Role of Calmodulin in the Generation of Reactive Oxygen Species and Apoptosis Induced by Tamoxifen in HepG2 Human Hepatoma Cells
The Korean Journal of Physiology & Pharmacology 6 Pages
The Korean Journal of Physiology & Pharmacology 2002, Vol.6 No.4 3 187-192 (6 pages)
Tamoxifen, an antiestrogen, has previously been shown to induce apoptosis in HepG2 human hepatoblastoma cells through activation of the pathways independent of estrogen receptors, i.e., intracellular Ca2 increase and generation of reactive oxygen species (ROS). However, the mechanism of tamoxifen to link increased intracellular Ca2 to ROS generation is currently unknown. Thus, in this study we investigated the possible involvement of calmodulin, a Ca2 activated protein, and... -
Increases in Effective Cleft Glutamate Concentration During Expression of LTP
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2002, Vol.6 No.2 8 113-120 (8 pages)
cleft since AMPA-type glutamate receptors are not generally saturated during basal transmission. Here we report evidence that peak cleft glutamate concentration ([glu]cleft) increases during LTP, as indicated by a lessening of the blocking effects of rapidly unbinding antagonists of AMPA. The efficacy of slowly equilibrating antagonists remained unchanged. The elevated [glu]cleft helps support the increased quantal amplitude of AMPA-type EPSCs (excitatory postsynaptic currents) during LTP. -
Increase of Intracellular Ca2+ Concentration Induced by Lysophosphatidylcholine in Murine Aortic Endothelial Cells
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2002, Vol.6 No.2 5 93-100 (8 pages)
Effects of oxidized low-density lipoprotein (ox-LDL), l-α-stearoyl-lysophosphatidylcholine (LPC), on intracellular Ca2 concentration were examined in mouse endothelial cells by measuring intracellular Ca2 concentration ([Ca2]i) with fura 2-AM and reverse transcription-polymerase chain reaction (RT-PCR). LPC increased [Ca2]i under the condition of 1.5 mM [Ca2]o but did not show any effect under the nominally Ca2-free condition. Even after the store... -
Correlation between mEPSC Amplitude and Rise Time upon the Blockade of AMPA Receptor Desensitization at Hippocampal Synapses
The Korean Journal of Physiology & Pharmacology 6 Pages
The Korean Journal of Physiology & Pharmacology 2002, Vol.6 No.2 3 81-86 (6 pages)
Conventional views of synaptic transmission generally overlook the possibility of "postfusional- control" the regulation of the speed or completeness of transmitter release upon vesicular fusion. However, such regulation often occurs in non-neuronal cells where the dynamics of fusion-pore opening is critical for the speed of transmitter release. In case of synapses, the slower the transmitter release, the smaller the size and rate-of-rise of postsynaptic responses would be expected if... -
Noradrenergic Modulation of Spontaneous Inhibitory Postsynaptic Currents in the Hypothalamic Paraventricular Nucleus
The Korean Journal of Physiology & Pharmacology 10 Pages
The Korean Journal of Physiology & Pharmacology 2002, Vol.6 No.2 2 71-80 (10 pages)
Previous studies have suggested that brain stem noradrenergic inputs differentially modulate neurons in the paraventricular nucleus (PVN). Here, we compared the effects of norepinephrine (NE) on spontaneous GABAergic inhibitory postsynaptic currents (sIPSCs) in identified PVN neurons using slice patch technique. In 17 of 18 type I neurons, NE (30∼100μM) reversibly decreased sIPSC frequency to 41⁑7% of the baseline value (4.4⁑0.8 Hz, p<0.001). This effect was blocked by yohimbine... -
Muscarine M2 Receptor-mediated Presynaptic Inhibition of GABAergic Transmission in Rat Meynert Neurons

of the Meynert neurons using the conventional whole-cell patch recording configuration. Muscarine (10μM) reversibly and concentration-dependently decreased mIPSC frequency without affecting the current amplitude distribution. Muscarine action on GABAergic mIPSCs was completely blocked by 1μM methoctramine, a selective M2 receptor antagonist, but not by 1μM pirenzepine, a selective M1 receptor antagonist. NEM (10μM), a G-protein uncoupler, attenuated the inhibitory action of muscarine on... -
Congenital LQT Syndromes From Gene to Torsade de Pointes
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2002, Vol.6 No.1 1 1-8 (8 pages)
Congenital Long QT syndrome (LQTs) is a relatively rare pathologic disorder but results frequently in sudden cardiac death. Of the six LQTs that have been clinically described, five have been worked out for their genetic and biophysical profile. Most are generated by mutations which cause a loss of function in two delayed Kcurrents, iKs and iKr. One syndrome is generated by mutations in the Na channel which causes essentially a gain of function in the channel. Clinically the... -
Nimodipine as a Potential Pharmacological Tool for Characterizing R-Type Calcium Currents
The Korean Journal of Physiology & Pharmacology 9 Pages
The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.6 8 511-519 (9 pages)
Nimopidine, one of dihydropyridine derivatives, has been widely used to pharmacologically identify L-type Ca currents. In this study, it was tested if nimodipine is a selective blocker for L-type Ca currents in sensory neurons and heterologous system. In mouse dorsal root ganglion neurons (DRG), low concentrations of nimodipine (<10μM), mainly targeting L-type Ca currents, blocked high-voltage-activated calcium channel currents by ∼38%. Interestingly, high concentrations of nimodipine... -
Forward-Mode Na+-Ca2+ Exchange during Depolarization in the Rat Ventricular Myocytes with High EGTA

the initial outward current was converted into inward current, 47⁑1% of which was suppressed by 0.1 mM CdCl2. 10 mM caffeine increased the remaining inward current after CdCl2 in a cAMP-dependent manner. This caffeine-induced inward current was blocked by 1μM ryanodine, 10μM thapsigargin, 5 mM NiCl2, or by Na and Ca2 omission, but not by 0.1μM isoproterenol. The I∼V relationship of the caffeine-induced current elicited inward current from 45 mV to 3 mV with... -
The Inhibitory Effect of Opioid on the Hyperpolarization-Activated Cation Currents in Rat Substantia Gelatinosa Neurons
The Korean Journal of Physiology & Pharmacology 8 Pages
The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.5 2 373-380 (8 pages)
The action of opioid on the hyperpolarization-activated cation current (Ih) in substantia gelatinosa neurons were investigated by using whole-cell voltage-clamp recording in rat spinal brain slices. Hyperpolarizing voltage steps revealed slowly activating currents in a subgroup of neurons. The half-maximal activation and the reversal potential of the current were compatible to neuronal Ih. DAMGO (1μM), a selective- opioid agonist, reduced the amplitude of Ih reversibly. This reduction was...
