Endonuclease I과 Exonuclease III가 없는 E. Coli균주 BW9062에서 3-methyladenine과 $O^6$-methylguanine을 제거하는 두 효소를 상당히 높은 순도로 정제하고 이들을 각각 3-methyladenine DNA Glycosylase 및 3-methyladenine과 $O^6$-methylguanine을 DNA Glycosylase라고 명명했다. 이 두 효소는 DNA-Cellulose Column을 사용하여 분리하였다. 그리고 3-methyladenine DNA glycosylase는 Isoeletric focusing column을 사용했을때 pI 값이 6.0과 7.7인 두 형태의 효소로 더욱 분리되었다. pI 7.7에서 얻은 3-methyladenine DNA glycosylase는 3-methyladenine만 주로 배출하고 pI 6.0에서 얻어진 효소는 3-methyladenine 및 7-methylguanine을 배출하였다. 3-methyladenine DNA glycosylase와 $O^6$-methylguanine DNA glycosylase를 Phosphocellulose column을 사용하여 분리할 수 있었다. $O^6$-methylguanine DNA glycosylase는 Isoelectric focusing column에 의해 재확인할 수 있었으나 대단히 불안정 하였다.
Two enzymes which release 3-methyladenine and $O^6$-methylguanine was highly purified from E. coli, BW9062 which lacks Endonuclease I and Exonuclease III and designated as 3-methyladenine DNA glycosylase and $O^6$-methylguanine DNA glycosylase respectively. These two enzymes was purified by using DNA-cellulose column chromatography. The 3-methyladenine DNA glycosylase was further separated to two isoenzymes with isoelectricfocusing column showing pI value of 6.0 and 7.7. 3-Methyladenine DNA glycosylase having pI value of 7.7 released mainly 3-methyladenine. The other 3-methyladenine DNA glycosylase having pI value of 6.0 released both 3-methyladenine and 7-methylguanine. The 3-methyladenine DNA glycosylase and $O^6$-methylguanine DNA glycosylase can be separated with phosphocellulose column. $O^6$-methylguanine DNA glycosylase was identified by using isoelectric focusing column but was highly labile.
