경피용제제의 전신순환으로의 약물흡수와 약동학적 특성을 분석하기 위한 모델을 평가하고자 두 가지 ketoprofen 첩포제로 부터 얻어진 혈장농도 데이터를 인용하여 PCNONLIN 프로그램을 이용한 1-compartment 모델과 초기용량과 유지용량의 서로 다른 두 가지 zero-order 흡수속도를 포함하는 모델을 적용하였다. 그 결과, 이러한 모델은 관찰된 혈장농도를 잘 반영하였고 산출된 청소율, 흡수속도, 흡수기간 등의 약동학적 파라메터들도 non-compartment 모델을 이용한 분석과 일치 하였다. 따라서 이 모델을 이용한 분석은 경피흡수제제의 흡수속도, 흡수양상, 청소율, 피부잔류여부, 및 항정상태 평균 혈장농도 등을 분석하고 예측하는데 유용할 것으로 사료되었다.
A complemental pharmacokinetic model with two different zero order absorption rate from transdermal drug delivery formulation to systemic circulation was applied to analyze the plasma concentration data obtained from 2 different patches of ketprofen(Ketotop and Mohrus). This model incorporated in PCNONLIN program, predicted the plasma concentration profiles of both patches with good accuracy. The calculated pharmacokinetic parameters of CL/F, Vd/F, initial burst infusion time, and maintenance infusion time were consistent with the results of non-compartment analysis. Those pharmacokinetic parameters were applied to estimate steady-state drug concentrations of both ketoprofen transdermal delivery formulations.
