Effect of Cadmium on Organic Acid Transport System in Renal Basolateral Membrane
10 Pages
10 279-288 (10 pages)
in Na+ and αKG-independent transport component. Kinetic analysis indicated that Vmax, but not Km, of the Na+-dependent, αKG-driven component was reduced. A similar reduction of Na+-dependent, αKG-driven PAH uptake was observed in normal membrane vesicles directly exposed to inorganic cadmium in vitro, and this was accompanied by an inhibition of both Na+-dependent αKG uptake and αKG-PAH exchange activity. These results indicate that during chronic exposure to cadmium, free cadmium ions... -
Effect of Probenecid on Tetraethylammonium (TEA) Transport Across Basolateral Membrane of Rabbit Proximal Tubule
8 Pages
7 249-256 (8 pages)
mM probenecid. Kinetic analysis indicated that probenecid decreased Vmax without significant change in Km. Probenecid inhibited significantly tissue oxygen consumption at concentrations of 3 and 5 mM. However, probenecid did not significantly reduce TEA uptake in brush border and basolateral membrane vesicles prepared from renal cortex even at a concentration as high as 10 mM. These results indicate that probenecid reduces TEA uptake in cortical slices by inhibiting tissue metabolism rather than... -
Decreased GLUT 4 mRNA Levels did not Related with Degree of Hyperglycemia in Skeletal Muscles of Streptozotocin-induced Diabetic Rats
6 Pages
5 231-236 (6 pages)
In our previous study (Kim et al, 1991), GLUT 4 protein content correlated negatively with plasma glucose levels in skeletal muscles of STZ-induced diabetic rats. Thus, in this study, to confirm whether expression of GLUT 4 correlate negatively with degree of hyperglycemia, we measured levels of GLUT 4 mRNA in red and white gastrocnemius muscles in STZ-induced mild and severe diabetic rats. Rats were randomly assigned to control, mild, and severe diabetic groups, and the diabetes was induced by... -
The Effects of Prolactin and Vasopressin on the Regulation of Amniotic Fluid Volume and Its Na+ Concentration through the Membrane Surrounding Amniotic Fluid
10 Pages
8 81-90 (10 pages)
The effects of prolactin and vasopressin on the regulation of amniotic fluid (AF) volume and its Na+ concentration ([Na+]) through the membrane surrounding the AF during increase in AF volume due to fetal urination, were studied. About 70% of AF volume was replaced with normal isotonic saline solution. Isotonic saline solution (0.5ml) containing Congored and LiCI was introduced into each amniotic sac. Vasopressin (25 ng/ml) or prolactin (1 mg/ml) of AF was then injected into experimental... -
Contribution of Bulk Flow to Transport Mechanisms of the Membranes Surrounding Amniotic Fluid in the Rabbit
12 Pages
8 79-90 (12 pages)
The objective of the present study is to assess the contribution of bulk flow to the regulatory mechanism of amniotic fluid volume and its ionic concentration in the membranes surrounding the amniotic fluid. For quantitative assessment, we prepared 4 kinds of artificial amniotic fIuids (isotonic isovolumetric, hypotonic isovolumetric, isotonic hypervolumetric and hypotonic hypervolumetric ones) by replacing 70% of amniotic fluid of pregnant rabbits with water or normal Tyrode solutions.... -
Effect of Changing Amniotic Fluid Osmolarity on the Li+ Transport Through the Membrane Surrounding Amniotic Fluid in the Rabbit
14 Pages
2 13-26 (14 pages)
To study the regulation of amniotic fluid volume and electrolyte concentration by the Membranes surrounding the amniotic fluid, the rate of Li+ disappearance from amniotic sac of expired fetuses were examined while increasing the amniotic volume and osmolarity in rabbits. After intraamniotic injection of 1 ml isosmotic saline (about 20% of the amniotic fluid volume) containing 15 mM LiCl and 0.5 g/L Censored, the time courses of Li+ and Censored disappearance were determined. From there the Li+... -
A Study on Na+ and Water Reabsorption in the Nephron Segment Beyond Proximal Tubule Measured by Lithium Clearance

with a bolus of LiCl solution (2 mM/kg, i.v.) for conditioning the CLi and urine flow rate. To rule out the effect of Li+ on tubular functions, a bolus of NaCl solution (2 mM/kg, i.v.) was administered. Fifteen, thirty, and sixty minutes after injection of hypertonic saline arterial blood and urine samples were taken. Urinary and plasma concentrations as well as urinary output of Li+, Na+ and K+ were measured. From these CLi, CNa and the reabsorption fraction of Na and water (FrNa & FrH2O) were... -
Effect of Renal Ischemia in Tetraethylammonium Transport in Rabbit Renal Coritcal Slices
8 Pages
7 171-178 (8 pages)
This study was carried out to determine effect of acute renal ischemia on transport function of organic cation, tetraethylammonium (TEA), in rabbit kidney proximal tubule. Clamping of the renal artery for 30 and 60 min produced a polyuria which was accompanied by an increase in Na+ excretion. The capacity of kidney cortical slices to accumulate TEA was increased after 30 and 60 min of ischemia. When blood flow was restored for 30 min after 30 and 60 min of ischemia, the augmented TEA uptake was... -
Proton Transport in Human Placental Microvillous Membrane Vesicles
12 Pages
5 49-60 (12 pages)
The change of the acridine orange absorbance was used to monitor the formation and/or dissipation of a pH gradient in microvillous membrane vesicles (MVV) isolated from human term placenta. Under Na+ efflux conditions, an acidification of the intravesicular space occured and it was completely inhibited by 0.1 mM amiloride. Under K+ efflux conditions, an acidification of the intravesicular space occured and it was potentiated by valinomycin or FCCP. An inwardly directed chloride gradient also... -
Effect of Probenecid on Urate Excretion in the Cat Kidney

increased urine flow. Thus, an increase in FEur by probenecid could be considered to be resulted from a change in urine flow. In the cat infusion of probenecid (2.5 mg/kg·min) into renal artery markedly suppressed FEPAH, but the effects on FEur and urine flow were similar to those when probenecid was infused into femoral vein. These results indicate that in the cat kidney urate filtered through glomerulus is reabsorbed by a probenecid-insensitive mechanism with no evidence for net secretion.
