Open Access : Specific and Sensitive Detection of Venturia nashicola, the Scab Fungus of Asian Pears, by Nested PCR
The Plant Pathology Journal 7 Pages
1 357-363 (7 pages)
sets was tested on strains representing three species of Venturia and 20 fungal plant pathogens. The nested PCR primer pair specific to V. nashicola was developed based on the sequence of the species-specific 525-bp DNA fragment amplified by primer set E11F/ E11R. The internal primer pair Na11F/Na11R amplified a 235-bp fragment from V. nashicola, but not from any other fungal species tested. The nested PCR assay was sensitive enough to detect the specific fragment in 50 fg of V. nashicola DNA. -
Mini-Review : Current Insights into Research on Rice stripe virus
The Plant Pathology Journal 11 Pages
1 223-233 (11 pages)
Rice stripe virus (RSV) is one of the most destructive viruses of rice, and greatly reduces rice production in China, Japan, and Korea, where mostly japonica cultivars of rice are grown. RSV is transmitted by the small brown plant-hopper (SBPH) in a persistent and circulative-propagative manner. Several methods have been developed for detection of RSV, which is composed of four single-stranded RNAs that encode seven proteins. Genome sequence data and comparative phylogenetic analysis have been... -
Biological Control of Apple Ring Rot on Fruit by Bacillus amyloliquefaciens 9001
The Plant Pathology Journal 6 Pages
6 168-173 (6 pages)
Apple ring rot disease, caused by Botryosphaeria dothidea (Moug. ex. Fr) Ces. et de Not., is one of the most important diseases on apple fruits. In this study, strain 9001 isolated from healthy apple fruits from an infested orchard was evaluated for its biocontrol activity against apple ring rot in vitro and in vivo. Strain 9001 showed obvious antagonistic activity to B. dothidea YL-1 when plated on potato dextrose agar. Soaking healthy apples in the bacterial suspensions of strain 9001 prior to... -
The First Identified Citrus tristeza virus Isolate of Turkey Contains a Mixture of Mild and Severe Strains
The Plant Pathology Journal 11 Pages
4 31-41 (11 pages)
The presence of Citrus tristeza virus (CTV) has previously been reported in citrus growing regions of Turkey. All serologically and biologically characterized isolates including I d r, which was the first identified CTV isolates from Turkey, were considered mild isolates. In this study, molecular characteristics of the I d r isolate were determined by different methods. Analysis of the I d r isolate by western blot and BD-RT-PCR assays showed the presence of MCA13 epitope, predominantly found in... -
The Complete Genome Sequence of Southern rice black-streaked dwarf virus Isolated from Vietnam
The Plant Pathology Journal 5 Pages
11 428-432 (5 pages)
We determined the complete genome sequence of a Vietnamese isolate of Southern rice black-streaked dwarf virus (SRBSDV). Whole genome comparisons and phylogenetic analysis showed that the genome of the Vietnamese isolate shared high nucleotide sequence identities of over 97.5% with those of the reported Chinese isolates, confirming a common origin of them. Moreover, the greatest divergence between different SRBSDV isolates was found in the segments S1, S3, S4 and S6, which differs from the... -
Identification of a Gene Encoding Adenylate Kinase Involved in Antifungal Activity Expression of the Biocontrol Strain Burkholderia pyrrocinia CH-67
The Plant Pathology Journal 8 Pages
4 373-380 (8 pages)
Burkholderia pyrrocinia CH-67 is a biocontrol bacterium with strong antifungal activity against several plant pathogenic fungi. Transposon mutagenesis was performed to identify the genes responsible for the antifungal activity of B. pyrrocinia CH-67. Of the 2,500 mutants tested using the Fulvia fulva spore screening method, a mutant deficient in antifungal activity, M208, was selected. DNA sequence analysis of the transposon-inserted region revealed that a gene encoding an adenylate... -
Begomoviruses and Their Emerging Threats in South Korea: A Review
The Plant Pathology Journal 14 Pages
1 123-136 (14 pages)
Diseases caused by begomoviruses (family Geminiviridae, genus Begomovirus) constitute a serious constraint to tropical and sub-tropical agro-ecosystems worldwide. In recent years, they have also introduced in temperate regions of the world where they have great impact and are posing a serious threat to a variety of greenhouse crops. Begomoviral diseases can in extreme cases reduce yields to zero leading to catastrophic losses in agriculture. They are still evolving and pose a serious threat to... -
Research Notes : Molecular Evidence of Recombination on Korean Isolates of Tomato yellow Leaf curl virus by Nucleotide Transversions and Transitions

and other TYLCV isolates [Thailand (AF206674), Iran (AJ132711), and Israel (X76319)]. TYLCV Jeju isolate was characterized by two recombination events (E1 and E2) caused by the presence of E1 in ORF V1 and C3, which may seem to be the mutations of the high nucleotide transversion and transition rate. Collectively, our results suggest that the occurrence of nucleotide transversions and transitions in TYLCV DNA-A might have induced novel recombination events within the TYLCV Korea isolates. -
RT-PCR Detection of Five Quarantine Plant RNA Viruses Belonging to Potyand Tospoviruses
The Plant Pathology Journal 6 Pages
12 291-296 (6 pages)
In order to detect quarantine plant viruses, we developed reverse transcription-polymerase chain reaction (RT-PCR) primer pairs for five single-stranded (ss) plant RNA viruses that are not currently reported in Korea but could be potential harmful plant viral pathogens. Three viruses such as Chilli veinal mottle virus (ChiVMV), Colombian datura virus (CDV), and Tobacco etch virus (TEV) belong to the genus Potyvirus while Chrysanthemum stem necrosis virus (CSNV) and Iris yellow spot virus (IYSV)... -
Identification of a New Potyvirus Associated with Chlorotic Vein Banding Disease of Spathiphyllum spp., in Andhra Pradesh, India
The Plant Pathology Journal 4 Pages
6 33-36 (4 pages)
The genome of a potyvirus isolate associated with chlorotic spots and vein banding symptoms on Spathiphyllum spp., in Andhra Pradesh state, India was amplified by RT-PCR using degenerate potyvirus primers, amplicons cloned, and sequence (1.6 kb) analyzed. This virus isolate shared maximum identity of 74.8% and 80.2% at coat protein (CP) gene nucleotide (906 nucleotides) and amino acid (302 amino acids) levels, respectively with Dasheen mosaic virus (DsMV)-M13 isolate reported from China. But its...
