Expression of Bax was not affected, whereas Bcl-2 was downregulated in SNP-treated PC12 cells. SNP augmented the release of cytochrome c from mitochondria into cytosol and enhanced caspase 8, 9, and 3 activities. SNP upregulated both Fas and Fas-L, which are known to be components of death receptor assembly. These results suggest that NO induces apoptosis of PC12 cells through both mitochondria- and death receptor-mediated pathways mediated by ROS and Bcl-2 family. -

of TNF-α induced superoxide production in the dihydroethidine- loaded endothelial cells. Diphenyleneiodonium (DPI, 10μmol/L), an inhibitor of NADPH oxidase, and rotenone (1μmol/L), an inhibitor of mitochondrial complex I inhibited TNF-α-induced superoxide production, VCAM-1 expression and monocyte adhesion in the endothelial cells. Taken together, our data suggest that NADPH oxidase and mitochondrial ROS were involved in TNF-α-induced VCAM-1 and monocyte adhesion in the endothelial cells. -

The promoting effect of hydrogen peroxide (H2O2) against the cytotoxicity of 1-methyl-4-phenylpyridinium (MPP) in differentiated PC12 cells was assessed by measuring the effect on the mitochondrial membrane permeability. Ratio-difference before and after the application of DTBNP after pretreatment with CCCP was 0.46⁑0.09 (n=7).
