Seong-ChunKwon, Ki-YoungPark, Duck-SunAhn, Young-HoLee, Bok-SoonKang 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.1 5 33-40 (8 pages)
This study was designed to clarify the mechanism of the inhibitory action of a nitric oxide (NO) donor, 3-morpholino-sydnonimine (SIN-1), on contraction, cytosolic Ca2 level ([Ca2]i), and ionic currents in guinea-pig ileum. SIN-1 (0.01∼100μM) inhibited 25 mM KCl- or histamine (10μM)-induced contraction in a concentration-dependent manner. SIN-1 reduced both the 25 mM KCl- and the histamine-stimulated increases in muscle tension in parallel with decreased [Ca2]i. Using the... -

Yoon-YoungJang, Eun-SookHan, Chung-SooLee, Young-KiKim, Jin-HoSong, Yong-KyooShin 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 10 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.6 12 631-640 (10 pages)
The present study investigated the stimulatory effects of iron (or ascorbate) on cyclosporine-induced kidney mitochondrial damage. Damaging effect of 50 μM cyclosporine plus 20 μM Fe2 on mitochondrial lipids and proteins of rat kidney and hyaluronic acid was greater than the summation of oxidizing action of each compound alone, except sulfhydryl oxidation. Cyclosporine and 100 μM ascorbate showed an enhanced damaging effect on lipids but not on proteins. The peroxidative action of... -
Role of K+ Channels to Resting Membrane Potential of Rabbit Middle Cerebral Arterial Smooth Muscle Cells

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.6 2 547-554 (8 pages)
The aim of the present study is to investigate the contribution of Ca2-activated K (KCa) channels and delayed rectifier K (KV) channels to the resting membrane potential (RMP) in rabbit middle cerebral arterial smooth muscle cells. The RMP and membrane currents were recorded using the whole-cell patch configuration and single KCa channel was recorded using the outside-out patch configuration. Using the pipette solution containing 0.05 mM EGTA, the RMP was... -

Glutamate and γ-aminobutyric acid (GABA) are major excitatory and inhibitory neurotransmitters in the vertebrate retina, respectively. Using the whole-cell patch clamp technique and a rapid solution changer, glutamate and GABA receptors have been extensively investigated in carp retina. Glutamate receptors on both horizontal and amacrine cells may be an AMPA preferring subtype, which predominantly consists of flop splice variants. GABAA and GABAC receptors coexist in bipolar cells and they both... -
Effect of Imipramine on Calcium Utilization of Single Cells Isolated from Canine Detrusor

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.4 9 439-445 (7 pages)
This study is to investigate the mechanism of inhibitory effect of imipramine on the calcium utilization in single cells isolated from canine detrusor. 2 mm thick smooth muscle chops were incubated in 0.12% collagenase solution at 36oC, and aerated with 95% O2/5% CO2, and then cell suspension was examined. Acetylcholine (ACh) evoked a concentration-dependent contraction of the isolated detrusor cells in normal physiologic salt solution (PSS), and the ACh-induced contraction was significantly... -
Effects of Acarbose on the Expression of Obese and Neuropeptide Y (NPY) Genes in Mice on High-Carbohydrate Diet

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.4 8 433-438 (6 pages)
Two components of the neuroendocrine-hormonal response to long-term treatment of acarbose, adipose tissue-derived leptin and central neuropeptide Y (NPY), were investigated in the ICR mice on a high- carbohydrate diet. Acarbose, administered 5 or 50 mg per 100 g diet for four weeks, dose dependently suppressed body weight gain. The body weight gain was reduced along with the amount of daily food intake in 50 mg acarbose-treated group at 7th and 28th day. 5 or 50 mg acarbose treatment... -

[K]o can be increased under a variety of conditions including subarachnoid hemorrhage. The increase of [K]o in the range of 5∼15 mM may affect tensions of blood vessels and cause relaxation of agonist-induced precontracted vascular smooth muscle (K-induced relaxation). In this study, effect of the increase in extracellular K concentration on the agonist-induced contractions of various arteries including resistant arteries of rabbit was examined, using home-made... -

We investigated the role of Ca2 and protein kinases/phosphatases in the stimulatory effect of insulin on glucose transport. In isolated rat adipocytes, the simple omission of CaCl2 from the incubation medium significantly reduced, but did not abolish, insulin-stimulated 2-deoxy glucose (2-DG) uptake. Pre-loading adipocytes with intracellular Ca2 chelator, 5,5'-dimethyl bis (o-aminophenoxy)ethane-N,N,N'N' tetraacetic acetoxymethyl ester (5,5'-dimethyl BAPTA/AM) completely blocked... -

IC50 of 0.24 and 927μM, respectively. The inhibitory action of DIDS was irreversible whereas that of niflumic acid was reversible. Furthermore, DIDS-induced block was not recovered despite exposure to ATP (1 mM). In cell-attached and inside-out patches, DIDS blocked the pinacidil- or 2,4-dinitrophenol (DNP)-induced KATP channel openings. In contrast, niflumic acid did not block the pinacidil-induced KATP channel openings in inside-out patches, but inhibited it in cell-attached patches. DIDS and... -

partial rundown state. The effect of pinacidil (10 μM) was not affected by the pretreatment with protein tyrosine phosphatase 1B (PTP1B, 10 μg ml1), but blocked by the pretreatment of protein phosphatase 2A (PP2A, 1 U ml1). In addition, pinacidil (10 μM) could not induce the opening of the reactivated KATP channels in the presence of H-7 (100 μM) but enhanced it in the presence of ATP (1 mM) and genistein (30 μM). These results indicate that the KATP channel-opening effect of...
