The ryanodine receptor, a Ca2 release channel of the sarcoplasmic reticulum (SR), is responsible for the rapid release of Ca2 that activates cardiac muscle contraction. In the excitation-contraction coupling cascade, activation of SR Ca2 release channel is initiated by the activity of sarcolemmal Ca2 channels, the dihydropyridine receptors. Previous study showed that the relaxation defect of diabetic heart was due to the changes of the expressional levels of SR... -

TFA enhanced channel activity in a concentration-dependent fashion. The concentration of TFA for half-maximal activation and the Hill coefficient were 0.03 mM and 1.2, respectively. TFA did not affect the single channel conductance of KATP channels. Analysis of open and closed time distributions showed that TFA increased burst duration and decreased the interburst interval without changes in open and closed time distributions shorter than 5 ms. TFA diminished ATP sensitivity of KATP channels in ... -

site of carrier-mediated transport from blood into urine. During the last decade, molecular cloning has identified several families of multispecific organic anion and cation transporters, such as organic anion transporter (OAT), organic cation transporter (OCT), and organic anion-transporting polypeptide (oatp). Additional findings also suggested ATP-dependent organic ion transporters such as MDR1/P-glycoprotein and the multidrug resistance-associated protein (MRP) as efflux pump. The substrate... -
Influence of Staurosporine on Catecholamine Release Evoked by Cholinergic Stimulation and Membrane Depolarization from the Rat Adrenal Gland

The present study was attempted to examine the effect of staurosporine (STS) on secretion of catecholamines (CA) evoked by cholinergic stimulation and membrane depolarization from the isolated perfused rat adrenal gland and to establish its mechanism of action. The perfusion of STS (3⁓107∼3⁓108 M) into an adrenal vein for 20 min produced a dose-dependent inhibition in CA secretion evoked by ACh (5.32⁓103 M), high K (5.6⁓102 M), DMPP... -
Cadmium Inhibition of Renal Endosomal Acidification

Chronic exposure to cadmium (Cd) results in an inhibition of protein endocytosis in the renal proximal tubule, leading to proteinuria. In order to gain insight into the mechanism by which Cd impairs the protein endocytosis, we investigated the effect of Cd on the acidification of renal cortical endocytotic vesicles (endosomes). The endosomal acidification was assessed by measuring the pH gradient-dependent fluorescence change, using acridine orange or FITC-dextran as a probe. In renal endosomes... -
The Enhancement of Endotoxin-Induced Nitric Oxide Production by Elevation of Glucose Concentration in Macrophage
HyunGooWoo, Yi-SookJung, EunJooBaik, Chang-HyunMoon, SooHwanLee 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages

The production of nitric oxide (NO) and the expression of inducible nitric oxide synthase (iNOS) are known to be modulated by a variety of factors. Recent study showed that endotoxin-induced NO synthesis and iNOS expression were greatly enhanced by elevation of extracellular glucose concentration in murine macrophages. Although this was suggested to be due to the activation of protein kinase C (PKC) via sorbitol pathway, there was lack of evidence for this speculation. This study was performed... -
Mechanism of Pituitary Adenylate Cyclase-Activating Polypeptide- Induced Inhibition on Catecholamine Secretion Evoked by Cholinergic Stimulation and Membrane Depolarization in the Rat Adrenal Gland
Dong-YoonLim, Jeong-WonKang, Young-JoKim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 12 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.3 12 339-350 (12 pages)
The present study was attempted to examine the effect of pituitary adenylate cyclase-activating polypeptide (PACAP) on catecholamine (CA) secretion evoked by cholinergic stimulation, membrane depolarization and calcium mobilization from the isolated perfused rat adrenal gland. The perfusion of PACAP (10 nM) into an adrenal vein for 60 min produced a great inhibition in CA secretion evoked by ACh (5.32⁓103 M), high K (5.6⁓102 M), DMPP (104 M for 2 min),... -

of theobromine (TB) and caffeine (CA) was determined. Their urine was collected for 1 hr (between 4 and 5 hrs) after they drank a cup of coffee containing 115 mg CA and analyzed by an HPLC system. The lowest TB/CA ratio obtained was 0.40, the highest ratio was 15.17 (38-fold difference), and the median ratio for all subjects was 1.87. The mean was 2.66 with 2.36 S.D.. In 134 nonsmokers, the mean ratio was 2.35⁑1.93, that of 51 males was 2.30⁑2.26 and 83 females was 2.37⁑1.85,... -
Minimal Amount of Insulin Can Reverse Diabetic Heart Function Sarcoplasmic Reticulum Ca2+ Transport and Phospholamban Protein Expression
HaeWonKim, YongSunCho, YunSongLee, EunHeeLee, HeeRanLee 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1999, Vol.3 No.2 7 175-182 (8 pages)
In the present study, the underlying mechanisms for diabetic functional derangement and insulin effect on diabetic cardiomyopathy were investigated with respect to sarcoplasmic reticulum (SR) Ca2-ATPase and phospholamban at the transcriptional and translational levels. The maximal Ca2 uptake and the affinity of Ca2-ATPase for Ca2 were decreased in streptozotocin-induced diabetic rat cardiac SR, however, even minimal amount of insulin could reverse both parameters.... -

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.4 6 443-454 (12 pages)
The present study was attempted to investigate the effect of vasoactive intestinal polypeptide (VIP) on secretion of catecholamines (CA) and to establish whether there is the existence of a noncholinergic mechanism in adrenomedullary CA secretion from the isolated perfused rat adrenal gland. The perfusion into an adrenal vein of VIP (3⁓106 M) for 5 min or the injection of acetylcholine (ACh, 5.32⁓103 M) resulted in great increases in CA secretion. Tachyphylaxis to...
