Characterization of ETB Receptor-mediated Relaxation in Precontracted Mesenteric Artery from Streptozotocin-induced Diabetic Rats
YangKiEom, KoanHoiKim, ByungYongRhim 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 9 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2005, Vol.9 No.5 8 305-313 (9 pages)
Diabetes mellitus is associated with vascular complications, including an impairment of vascular function and alterations in the reactivity of blood vessels to vasoactive substances in various vasculature. In the present study, the authors have observed endothelin-B (ETB) receptor agonist-induced relaxation in precontracted mesenteric arterial segments from streptozotocin (STZ)-induced diabetic rats, which was not shown from control rats or in other arterial segments from diabetic rats.... -
Changes of Cytosolic Ca2+ under Metabolic Inhibition in Isolated Rat Ventricular Myocytes

To characterize cytosolic Ca2 fluctuations under metabolic inhibition, rat ventricular myocytes were exposed to 200μM 2,4-dinitrophenol (DNP), and mitochondrial Ca2, mitochondrial membrane potential (ΔΨm), and cytosolic Ca2 were measured, using Rhod-2 AM, TMRE, and Fluo-4 AM fluorescent dyes, respectively, by Laser Scanning Confocal Microscopy (LSCM). Furthermore, the role of sarcolemmal Na/Ca2 exchange (NCX) in cytosolic Ca2 efflux was studied in... -
Naltrexone Inhibits Catecholamine Secretion Evoked by Nicotinic Receptor Stimulation in the Perfused Rat Adrenal Medulla

greatly CA secretion evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors as well as that by membrane depolarization. It seems that this inhibitory effect of naltrexone does not involve opioid receptors, but might be mediated by blocking both the calcium influx into the rat adrenal medullary chromaffin cells and the uptake of Ca2 into the cytoplasmic calcium store, which are at least partly relevant to the direct interaction with the nicotinic receptor itself. -
The Role of Mitochondrial ATP-sensitive Potassium Channel on Intestinal Pacemaking Activity

using whole-cell patch clamp techniques. Under current clamp mode, at 10μM glibenclamide, there was no change in pacemaking activity of ICCs. At 30μM glibenclamide, an inhibitor of the ATP sensitive K channels, we could find two examples. If pacemaking activity of ICCs was irregulating, pacemaking activity of ICCs was changed into regulating and if in normal conditions, membrane potential amplitude was increased. At 50μM glibenclamide, the resting membrane potential was depolarized. At... -
The Effect of External Divalent Cations on Intestinal Pacemaking Activity

effect of 10 mM Ba2, Mn2 and Sr2 on pacemaking activity of ICC in the presence of external 0 mM Mg2, and found that 10 mM Ba2 and Mn2 induced large inward current and stopped the pacemaking activity of ICC (n=5). Interestingly, 10 mM Sr2 induced small inward current and potentiated the amplitude of pacemaking activity of ICC (n=5). These results indicate that extracellular Ca2 and Mg2 are requisite for the pacemaking activity of ICC. -
N-methyl-D-aspartate (NMDA) and Non-NMDA Receptors are Involved in the Production and Maintenance of Nociceptive Responses by Intraplantar Injection of Bee Venom and Melittin in the Rat

have been reported to induce long-lasting spontaneous flinchings and hyperalgesia. The current study was designed to elucidate the peripheral and spinal mechanisms of N-methyl-D-aspartate (NMDA) and non-NMDA receptors by which intraplantar (i.pl.) injection of WBV and melittin induced nociceptive responses. Changes in mechanical threshold and flinching behaviors were measured after the injection of WBV (0.04 mg or 0.1 mg/paw) and melittin (0.02 mg or 0.05 mg/paw) into the mid-plantar area of a... -
Peripheral Nerve Injury Alters Excitatory and Inhibitory Synaptic Transmission in Rat Spinal Cord Substantia Gelatinosa
Dong-HoYoun 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2005, Vol.9 No.3 2 143-148 (6 pages)
Following peripheral nerve injury, excessive nociceptive inputs result in diverse physiological alterations in the spinal cord substantia gelatinosa (SG), lamina II of the dorsal horn. Here, I report the alterations of excitatory or inhibitory transmission in the SG of a rat model for neuropathic pain ("spared nerve injury"). Results from whole-cell recordings of SG neurons show that the number of distinct primary afferent fibers, identified by graded intensity of stimulation, is... -

we found Iberiotoxin- and TEA-sensitive outward rectifying oscillatory current with whole-cell recordings. Single channel analysis showed large conductance K channels (106 pS of chord conductance at 40 mV in physiological K gradient). The 106 pS channels were activated by membrane potential and [Ca2]i, consistent with the known properties of BKCa channels. BKCa channels in CRL-1474 were positively regulated by adenylate cyclase activator (10μM forskolin), 8-Br-cyclic... -

1 mmol/L, and 10 mmol/L BAPTA, respectively. In these situations, ryanodine-sensitive inward NCX current was compared using 100μmol/L ryanodine, Na depletion, 5 mmol/L NiCl2 and 1μmol/L nifedipine. Surprisingly, the result showed that the ryanodine-sensitive inward NCX current was well preserved after 10 mmol/L BAPTA to 91 % of that obtained after 1 mmol/L BAPTA. From this result, it is concluded that most of the NCX-induced Ca2 extrusion occurs before the Ca2 diffuses to... -
Ca2+-induced Ca2+ Release from Sarcoplasmic Reticulum Negatively Regulates Myocytic ANP Release in Beating Rabbit Atria
DanLi, HeXiuQuan, JinFuWen, JingYuJin, SungHunPark, SunYoungKim, SungZooKim, KyungWooCho 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 8 Pages

It is not clear whether Ca2-induced Ca2 release from the sarcoplasmic reticulum (SR) is involved in the regulation of atrial natriuretic peptide (ANP) release. Previously, we have shown that nifedipine increased ANP release, indicating that Ca2 entry via voltage-gated L-type Ca2 channel activation decreases ANP release. The purpose of the present study was two-fold: to define the role of SR Ca2 release in the regulation of ANP release and whether Ca2...
