single dose of 40 mg/kg body weight of moxifloxacin. Blood plasma was collected at 5, 10, 20, 30, 60, 120, 240, 480, 1440 min after drug injection. A PBPK model was developed in rats and extrapolated to human using GastroPlus software. The predictions were assessed by comparing predictions and observations. In the plasma concentration versus time profile of moxifloxcinin rats, Cmax was 11.151 μg/mL at 5 min after the intravenous injection and t1/2 was 2.936 h. Plasma concentration and kinetics... -
Activation of Lysophosphatidic Acid Receptor Is Coupled to Enhancement of Ca2+-Activated Potassium Channel Currents

G protein- coupled LPA receptor subtypes. The activation of LPA receptors induces transient elevation of intracellular Ca2+ levels through diverse G proteins such as GՁq/11, GՁi, GՁ12/13, and GՁs and the related signal transduction pathway. In the present study, we examined LPA effects on BKCa channel activity expressed in Xenopus oocytes, which are known to endogenously express the LPA receptor. Treatment with LPA induced a large outward current in a reversible and... -
Acute Toxicity and General Pharmacological Action of QGC EXT

it. We examined acute toxicity and the general pharmacological action of QGC EXT to search for any side effects of it in rats, mice, guinea pigs, and cats. In a single dose toxicity study, QGC EXT didn't show toxicological effects in rats and mice, and the LD50 was over 5 g/kg in both animals, and there were also no changes in weight, feed and water intake during these toxicological experimental periods. We examined the general pharmacological action on central controlled behavior responses, and... -
Changes of Cytosolic Ca2+ under Metabolic Inhibition in Isolated Rat Ventricular Myocytes

by 1μM thapsigargin plus 10μM ryanodine, the initial increase of Fluo-4 AM fluorescence intensity was unaffected, however, the subsequent progressive increase was abolished. KB-R7943 delayed both the first and the second phases of cytosolic Ca2 overload, while Na-free solution accelerated the second. The above results suggest that: 1) the initial rise in cytosolic Ca2 under DNP results from mitochondrial depolarization; 2) the secondary increase is caused by progressive... -
Responses of Inferior Olive Neurons to Stimulation of Semicircular Canals. II. Vertical Semicircular Canalss

about 45 degree counterclockwise to the longitudinal axis of the animal, and that those inputs were distributed to RSC in the rostral part of IOβ. On the other hand, those from the posterior semicircular canal were related with the NRP at about 45 degree clockwise and with the caudal part of the IOβ. These results suggest that IO neurons receive and encode the vestibular information, the priority of which seems to be the vertical component of the body movement rather than the horizontal ones. -
Effects of Noradrenaline on the Membrane Potential of Prostatic Neuroendocrine Cells of Rat

of cytosolic Ca2 concentration ([Ca2]c), measured by the fura-2 fluorescence, and the voltage clamp study showed the presence of charybdotoxin-sensitive Ca2-activated K currents. In summary, adrenergic stimulation induced either depolarization or hyperpolarization of RPNECs, depending on the initial level of RMP. The inward current evoked by NA and the Ca2-activated K current might partly explain the depolarization and hyperpolarization, respectively. -
Dietary Ascorbate Supplementation Reduces Oxidative Tissue Damage and Expression of iNOS in the Kidney of Streptozotocin Induced Diabetic Rats

were significantly increased in STZ-treated rats that were decreased by ascorbate supplimentation. The ascorbate supplimentation had no effect on vasorelaxation of isolated thoracic aorta. These results indicate that ascorbate supplimentation may exert an inhibitory effect on STZ-induced oxidative tissue damage through protection of pancreatic islet cells by scavanging reactive oxygen species. The ascorbate supplimentation may possibly attenuate the renal complication of diabetes mellitus. -
Role of Phospholipase A2 in Oxidant-induced Alteration in Phosphate Transport in Primary Cultured Rabbit Renal Proximal Tubule Cells

it did not alter the inhibition of phosphate uptake and the decrease in cell viability induced by tBHP. tBHP-induced inhibition of phosphate transport was not affected by a PKC inhibitor, staurosporine. tBHP at 0.1 mM did not produce the inhibition of Na-K-ATPase activity in microsomal fraction, although it significantly inhibited at 1.0 mM. These results suggest that tBHP can inhibit phosphate uptake through a mechanism independent of PLA2 activation, irreversible cell injury, and... -
The Staging of Major Mood Disorders

gaining momentum and the purpose of this review is to examine whether major mood disorders can be defined according to stages. Methods_In April 2018 the PubMed electronic data base was scrutinized by a combination of various search terms like “major depressive disorder and staging,” “bipolar disorder and neuroprogression,” etc. To incorporate the latest findings the search was limited to the last 10 years. Both original and review articles were examined by reading the abstracts, and papers... -
Mode of Inhibitory Action of Amitriptyline on Carbachol-Induced Contraction of Isolated Rabbit Detrusor Muscle

free medium, the contraction response to carbachol was markedly decreased, which was resumed by the addition of 2 (2.5mM), but not in the presence of either amitriptyline or nifedipine. KCI (60 mM) produced a potent contraction, which was abolished in the presence of amitriptyline or nifedipine. These results suggest that amitriptyline, unlike atropine, not only acts as a noncompetitive antagonist at cholinergic muscarine receptors but also inhibits Ca-influx through the muscle cell membrane.
