배추흰나비의 5령말 유충의 중장에서의 protease를 Sephadex G 100과 DEAE-Cellulose를 이용하여 정제하였다. 여기서 분리된 protease는 전기영동법과 면역학적방법에서 완전정제됨이 증명되었으며 효소의 최종 순도는 약 8.6이며, pH 8.9에서 Rm 값은 0.1이었다. 면역확산법을 이용하여 protease가 5령말 유충의 혈림프 및 지방체내에는 존재하지만 전장, 후장, 타액선 및 말피기씨소관에는 존재하지 않는 것으로 밝혀졌다.
A protease from the late fifth instar larval midgut of Pieris rapae was purified by gel filtration on Sephadex G 100 and ion exchange chromatography on DEAE-cellulose. This purified enzyme was proved to be homogeneous by electrophoresis on polyacrylamide gel. The final purity of enzyme is about 8.6 and Rm is 0.1 at pH 8.9. Immunological study revealed that protease in the midgut is present in haemolymph and fat body of the late fifth instar larva but not in the foregut, hindgut, salivary gland and Malpighian tuble.
