Modulation of Ca2+-Activated Potassium Channels by cGMP- Dependent Signal Transduction Mechanism in Cerebral Arterial Smooth Muscle Cells of the Rabbit
The Korean Journal of Physiology & Pharmacology 6 Pages
The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.6 2 439-444 (6 pages)
The present investigation tested the hypothesis that the activation of protein kinase G (PKG) leads to a phosphorylation of Ca2-activated potassium channel (KCa channel) and is involved in the activation of KCa channel activity in cerebral arterial smooth muscle cells of the rabbit. Single-channel currents were recorded in cell-attached and inside-out patch configurations of patch-clamp techniques. Both molsidomine derivative 3-morpholinosydnonimine-N-ethylcarbamide (SIN-1, 50μM) and... -
Effects of Prostaglandin F2α on Membrane Potentials and K+ Currents in Rabbit Middle Cerebral Arterial Cells
The Korean Journal of Physiology & Pharmacology 9 Pages
The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.4 5 301-309 (9 pages)
The purpose of our investigation was to examine the effects of prostaglandin F2α (PGF2α) on membrane potentials, Ca2-activated K (KCa) channels, and delayed rectifier K (KV) channels using the patch-clamp technique in single rabbit middle cerebral arterial smooth muscle cells. PGF2α significantly hyperpolarized membrane potentials and increased outward whole-cell K currents. PGF2α increased open-state probability of KCa channels without the change of the open and... -
Effect of Atrial Natriuretic Peptide on the Proliferation and Activity of Osteoblastic Cells
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.4 3 283-289 (7 pages)
Natriuretic peptides comprise a family of three structurally related peptides; atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP), and C-type natriuretic peptide (CNP). The present study was performed to investigate the effect of ANP on the proliferation and activity of ROS17/2.8 and HOS cells which are well-characterized osteoblastic cell lines. ANP dose-dependently decreased the number of ROS17/2.8 and HOS cells after 48-hour treatment. ANP generally increased the alkaline... -
Effects of Cholecystokinin Octapeptide on Neuronal Activities in the Rat Nucleus Tractus Solitarius
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.4 2 275-281 (7 pages)
Cholecystokinin (CCK) is a gastrointestinal hormone which plays an important role in satiety and gastric motility. It is also widely distributed throughout the central nervous system, where it appears to be involved in the central control of anxiety, feeding behavior and nociception. Two distinct CCK receptor types, CCKA and CCKB, have been found in the brain. Both CCK receptors coexist in the rat nucleus tractus solitarius (NTS), which is the primary center for the coordination of peripheral... -
Tyrosine Phosphorylation of Paxillin May be Involved in Vascular Smooth Muscle Contraction
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.3 5 211-217 (7 pages)
Paxillin is a regulatory component of the complex of cytoskeletal proteins that link the actin cytoskeleton to the plasma membrane. However, the role of paxillin during smooth muscle contraction is unclear. We investigated a possible role for the membrane-associated dense plaque protein paxillin in the regulation of contraction in rat aortic vascular smooth muscle. The tyrosine phosphorylation of paxillin, which was increased by norepinephrine, reached a peak level after 1 min stimulation and... -
Relatoinship between Sarcoplasmic Reticular Calcium Release and Na+-Ca2+ Exchange in the Rat Myocardial Contraction

The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.3 4 197-210 (14 pages)
Suppressive role of Na-Ca2 exchange in myocardial tension generation was examined in the negative frequency-force relationship (FFR) of electric field stimulated left atria (LA) from postnatal developing rat heart and in the whole-cell clamped adult rat ventricular myocytes with high concentration of intracellular Ca2 buffer (14 mM EGTA). LA twitch amplitudes, which were suppressed by cyclopiazonic acid in a postnatal age-dependent manner, elicited frequency-dependent and... -
Effect of Brain Angiotensin II Receptor Antagonists and Antisense Oligonucleotide on Drinking and Renal Renin in Rats

The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.2 8 137-142 (6 pages)
The physiological roles of brain angiotensin II in mediating water deprivation-induced drinking and in regulating renal renin release were assessed in male Sprague-Dawley rats. Specific AT1 receptor antagonists, losartan and SK 1080, and antisense oligonucleotide (AS-ODN) directed to AT1 receptor mRNA were intracerebroventricularly (i.c.v.) administered in conscious unrestrained rats. When water was given 20 min after i.c.v. injection of AT1 receptor antagonists in 48-h water-deprived rats,... -
Muscarinic Receptor Subtype Controlling the Carbachol-Induced Muscle Contraction in Guinea Pig Gastric Antrum
The Korean Journal of Physiology & Pharmacology 7 Pages
The Korean Journal of Physiology & Pharmacology 2000, Vol.4 No.2 4 105-111 (7 pages)
Stimulation of muscarinic receptors by carbachol (CCh) in the circular smooth muscle of the guinea pig gastric antrum causes muscle contraction. In the present study, muscarinic receptor subtype controlling the muscle contraction in response to CCh was studied using putative muscarinic receptor antagonists. Isometric force of the isolated circular muscle strips was measured in an organ bath. CCh contracted the muscle in a dose-dependent way, and each of the three muscarinic receptor... -

We investigated the action of NOS inhibitors on NOS in rats. Both of nitric oxide synthase inhibitors, NG-monomethyl-L-arginine (L-NMMA, 3μM) or NG-nitro-L-arginine methylester (L-NAME, 30μM), augmented phenylephrine (PE, 107 M)-induced contraction which was inhibited by acetylcholine (ACh) in rat thoracic aorta. This augmentation by L-NAME or L-NMMA was attenuated with the treatment of NO precursor, arginine. ACh, however, decreased the augmentation induced by L-NMMA, but not by... -
