Evidence for Adenosine Triphosphate (ATP) as an Excitatory Neurotransmitter in Guinea-Pig Gastric Antrum

We explore the question of whether adenosine 5'-triphosphate (ATP) acts as an excitatory neurotransmitter in guinea-pig gastric smooth muscle. In an organ bath system, isometric force of the circular smooth muscle of guinea-pig gastric antrum was measured in the presence of atropine and guanethidine. Under electrical field stimulation (EFS) at high frequencies (>20 Hz), NO-mediated relaxation during EFS was followed by a strong contraction after the cessation of EFS (a... -
Effects of Dexamethasone and DHEA on the Responses of Rat Cerebral Cortical Astrocytes to Lipopolysaccharide and Antimycin A

As part of a study on the effects of dexamethasone and dehydroepiandrosterone (DHEA) on the biological roles of astrocytes in brain injury, this study evaluated the effects of dexamethasone and DHEA on the responses of primary cultured rat cortical astrocytes to lipopolysaccharide (LPS) and antimycin A. Dexamethasone decreased spontaneous release of LDH from astrocytes, and the dexamethasone effect was inhibited by DHEA. However, the inhibitory effect of DHEA on the dexamethasone-induced... -

or by depleting Ca2 from SR with CPA. They persisted in the presence of Ca2-sensitive Cl-channel blockers, niflumic acid and DIDS or Co2, a non selective Ca2-channel blocker. These results suggest that spontaneous activity of gastric smooth muscle results from Ca2 release from SR, followed by activation of Ca2-dependent ion channels other than Cl channels, with the release of Ca2 from SR being triggered by membrane depolarization. -
The Effects of Sera from Amyotrophic Lateral Sclerosis Patients on Neuromuscular Transmission and Calcium Channels in Mice

potential of ICa nor in the I-V curve. From these results it was concluded that: 1) The serum factors of sporadic ALS patients increase neuromuscular transmission and can alter motor nerve terminal presynaptic function. This suggests that ALS serum factors may play an important role in the early stage of ALS, and 2) Calcium currents in DRG cells were reduced and rapidly inactivated by ALS sera, suggesting that in these cells, ALS serum factors may exert interaction with the calcium channel. -
Regulation of Magnesium Release by cAMP during Chemical Hypoxia in the Rat Heart and Isolated Ventricular Myocytes

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.1 8 59-68 (10 pages)
Chemically induced hypoxia has been shown to induce a depletion of ATP. Since intracellular free Mg2 ([Mg2]i) appears to be tightly regulated following cellular energy depletion, we hypothesized that the increase in [Mg2]i would result in Mg2 extrusion following hormonal stimulation. To determine the relation between Mg2 efflux and cellular energy state in a hypoxic rat heart and isolated myocytes, [Mg2]i, ATP and Mg2 content were measured by... -
Roles of Na+-Ca2+ Exchange in the Negative Force-Frequency Relationship

Frequency-force relationships (FFR) were studied in electrically field stimulated rat left atria (LA) by reducing the stimulation frequency from resting 3 Hz to test frequencies (0.1-1 Hz) for 5 minutes. The twitch amplitudes of LA elicited the typical negative staircases with 3-phased changes: the initial rapid increase, the second decrease and the following plateau at test frequencies. Verapamil (3⁓105 M) pretreatment elicited frequency-dependent suppression of the twitch... -
Effects of Exogenous ATP on Calcium Mobilization and Cell Proliferation in C6 Glioma Cell

To clarify the effect of extracellular ATP in cultured C6 glioma cells, ATP-induced cytosolic free calcium ([Ca2]i) mobilization and cell proliferation were investigated. ATP-induced [Ca2]i increased in a dose-dependent manner (107 M∼103 M). ATP-induced [Ca2]i increases were slightly slowed in extracellular calcium-free conditions especially in sustained phase. ATP-induced [Ca2]i increment was also inhibited by the pretreatment of U73122, a... -

on benzodiazepinergic receptor and the GABA receptor-chloride ion channel complex. Moreover, it is known that barbiturates also have some cardiovascular regulatory effects mediated by the central GABAergic system. Therefore, this study was undertaken to delineate the regulatory actions and interactions of these systems by measuring the responses of the cardiovascular system and renal nerve activity to muscimol, diazepam and pentobarbital, administered intracerebroventricularly in rabbits. When... -

To investigate the properties of ryanodine binding sites of the bird skeletal SR vesicles, SDS PAGE, purification of RyR, and [3H]ryanodine binding study were carried out in the SR vesicles prepared from the chicken pectoral muscle. The chicken SR vesicles have two high molecular weight (HMW) protein bands as in eel SR vesicles on SDS PAGE. The HMW bands on SDS PAGE were found in the [3H] ryanodine peak fraction (Fr3-5) obtained from the purification step of the ryanodine receptor protein. Bmax...
