Despite the potential importance of the human regulator of calcineurin 1 (RCAN-1) gene in the modulation of cell survival under stress, little is known about its role in death-inducing signal pathways. In this study, we addressed the effects of RCAN1.4 knockdown on cellular susceptibility to apoptosis and the activation of death pathway proteins. Transfection of siRNAs against RCAN1.4 resulted in enhanced Fas- and etoposide-induced apoptosis, which was associated with increased expression and... -
Rifampicin Inhibits the LPS-induced Expression of Toll-like Receptor 2 via the Suppression of NF-κB DNA-binding Activity in RAW 264.7 Cells

determined that rifampicin suppresses LPS-induced TLR2 mRNA expression. The down-regulation of TLR2 expression coincided with decreased production of TNF-Ձ. Since NF-ՊB is a major transcription factor that regulates genes for TLR2 and TNF-Ձ, we examined the effect of rifampicin on the LPS-induced NF-ՊB activation. Rifampicin inhibited NF-ՊB DNA-binding activity in LPS-activated RAW 264.7 cells, while it did not affect IKKՁ/Ղ activity. However, rifampicin... -

Heat shock proteins (HSPs) serve as molecular chaperones and play a role in cell protection from damage in response to stress stimuli. The aim of this article is to investigate whether HSP22 affects IL-8 expression in vascular smooth muscle cells (VSMCs), and which cellular factors are involved in the HSP-mediated IL-8 induction in that cell type in terms of mitogen activated protein kinase (MAPK) and transcription element. Exposure of aortic smooth muscle cells (AoSMCs) to HSP22 not only... -

TGF-<FONT FACE="HJ중명조">Ղ1 mRNA was effectively reduced. TGF-<FONT FACE="HJ중명조">Ղ1 RiAS, mismatched RiAS, and normal saline were each injected to mice via tail veins. When examined for the biochemical effects on the liver, TGF-<FONT FACE="HJ중명조">Ղ1 mRNA levels were significantly reduced only in the TGF-<FONT FACE="HJ중명조">Ղ1 RiAS-treated group. The results of immunohistochemical studies showed that TGF-<FONT FACE="HJ중명조">Ղ1... -
Regulation of Vacuolar H+-ATPase c Gene Expression by Oxidative Stress

was lost in mutants of the GC box. We confirmed by electrophoretic mobility shift and supershift assays that Sp1 protein specifically binds to the GC box. The promoter activity was not changed by the H2O2 treatment and incubation in hypoxic chamber, however, H2O2 increased the stability of ATP6L mRNA. These data suggest that the expression of the ATP6L gene by oxidative stresses is regulated at posttranscriptional level, whereas the GC box is important in basal activities of the promoter. -
Enhanced Expression of Cell Adhesion Molecules in the Aorta of Diabetic Mice is Mediated by gp91phox-derived Superoxide

Endothelial activation and subsequent recruitment of inflammatory cells are important steps in atherogenesis. The increased levels of cell adhesion molecules (CAM) have been identified in diabetic vasculatures, but the underlying mechanisms remain unclear. To determine the relationship among vascular production of superoxide, expression of CAM and diabetes, superoxide generation and expression of intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), E- and... -

concentration-dependently reduced iNOS mRNA and protein expression, as evidenced by Northern and Western blot analysis, respectively. The underlying mechanism by which YS 49 suppressed iNOS expression was not to affect iNOS mRNA stability but to inhibit activation and translocation of NF-κB by preventing the degradation of its inhibitory protein IκBα. As expected, YS 49 prevented NO-induced apoptotic cell death by sodium nitroprusside. Taken together, it is concluded that YS 49 inhibits iNOS... -
Kinesin Superfamily KIF5 Proteins Bind to βIII Spectrin

The kinesin proteins (KIFs) make up a large superfamily of molecular motors that transport cargo such as vesicles, protein complexes, and organelles. KIF5 is a heterotetrameric motor that conveys vesicles and plays an important role in neuronal function. Here, we used the yeast two-hybrid system to identify the neuronal protein(s) that interacts with the tail region of KIF5 and found a specific interaction with βIII spectrin. The amino acid residues between 1394 and 1774 of βIII spectrin were... -
Transduction of Tat-Superoxide Dismutase into Insulin-producing MIN6N Cells Reduces Streptozotocin-induced Cytotoxicity

The reactive oxygen species (ROS) are considered to be an important mediator in pancreatic β cell destruction, thereby triggering the development of insulin-dependent diabetes mellitus. In the present study, HIV-1 Tat-mediated transduction of Cu,Zn-superoxide dismutase (SOD) was investigated to evaluate its protective potential against streptozotocin (STZ)-induced cytotoxicity in insulin-producing MIN6N cells. Tat-SOD fusion protein was successfully delivered into MIN6N cells in a... -

the functional consequences of these changes in various thyroid states were investigated. The effects of thyroid hormones on (1) SR Ca2 uptake, (2) phosphorylation levels of phospholamban, (3) SR Ca2-ATPase and phospholamban protein levels, (4) phospholamban mRNA levels were examined. Our findings indicate that hyperthyroidism is associated with increases in Ca2-ATPase and decreases in phospholamban levels whereas opposite changes in these proteins occur in hypothyroidism.
