a detection limit of up to eight copies per reaction, which was equivalent to that of the real-time quantitative polymerase chain reaction (qPCR) assay. The assay was specific, as it did not amplify genomes from five other Mycobacterium spp. or five pathogenic enteric bacteria. Six hundred-twelve clinical samples (320 fecal and 292 serum) were assessed by RPA-LFD, qPCR, and enzyme-linked immunosorbent assay, respectively. The RPA-LFD assay yielded 100% sensitivity, 97.63% specificity, and 98.44%... -

a colloidal gold-labeled recombinant Nsp7 antigen probe was successfully developed for the detection of anti-porcine reproductive and respiratory syndrome virus (PRRSV) antibodies in swine. Recombinant Nsp7 protein of PRRSV labeled with colloidal gold was dispensed on a conjugate pad for use as the detector. Staphylococcal protein A and purified porcine anti-Nsp7 antibodies were blotted on a nitrocellulose membrane to form test and control lines, respectively. A comparison of the strip with... -
Uracil-DNA glycosylase-treated reverse transcription loop-mediated isothermal amplification for rapid detection of avian influenza virus preventing carry-over contamination
Eun-Mi Kim, Hyo-Sung Jeon, Ji-Jung Kim, Yeun-Kyung Shin, Youn-Jeong Lee, Sang-Geon Yeo, Choi-Kyu Park 대한수의학회 Journal of Veterinary Science 5 Pages
대한수의학회 Journal of Veterinary Science 2016, 제 17권 제 3호 21 421-425 (5 pages)
Here, we describe a uracil-DNA glycosylase (UNG)-treated reverse transcription loop-mediated isothermal amplification (uRT-LAMP) for the visual detection of all subtypes of avian influenza A virus (AIV). The uRT-LAMP assay can prevent unwanted amplification by carryover contamination of the previously amplified DNA, although the detection limit of the uRT-LAMP assay is 10-fold lower than that of the RT-LAMP without a UNG treatment. To the best of our knowledge, this is the first successful... -
Accuracy of capillary blood 3-β-hydroxybutyrate determination for the detection and treatment of canine diabetic ketoacidosis
Francesca Bresciani, Marco Pietra, Sara Corradini, Massimo Giunti, Federico Fracassi* 대한수의학회 Journal of Veterinary Science 8 Pages
대한수의학회 Journal of Veterinary Science 2014, 제 15권 제 2호 18 309-316 (8 pages)
In human medicine, diagnosis of diabetic ketoacidosis (DKA) is usually based on measurement of capillary 3-β-hydroxybutyrate (3-HB) with a hand held ketone sensor. This study was conducted to determine if measurement of capillary 3-HB could be useful for the diagnosis and monitoring of canine DKA. Fifteen dogs with diabetic ketosis and 10 with DKA were evaluated. Paired measurements of 3-HB of capillary and venous blood samples were analysed by the electrochemical sensor and reference method.... -
Development of a real-time SYBR Green PCR assay for the rapid detection of Dermatophilus congolensis

as real time (RT)-PCR have not been developed for the rapid detection and diagnosis of Dermatophilus(D.)congolensis infection. In the present study, a D. congolensis-specific SYBR Green RT-PCR assay was evaluated. The detection limit of the RT-PCR assay was 1 pg of DNA per PCR reaction. No cross-reaction with nucleic acids extracted from Pseudomonas aeruginosa, Mycobacterium tuberculosis, Staphylococcus aureus, or Austwickia chelonae was observed. Finally, the RT-PCR assay was used to evaluate... -

placed on the top of adsorbent pads enclosed in a plastic cassette with a test zone at the center. The FTA could be completed within 10 min. Clear purple dots against a white background indicated the presence of Aphanomyces (A.) invadans. The FTA limit of detection was 7 μg/mL for A.invadans compared to 56 μg/mL for the immunodot. FTA and polymerase chain reaction (PCR) could detect A.invadans in fish tissue homogenates at a 10 dilution compared to a 10 dilution by immunodot. In fish suffering... -
Serotype- and serogroup-specific detection of African horsesickness virus using phage displayed chicken scFvs for indirect double antibody sandwich ELISAs
Wouter van Wyngaardt*, Cordelia Mashau, Isabel Wright, Jeanni Fehrsen 대한수의학회 Journal of Veterinary Science 4 Pages
대한수의학회 Journal of Veterinary Science 2013, 제 14권 제 1호 14 95-98 (4 pages)
There is an ongoing need for standardized, easily renewable immunoreagents for detecting African horsesickness virus (AHSV). Two phage displayed single-chain variable fragment (scFv) antibodies, selected from a semi-synthetic chicken antibody library, were used to develop double antibody sandwich enzyme-linked immunosorbent assays (DAS-ELISAs) to detect AHSV. In the DAS-ELISAs, the scFv previously selected with directly immobilized AHSV-3 functioned as a serotype-specific reagent that recognized... -
Application of a multiplex PCR assay for Campylobacter fetus detection and subspecies differentiation in uncultured samples of aborted bovine fetuses
Gregorio Iraola, Mart?n Hern?ndez, Luc?a Calleros, Fernando Paolicchi, Silvia Silveyra, Alejandra Velilla, Luis Carretto, Eliana 대한수의학회 Journal of Veterinary Science 6 Pages
대한수의학회 Journal of Veterinary Science 2012, 제 13권 제 4호 6 371-376 (6 pages)
Campylobacter (C.) fetus (epsilonproteobacteria) is an important veterinary pathogen. This species is currently divided into C. fetus subspecies (subsp.) fetus (Cff) and C. fetus subsp. venerealis (Cfv). Cfv is the causative agent of bovine genital Campylobacteriosis, an infectious disease that leads to severe reproductive problems in cattle worldwide. Cff is a more general pathogen that causes reproductive problems mainly in sheep although cattle can also be affected. Here we describe a... -
Development and evaluation of a multiplex PCR assay for simultaneous detection of Flavobacterium psychrophilum, Yersinia ruckeri and Aeromonas salmonicida subsp. salmonicida in culture fisheries
Ertan Emek Onuk, Alper Ciftci*, Arzu Findik, Yuksel Durmaz 대한수의학회 Journal of Veterinary Science 7 Pages
대한수의학회 Journal of Veterinary Science 2010, 제 11권 제 3호 9 235-241 (7 pages)
In optimized conditions, neither false specific nor nonspecific amplification occurred. The detection limits of the m-PCR method using DNA extracts from dilutions of pure cultures of bacteria were 35 pg for Y. ruckeri and F. psychrophilum and 70 pg for A. salmonicida subsp. salmonicida. It was determined that 15 CFU Y. ruckeri and F. psychrophilum and 30 CFU A. salmonicida subsp. salmonicida could be detected by m-PCR developed using genomic DNA extracted from dilutions of the suspensions. The... -
Detection of Streptococcus dysgalactiae subsp. equisimilis in equine nasopharyngeal swabs by PCR
Silvia Preziuso, *, Fulvio Laus, Aurora Romero Tejeda, Carlo Valente, Vincenzo Cuteri2 대한수의학회 Journal of Veterinary Science 6 Pages
대한수의학회 Journal of Veterinary Science 2010, 제 11권 제 1호 10 67-72 (6 pages)
Streptococcus (S.) dysgalactiae subsp. equisimilis is responsible for severe diseases in humans, including primary bacteraemia, pneumonia, endocarditis, and toxic shock syndrome. Infection in some animal species can also occur, although a few studies have looked into cross-species infectivity. In horses, S. equisimilis is generally considered infrequent or opportunistic, but has recently been isolated from cases of strangles-like disease. Rapid and sensitive diagnostic techniques could enable...
