망개나무는 낙엽교목에 속하는 식물로서 전통적으로 간기능 치료제로 사용되어 왔다. 그러나 항우 울효과 및 그에 대한 분자기전에 대한 연구는 미흡한 실정이다. 86, 285 mg/kg 용량의 망개나무추출 물을 쥐실험군에게 경구투여한 후 변화하는 부동시간의 변화와 면역변화를 분석함으로 추출물의 효 능을 알고자 실험하였다. 그 결과 경구투여한 추출물의 농도에 비례하여 부동시간, 코티졸농도, 그리 고 IL-6, IL-1β, TNF-α 같은 전염증성사이토카인의 농도가 감소하였다. 이와 유사한 경향으로 p-p65과 p-65의 활성도 역시 망개나무추출물 경구투여군이 비투여실험군에 비하여 감소하였다. 위의 결과를 종합하여 볼 때 망개나무 추출물은 우울증에 효과적인 치료제가 될 수 있는 경쟁력 있는 물질이며 이러한 항우울효과는 NFkB 활성 저하조절과 관련이 있다고 생각된다.
Berchemia berchemiaefolia(BB) is a genus of plants in the family Rhamnaceae. Traditionally BB has been already used for liver defender in Korea. But the study about antidepressant-like activity and related molecular mechanisms of BB were not studied well. The effects of subacute treatment with BB(86 and 285 mg/kg, p.o.) on immobility and FST-induced changes of immune parameters were investigated by using the forced swimming test (FST) in rats. As a result, BB immobility decreases with a dose-dependency in the FST. The levels of cortisol, IL-6, IL-1β, and TNF-α of treatment group were significantly decreased than those of the FST group. Similarly p-p65 and p-65 activity of treatment group were decreased than those of the FST group. In all, these results indicate that BB extract can be a very effective treatment for Antidepressant-like effects of Berchemia berchemiaefolia via NFkB signal transduction in rat models of depression depression, suggesting that the down-regulation of NFkB activity is related to Antidepressant Effect of Berchemia berchemiaefolia.
