Comparison of an enzyme-linked immunosorbent assay with serum neutralization test for serodiagnosis of porcine epidemic diarrhea virus infection
Jin Sik Oh, Dae Sub Song, Jeong Sun Yang, Ju Young Song, Hyoung Joon Moon, Tae Yung Kim, Bong Kyun 대한수의학회 Journal of Veterinary Science 4 Pages
대한수의학회 Journal of Veterinary Science 2005, 제 6권 제 4호 14 349-352 (4 pages)
An indirect porcine epidemic diarrhea (PED)virus (PEDV)enzyme-linked immunosorbent assay (ELISA) was compared with the serum neutralization (SN)test by testing 46 samples from experimentally infected sows,73 samples from naive sows, and 1,024 field sow samples from 48 commercial swine farms of undefined PED status. The SN test and the ELISA were performed using PEDV, KPEDV-9 strain.Viral proteins as a coating antigen of PEDV ELISA were extracted from the cytoplasm of PEDV-infected Vero cells... -
Identification of a putative cellular receptor 150 kDa polypeptide for porcine epidemic diarrhea virus in porcine enterocytes
Jin Sik Oh, Dae Sub Song, Bong Kyun Park 대한수의학회 Journal of Veterinary Science 7 Pages
대한수의학회 Journal of Veterinary Science 2003, 제 4권 제 3호 12 269-275 (7 pages)
Porcine epidemic diarrhea virus (PEDV) causes an acute enteritis in pigs of all ages, often fatality for neonates. PEDV occupies an intermediate position between two well characterized members of the coronavirus group I, human coronavirus (HCoV-229E) and transmissible gastroenteritis virus (TGEV) which uses aminopeptidase N (APN), a 150 kDa protein, as their receptors. However, the receptor of the PEDV has not been identified yet. A virus overlay protein binding assay (VOPBA) was used to... -
Seroprevalence of Antibody to Procine Reproductive and Respiratory Syndrome Virus in Diagnostic Submissions

of the PRRSV antibody was 52.1% (1765/3391). Most of the pigs seemed to be infected with PRRSV at around 50 to 60 days old. The seroprevalence of the antibody became higher with age, and peaked at around 100 days old. More than one-third of the adult pigs, including boars, gilts, and sows, was positive for the PRRSV antibody. The infection of PRRSV was chronic and confined to growers and/or finishers in most farms. However, the antibody was detected in all production phases at some farms. -
Localization of Porcine Reproductive and Respiratory Syndrome Virus Infection in Boars by In Situ Riboprobe Hybridization
Jin-Ho Shin and Thomas W. Molitor 대한수의학회 Journal of Veterinary Science 10 Pages
대한수의학회 Journal of Veterinary Science 2002, 제 3권 제 2호 5 87-96 (10 pages)
(PRRSV) to be shed in semen for extended periods of time has been suggested to be a principal factor for viral transmission via insemination. In attempts to gain insights into the mechanism of PRRSV persistence in boars, tissue distribution and sites of viral infection were investigated by in situ hybridization (ISH) using digoxigenin-labeled RNA probe and the ISH results were compared with those of reverse transcription-nested polymerase chain reaction (RT-nested PCR). Animals were intranasally... -
Assessment of Porcine Reproductive and Respiratory Syndrome Virus RNA Load in Sera and Tissues during Acute Infection
Jin-Ho Shin and Thomas W. Molitor 대한수의학회 Journal of Veterinary Science 12 Pages
대한수의학회 Journal of Veterinary Science 2002, 제 3권 제 2호 4 75-86 (12 pages)
days 5 to 14 PI showed that the viral RNA was cleared at the rate of 0.37 log reduction in the number of PRRSV RNA copies per day. It was estimated to be day 27 PI when the viral RNA in the serum of infected pigs becomes undetectable. When correlation analysis was performed between the systemic clearance rate and viral RNA concentrations in tissues of 9 infected pigs obtained at day 14 PI, moderately strong negative correlation was observed in the thymus (r = -0.62) and brain stem (r = -0.48),... -
Assessment of Replication and Virulence of Attenuated Pseudorabies Virus in Swine
T. J. Newby, D. P. Carter, K.-J. Yoon, M. W. Jackwood and P. A. Hawkins 대한수의학회 Journal of Veterinary Science 6 Pages
대한수의학회 Journal of Veterinary Science 2002, 제 3권 제 2호 1 61-66 (6 pages)
A nonclinical study was conducted to characterize the replication behavior of a modified live gE-deleted pseudorabies virus (PRV MS) in swine and potential for reversion to virulence after animal passages. Two to 3 week-old weaned pigs, negative for PRV, were maintained in isolation and challenged by intranasal instillation. For the first passage, 6 pigs were given 1 mL of PRV MS (10(7.3)TCID(50)/mL) and 2 were necropsied at 3, 4 and 5 days post-inoculation (PI). Brain and secondary lymphoid... -
Genetic characterization of porcine circovirus - 2 field isolates from PMWS pigs
Jin H. Kim, Young S. Lyoo 대한수의학회 Journal of Veterinary Science 9 Pages
대한수의학회 Journal of Veterinary Science 2002, 제 3권 제 1호 6 31-39 (9 pages)
PMWS is a new emerging disease in swine herds worldwide. Field isolates of PCV-2, a putative major causative agent of PMWS, were isolated and genetically characterized. Viral genome of two field isolates (PC201DJ and PC201SS) from pigs showing typical PMWS was sequenced. The nucleotide sequence homology with other PCV-2 isolates was ranging from 95% to 99% in complete viral genomic sequence. The highly conserved nonanucleotide motif of replication origin was identical to that of other PCV-2... -
Expression of Open Reading Frame 5 Protein of Porcine Reproductive and Respiratory Syndrome Virus Using Semliki Forest Virus Expression System

primary candidate for the PRRSV subunit vaccine. To produce the native form of GP5 in mammalian cells, we have cloned the ORF5 gene from PRRSV CNV-1 into the Semliki Forest virus (SFV)-based expression vector, resulting in recombinant pSFV-ORF5. By the infection with recombinant pSFV-ORF5 to BHK-21 cells, the GP5 expression was confirmed by immunocytochemistry and immunoblotting assay. The recombinant virus particle harboring ORF5 gene was infectious to BHK-21 and MARC-145. The RNA synthesis and... -
Acute porcine reproductive and respiratory syndrome outbreaks in immunized sow herds: from occurrence to stabilization under whole herd vaccination strategy

a type 2 PRRSV modified live vaccine (MLV). Therefore, to control the type 1 PRRSV, an alternative vaccination program utilizing both type 1 and type 2 MLV was undertaken. Following whole herd vaccinations with both types of MLV, successful stabilization of PRRS outbreaks was identified based on serological data (no viremia and downward trends in ELISA antibody titers in both sows and suckling piglets) and recovery of reproductive performance. Additionally, through comparison of the reproductive...
