To investigate whether VacA (vacuolating toxin) produced by Helicobacter pylori `Korean stain 99 induces intestinal secretion, purified VacA was added to T84 cell monolayers mounted in Ussing chambers, and electrical parameters were monitored. Mucosal addition of low pH-pretreated VacA increased short circuit current (Isc). The effect was time- and dose-dependent and saturable. The time-to-peak Isc was concentration-dependent. Chloride channel inhibitors, niflumic acid or 5-... -
Identification of a Marker Protein for Cardiac Ischemia and Reperfusion Injury by Two-Dimensional Gel Electrophoresis and Matrix-Assisted Laser Desorption Ionization Mass Spectrometry

evaluate the expression of cardiac marker protein in rabbit cardiac tissue that was exposed to ischemic preconditioning (IPC), or ischemiareperfusion injury (IR) using two-dimensional gel electrophoresis (2DE) and matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS). We compared 2DE gels of control (uninjured) cardiac tissue with those of IPC and IR cardiac tissue. Expression of one protein was detected in IR heart tissue, however the protein was not detected in the samples... -
Kinesin Superfamily KIF5 Proteins Bind to βIII Spectrin
Jae-EunPaik, NariKim, SungSuYea, WonHeeJang, Joon-YoungChung, Sang-KyoungLee, Yeong-HongPark, JinHan, Dae-HyunSeog 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages

The kinesin proteins (KIFs) make up a large superfamily of molecular motors that transport cargo such as vesicles, protein complexes, and organelles. KIF5 is a heterotetrameric motor that conveys vesicles and plays an important role in neuronal function. Here, we used the yeast two-hybrid system to identify the neuronal protein(s) that interacts with the tail region of KIF5 and found a specific interaction with βIII spectrin. The amino acid residues between 1394 and 1774 of βIII spectrin were... -
Role of STAT3-Interacting Protein (STIP1) in Δ12-Prostaglandin J2-Induced Cell Death

was increased, compared with the vector control cells. The cytotoxicity of Δ12-PGJ2 treatment was significantly accentuated by pretreatment of the STIP1-transfected cells with protein kinase inhibitor, genistein, and less activation of STAT3 in STIP1-transfected cells was shown, compared with the vector control cells. Expression of bax was also increased in the STIP1-transfected cells. These data suggest that STIP1 inhibits cell growth via inhibition of STAT3 activation in Δ12-PGJ2 treatment. -
Molecular Mechanism of Pancreatic Bicarbonate Secretion

대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2002, Vol.6 No.3 1 131-138 (8 pages)
Thanks to recent progress in availability of molecular and functional techniques it became possible to search for the basic molecular and cellular processes that mediate and control HCO3 and fluid secretion by the pancreatic duct. The coordinated action of various transporters on the luminal and basolateral membranes of polarized epithelial cells mediates the transepithelial HCO3 transport, which involves HCO3 absorption in the resting state and HCO3 secretion in the... -
Effects of Pretreatment of Serotonin Synthesis Inhibitor p-chlorophenylalanine on Lipopolysaccharide-induced Anorexia in Rats
So-youngPark, ByungSuckKim, SeoungSookBack 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 6 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 2001, Vol.5 No.2 3 133-138 (6 pages)
In the present study, we investigated the effect of pretreatment of p-chlorophenylalanine (PCPA), inhibitor of serotonin synthesis, on lipopolysaccharide (LPS)-induced anorexia in rats. First of all, effects of PCPA injection on food intake and body weight in rats were investigated. During 4 days of PCPA injection (300 mg/kg BW once a day), food intake was decreased by 33% and daily gain in body weight was inhibited compared with controls. Twenty-four hours after last PCPA injection, food... -
The Hyperthermic Effect of Nitric Oxide in Central Nervous System

The precise mechanism of set-point regulation in hypothalamus was not elucidated. Nitric oxide synthases(NOS) were detected in hypothalamus, however, the roles of NO in hypothalamus was not fully studied. So, we tested the effects of NO on body temperature because preoptic-anterior hypothalamus was known as the presumptive primary fever-producing site. NO donor sodium nitroprusside (SNP, 4 nmol, i.c.v.) elicited marked febrile response, and this febrile response was completely blocked by... -
Role of NF-κB Binding Sites in the Regulation of Inducible Nitric Oxide Synthase by Tyrosine Kinase

on NO production, iNOS mRNA expression, nuclear factor-κB (NF-κB) binding activity and the promoter activity of iNOS gene containing two NF-κB sites have been examined in a mouse macrophage RAW 264.7 cells. LPS or IFN-γ stimulated NO production, and their effect was enhanced synergistically by mixture of LPS and IFN-γ. The PTK inhibitor such as tyrphostin reduced LPS plus IFN-γ-induced NO production, iNOS mRNA expression and NF-κB binding activity. In contrast, PKC inhibitors such as H-7,... -
Comparative Effects of PKB-α and PKC-ζ on the Phosphorylation of GLUT4-Containing Vesicles in Rat Adipocytes

Insulin stimulates glucose transport in muscle and fat cells by promoting the translocation of glucose transporter (GLUT4) to the cell surface. Phosphatidylinositide 3-kinase (PI3-kinase) has been implicated in this process. However, the involvement of protein kinase B (PKB)/Akt and PKC-ζ, those are known as the downstream target of PI3-kinase in regulation of GLUT4 translocation, is not known yet. An interesting possibility is that these protein kinases phosphorylate GLUT4 directly in this... -
Effect of the Inhibition of Platelet Activating Factor on Oxidative Lung Injury Induced by Interleukin-1α
YoungManLeeYoon-YubPark 대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 13 Pages
대한생리학회-대한약리학회 The Korean Journal of Physiology & Pharmacology 1998, Vol.2 No.4 10 479-491 (13 pages)
In order to know the pathogenesis of adult respiratory distress syndrome (ARDS) in association with the oxidative stress by neutrophils, the role of platelet activating factor (1-0-alkyl-2-acetyl-sn- glycero-3-phosphocholine, PAF) was investigated during acute lung injury induced by interleukin- 1α (IL-1) in rats. An insufflation of IL-1 into the rat's trachea increased the acetyltransferase activity in the lung and the increase of PAF content was followed. As evidences of acute lung injury by...
