from intracellular stores. This study shows a strong inhibitory effect of caffeine on ICCh in guinea-pig gastric myocyte. In this study, the underlying mechanism of the inhibitory effect of caffeine was investigated. ICCh was completely... -

Thiobenzamide S-oxidation in Perfused Rat Liver Ex Vivo Determination of Hepatic Flavin-Containing Monooxygenase Activity
Woon-GyeChung, Hyung-KeunRoh, Young-NamCha 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 5 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1997, Vol.1 No.5 15 591-595 (5 pages)
An ex vivo assay determining the flavin-containing monooxygenase (FMO) activity in perfused rat liver has been developed by assessing the rate of thiobenzamide S-oxide (TBSO) formation from the infused thiobenzamide (TB). The hepatotoxicity by TB or TBSO was not a critical factor for maintaining the FMO activity for up to 50 min. The FMO activity expressed in nmoles TBSO produced/g liver/min was the same for the recycling and non-recycling perfusion. This implies that reduction of the oxidized...
