ATP-sensitive potassium channels (KATP channels) play an important role in insulin secretion from pancreatic beta cells. We have investigated the effect of propofol on KATP channels in cultured single pancreatic beta cells of rats. Channel activity was recorded from membrane patches using the patch-clamp technique. In the inside-out configuration bath-applied propofol inhibited the KATP channel activities in a dose-dependent manner. The half-maximal inhibition dose (ED50) was 48.6⁑8.4μM... -

middle cerebral arterial smooth muscle cells. The RMP and membrane currents were recorded using the whole-cell patch configuration and single KCa channel was recorded using the outside-out patch configuration. Using the pipette solution containing 0.05 mM EGTA, the RMP was 25.76⁑5.08 mV (n=12) and showed spontaneous transient hyperpolarizations (STHPs). The membrane currents showed time- and voltage-dependent outward currents with spontaneous transient outward currents (STOCs). When... -

The Kv channel activity in vascular smooth muscle cell plays an important role in the regulation of membrane potential and blood vessel tone. It was postulated that increased blood vessel tone in hypertension was associated with alteration of Kv channel and membrane potential. Therefore, using whole cell mode of patch-clamp technique, the membrane potential and the 4-AP-sensitive Kv current in cerebral arterial smooth muscle cells were compared between normotensive rat and one-kidney, one-clip... -
Involvement of Spontaneously Formed Cyclic Nucleotides in Cat Gastric Muscle Relaxation

Muscle strips and muscle cells from cat stomach were used to investigate whether spontaneously formed cyclic nucleotides were involved in the inhibition of gastric smooth muscle contraction. A phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine (IBMX), increased the levels of both cyclic GMP (cGMP) and cyclic AMP (cAMP) in resting state cells, while decreasing acetylcholine-induced muscle contraction. Under the influence of IBMX, SQ22536, an adenylyl cyclase inhibitor and methylene blue, a... -
Antioxidant Effects of Serotonin and L-DOPA on Oxidative Damages of Brain Synaptosomes

Antioxidant effects of serotonin and L-DOPA on neuronal tissues were examined by studying the oxidative damages of brain synaptosomal components. The study further explored the mechanism by which they exert protective actions. Serotonin and L-DOPA (1 μM to 1 mM) significantly inhibited lipid peroxidation of brain tissues by either Fe2 and ascorbate or t-butyl hydroperoxide in a dose dependent fashion. Protective effect of serotonin on the peroxidative actions of both systems was greater... -
Regulation of Magnesium Release by cAMP during Chemical Hypoxia in the Rat Heart and Isolated Ventricular Myocytes

Chemically induced hypoxia has been shown to induce a depletion of ATP. Since intracellular free Mg2 ([Mg2]i) appears to be tightly regulated following cellular energy depletion, we hypothesized that the increase in [Mg2]i would result in Mg2 extrusion following hormonal stimulation. To determine the relation between Mg2 efflux and cellular energy state in a hypoxic rat heart and isolated myocytes, [Mg2]i, ATP and Mg2 content were measured by... -
Sensitivity of Rabbit Cerebral Artery to Serotonin is Increased with the Moderate Increase of Extracellular K+

[K]O can be increased under a variety of conditions including subarachnoid hemorrhage. The increase of [K]O in the range of 5∼15 mM may affect tensions of blood vessels and can change their sensitivity to various vasoactive substances. Therefore, it was examined in the present study whether the sensitivity of cerebral arteries to vasoactive substances can be changed with the moderate increase of [K]O, using Mulvany-type myograph and [Ca2]c measurement. The... -

We examined effects of interleukin 1α (IL1α) and phorbol 12, 13 dibutyrate (PDB), an activator of protein kinase C, on mRNA for Prostaglandin H synthase (PGHS) and prostanoid production in cultured ovine meningeal fibroblasts. Immuno- and morphologically-identified fibroblasts were derived from cerebral cortex and white matter from fetal lambs (approximately 120 days gestation) and grown to confluence on glass coverslips in 12 well plates. Levels of prostaglandin F2α and the stable... -

The spontaneous contractions of gastric smooth muscles are regulated by slow waves, which are modulated by both nervous system and humoral agents. This study was designed to examine the effects of prostaglandin E2 (PGE2) on the contractile and electrical activities of antral smooth muscles in guinea-pig stomach, using an intracellular recording technique. To elucidate the underlying mechanism for its effect on contractility, ionic currents were also measured using a whole-cell patch clamp... -

inhibitory effect of caffeine on ICCh in guinea-pig gastric myocyte. In this study, the underlying mechanism of the inhibitory effect of caffeine was investigated. ICCh was completely suppressed by the addition of caffeine (10 mM) to the superfusing solution. Inhibition of ICCh by caffeine was not related to the intracellular cAMP accumulation which was expected from the phosphodiesterase-inhibiting effect of caffeine. The blockade of InsP3-induced Ca2 release by heparin had no significan...
